BCL11B is a Cys2-His2 zinc-finger (C2H2-ZnF) domain-containing, DNA-binding, transcription factor with established roles in the development of various organs and tissues, primarily the immune and nervous systems. BCL11B germline variants have been associated with a variety of developmental syndromes. However, genotype-phenotype correlations along with pathophysiologic mechanisms of selected variants mostly remain elusive.

Belzutifan for renal cell carcinoma: Balancing regulatory approval with societal and patient impact.

LITESPARK-005 evaluated belzutifan against everolimus in advanced renal cell carcinoma (RCC), demonstrating significant progression-free survival improvement but failing to meet the overall survival (OS) co-primary endpoint. Despite FDA approval, the trial highlights key obstacles in drug development in RCC, given the absence of OS improvement, lack of biomarker studies, high financial toxicity, and limited accessibility outside the United States.

Evaluating the physiological benefits of behavioral flexibility in chacma baboons (Papio ursinus) using a biophysical model.

As opportunistic generalists occupying a range of ecological niches, chacma baboons (Papio ursinus) are considered a highly flexible species of relatively low conservation priority. Underlying their ecological flexibility is a repertoire of behavioral strategies observed in response to ecological stressors. Although these strategies are relatively well-documented, we know very little about how they impact upon an individual's thermal and energetic physiology, which can influence population-level reproductive potential in the face of climatic warming.

The post-pandemic world still faces ongoing COVID-19 infections, although international travel has returned to pre-pandemic conditions. Wastewater-based epidemiology (WBE) is considered an efficient tool for the population-wide surveillance of COVID-19 infections during the pandemic. However, the performance of WBE in post-pandemic era with travel restrictions lifted remains unknown.

Advancing cultural humility in pharmacy education: Insights from interprofessional experiences for varied learning modalities.

Effective pharmacy education requires incorporating interactive and engaging strategies that encourage collaboration among people from diverse backgrounds. Activities that forward cultural humility in combination with interprofessional education (IPE) are beneficial to achieve a comprehensive educational experience for enhanced patient centered care and effective teamwork between colleagues. Moving beyond travel seminars and Advanced Pharmacy Practice Experiences (APPEs), these goals can be met through numerous educational formats, which can be tailored to the needs of the course using institutional resources available.

A generalizable methodology for predicting retention time of small molecule pharmaceutical compounds across reversed-phase HPLC columns.

Quantitative structure retention relation (QSRR) is an active field of research, primarily focused on predicting chromatography retention time (Rt) based on molecular structures of an input analyte on a single or limited number of reversed-phase HPLC (RP-HPLC) columns. However, in the pharmaceutical chemistry manufacturing and controls (CMC) settings, single-column QSRR models are often insufficient. It is important to translate retention time across different HPLC methods, specifically different stationary phases (SP) and mobile phases (MP), to guide the HPLC method development, and to bridge organic impurity profiles across different development phases and laboratories.

Dolomite dissolution, pH neutralization, and potentially toxic element immobilization in stormwater bioretention beds.

The importance of pH in stormwater bioretention beds cannot be overstated since it impacts plant and microbial populations and removal of potentially toxic elements (PTEs) from stormwater runoff. This study investigated the effects of dolomite amendment on pH neutralization and subsequent PTE immobilization in bioretention media. To assess dolomite dissolution, pH neutralization, and PTE immobilization, engineered bioretention media was amended with different dolomite ratios and samples of dolomite-amended media were collected from two bioretention beds, one and two months after installation.

Factors Contributing to Opioid Overprescribing at Surgical Discharge.

Introduction: Opioids remain the gold standard for treating acute pain, whereas overprescribing occurs regularly in the postoperative setting with little clinical guidance. The objective of this study is to examine whether the length of surgery is an independent risk factor for opioid overprescribing at discharge.

Methods: We conducted a retrospective case-control study to determine if there is an association between the length of surgery and overprescribed opioids.

Intermittent fasting and neurocognitive disorders: What the evidence shows.

Introduction: Intermittent fasting (IF) has emerged as a potential lifestyle intervention for mitigating cognitive decline and enhancing brain health in individuals with mild to major neurocognitive disorders. Unlike preventive strategies, this review evaluates IF as a therapeutic approach, focusing on its effects on neuroplasticity, inflammation, and cognitive function.

Methods: A narrative review was conducted using a comprehensive PubMed search with the terms "intermittent fasting AND neurocognition" and "intermittent fasting AND neuroplasticity".

Assessing the impact of perioperative anticoagulant continuation on DVT/PE rates in trauma patients.

Introduction: In the United States, deep vein thrombosis (DVT) and pulmonary embolism (PE) ranked high in terms of possibly preventable hospital deaths. Victims of trauma were at a higher risk of developing thromboembolic complications, and thus various agents were used for prophylaxis. Multiple studies recommended holding these agents in the perioperative period to decrease the potential complications of additional bleeding, wound issues, hematoma etc.

No penalty for blocking: Preoperative nerve blocks do not prevent motor nerve stimulation during TMR.

The timing of nerve blocks for amputation surgery with immediate targeted muscle reinnervation (TMR) has been disputed. Traditional practices often defer nerve blocks until post-amputation, fearing interference with motor nerve target identification for TMR. Here, we present a case series demonstrating that pre-amputation regional nerve blocks do not prevent the identification of motor nerve targets.

Sleep quality and duration as predictors of alcohol and cannabis use motives in daily life.

Objective: Alcohol and cannabis are two of the most widely used substances in the United States, where sleep problems are also prominent. Although poor sleep is linked to substance use, little is known about how prior-night sleep contributes to next-day decisions to use substances in daily life. This study tested the impact of prior-night sleep duration and quality on momentary motives for alcohol (Aim 1) and cannabis use (Aim 2).

Feasibility of a randomized controlled trial of enteral vs intravenous resuscitation for adults with major burn injuries in Nepal.

Introduction: Enterally-based resuscitation (EResus) is safe, efficacious, and has operational advantages, particularly in low-resource settings. However, there is a lack of real-world effectiveness studies and evidence-based protocols, which hinders implementation. To address this gap, we conducted a feasibility study ahead of a randomized controlled trial (RCT) of enterally based versus usual resuscitation at a tertiary burn center in Nepal which had no prior clinical trial experience.

Unlocking the phosphorus circularity potential of corn belt watersheds with biorefinery phosphorus recovery incentives.

As global phosphorus (P) stores rapidly decline, P fed algal blooms continue to threaten critical freshwater resources across the globe. In the Midwestern United States (US), particularly the Corn Belt, biorefineries could play a key role in addressing this issue. By recovering P from the byproducts of ethanol production these facilities could reduce the P content of distillers grain feed, thereby reducing P excreted in manures.

Quality-adjusted time without symptoms or toxicity analysis of trastuzumab deruxtecan versus trastuzumab emtansine in HER2- positive metastatic breast cancer patients based on secondary use of the DESTINY-Breast03 trial.

Objective: DESTINY-Breast03, a randomised, phase 3 trial, evaluated trastuzumab deruxtecan (T-DXd) versus trastuzumab emtansine (T-DM1) in patients with human epidermal growth factor receptor (HER2)-positive unresectable and/or metastatic breast cancer who progressed on or after treatment with trastuzumab and a taxane. At the current data cut off overall survival analysis, T-DXd demonstrated a substantial improvement in overall survival over T-DM1. This secondary analysis use of DESTINY-Breast03 aimed to further evaluate the treatment differences using quality-adjusted survival time without symptoms or toxicity (Q-TWiST) methods.

Effect of respiratory rehabilitation on quality of life in individuals with post-COVID-19 symptoms: A randomised controlled trial.

Background: Inspiratory and expiratory muscle training (RMT) has been shown to have beneficial effects in individuals with long-term post-COVID-19 symptoms.

Objective: To assess the effects of adding RMT to an aerobic exercise (AE) training program for health-related quality of life (HRQoL) and exercise tolerance in individuals with long-term post-COVID-19 symptoms, and to evaluate the effects on physical and lung function, and psychological status.

Methods: 64 individuals with long-term post-COVID-19 symptoms of fatigue and dyspnoea were randomly assigned to AE+RMT or AE+RMT groups for an 8-wk intervention (AE: 50min/day, 2 times/wk; RMT: 40min/day, 3 times/wk).

Measurement, decomposition and level-switching in historical science: Geochronology and the ontology of scientific methods.

Philosophers of the historical sciences have focused to a significant extent on the problem of epistemic access facing these sciences: how do historical scientists overcome the relative scarcity of data about the past, compared to the present? Solving this problem usually requires solving another one, which I call the 'problem of ontic access:' how do historical scientists get access to entities and processes with properties that are potentially informative about the past? The case of geochronology illustrates one solution to this problem: historical scientists can get access to entities and processes with properties that are potentially informative about the past by exploiting the metaphysical structure of their domain. Geochronology experienced a spectacular explosion of its research boundaries in the 20th century. I explain this productivity by analyzing the ontology implicit in geochronological techniques.

Association between diffusion tensor imaging measurements and cognitive performances in people with multiple sclerosis: A systematic review and meta-analysis.

Background: Alterations in structural connectivity of brain networks have been linked to complex cognitive functions in people with multiple sclerosis (PwMS). However, a definitive consensus on the optimal diffusion tensor imaging (DTI) markers as indicators of cognitive performance remains incomplete and inconclusive. This systematic review and meta-analysis aimed to explore the evidence on the correlation between DTI metrics and cognitive functions in PwMS.

Rising cardiovascular mortality among obstructive sleep apnea patients: United States epidemiological trends (1999-2019).

Background: Obstructive sleep apnea (OSA) prevalence has risen significantly, affecting millions globally and posing a major healthcare burden. OSA is strongly associated with cardiovascular diseases (CVD) such as heart failure, stroke, and ischemic heart disease. However, trends in CVD-related mortality among individuals with OSA remain underexplored.

DRopEridol for Abdominal pain in the emergency department for Morphine Equivalent Reduction. The DREAMER study.

Introduction: Droperidol is a dopamine-2 receptor antagonist in the class of butyrophenone antipsychotics with antiemetic, sedative, analgesic, and anxiolytic properties. In the postoperative setting, droperidol provides an opioid sparing effect and decreases nausea/vomiting. Another butyrophenone antipsychotic, haloperidol, has been shown to reduce morphine milliequivalents (MME) administered when used for abdominal pain in the emergency department (ED).
