Carbapenemase producing (CPEs) represent a group of multidrug resistant pathogens for which few, if any, therapeutics options remain available. CPEs generally harbor plasmids that encode resistance to last resort carbapenems and many other antibiotics. We previously performed a high throughput screen to identify compounds that can disrupt the maintenance and replication of resistance conferring plasmids through use of a synthetic screening plasmid introduced into K-12 cells.

Efficacy of parenteral bronchodilators on ventilatory outcomes in pediatric critical asthma: a national cohort study.

To evaluate the association of parenteral epinephrine and terbutaline use on ventilatory support in children admitted to the intensive care unit (ICU) with critical asthma in the United States. Data were obtained from the Pediatric Health Information System data base for children ages 2 to 18 years admitted to the ICU with a diagnosis of asthma exacerbation from January 1, 2016, to December 31, 2023. The primary outcomes included noninvasive ventilation (NIV) and/or invasive mechanical ventilation (IMV) use after receipt of terbutaline and/or epinephrine.

Background: Doulas, non-clinical professionals who provide support throughout the perinatal period, can positively impact patient experiences and clinical outcomes during birth. Doulas often support hospital-based births without being employed by the hospital system, resulting in varied relationships with hospitals and clinicians. Systems-level changes are needed to maximize collaboration between hospitals and doulas to ensure facilitation of, and not barriers to, doula support.

Patient-derived response estimates from zero-passage organoids of luminal breast cancer.

Background: Primary luminal breast cancer cells lose their identity rapidly in standard tissue culture, which is problematic for testing hormone interventions and molecular pathways specific to the luminal subtype. Breast cancer organoids are thought to retain tumor characteristics better, but long-term viability of luminal-subtype cases is a persistent challenge. Our goal was to adapt short-term organoids of luminal breast cancer for parallel testing of genetic and pharmacologic perturbations.

Supporting teams with designing for dissemination and sustainability: the design, development, and usability of a digital interactive platform.

Background: Designing for Dissemination and Sustainability (D4DS) principles and methods can support the development of research products (interventions, tools, findings) that match well with the needs and context of the intended audience and setting. D4DS principles and methods are not well-known or used during clinical and public health research; research teams would benefit from applying D4DS. This paper presents the development of a new digital platform for research teams to learn and apply a D4DS process to their work.

Implementation of malaria vector surveillance and insecticide resistance monitoring interventions in Nigeria.

Malaria vector surveillance is required to determine disease transmission dynamics, vector insecticide susceptibility status, suitable control strategies and impact of control interventions. However, capacity and resources for vector surveillance and insecticide resistance monitoring is often inadequate in most countries at risk of vector-borne diseases. Collaborations and linkages between malaria control policy makers and existing research institutions generating vector surveillance research data are often weak, thereby hindering the availability of data for decision-making.

Deep learning enhanced transmembranous electromyography in the diagnosis of sleep apnea.

Obstructive sleep apnea (OSA) is widespread, under-recognized, and under-treated, impacting the health and quality of life for millions. The current gold standard for sleep apnea testing is based on the in-lab sleep study, which is costly, cumbersome, not readily available and represents a well-known roadblock to managing this huge societal burden. Assessment of neuromuscular function involved in the upper airway using electromyography (EMG) has shown potential to characterize and diagnose sleep apnea, while the development of transmembranous electromyography (tmEMG), a painless surface probe, has made this opportunity practical and highly feasible.

Systemic adverse drug events to topical prostaglandin analogs for treating glaucoma: a retrospective focused pharmacovigilance study.

Background: Prostaglandin analogs are first-line treatments for open-angle glaucoma due to their proven efficacy in reducing intraocular pressure. Despite their topical administration, systemic adverse drug Events (ADEs) have been reported. This study investigates the systemic ADEs associated with topical prostaglandin analogs using the United States Food and Drug Administration (USFDA) Adverse Drug Event Reporting System (AERS) database.

Telemedicine for HIV care: a cross-sectional survey of people living with HIV receiving care at two federally qualified health centers in Los Angeles during a mature phase of the COVID-19 pandemic.

Background: The COVID-19 pandemic resulted in the rapid implementation of telemedicine for HIV care at federally qualified health centers (FQHCs) in the United States. We sought to understand use of telemedicine (telephone and video) at two FQHCs in Los Angeles, and the client attitudes towards and experiences with telemedicine as part of future HIV care.

Methods: We conducted surveys with 271 people living with HIV (PLHIV), with questions covering sociodemographic factors, telemedicine attitudes and experiences, technological literacy, and access to technological resources and privacy.

Modeling bladder mechanics with 4D reconstruction of murine ex vivo bladder filling.

This study presents a novel methodology for high-resolution 3D bladder modeling during filling, developed by leveraging improved imaging and computational techniques. Using murine bladder filling data, the methodology generates accurate 3D geometries across time, enabling in-depth mechanical analysis. Comparison with a traditional spherical model revealed similar stress trends, but the 3D model permitted nuanced quantifications, such as localized surface curvature and stress analysis.

Background: New surgeons experience heavy workload during robot-assisted surgery partially because they must use vision to compensate for the lack of haptic feedback. We hypothesize that providing realistic haptic feedback during dry-lab simulation training may accelerate learning and reduce workload during subsequent surgery on patients.

Methods: We conducted a single-blinded study with 12 general surgery residents (third and seventh post-graduate year, PGY) randomized into haptic and control groups.

We searched the Web of Science database for articles related to PE management from 2000 to 2022.

Sensors are indispensable tools of modern life that are ubiquitously used in diverse settings ranging from smartphones and autonomous vehicles to the healthcare industry and space technology. By interfacing multiple sensors that collectively interact with the signal to be measured, one can go beyond the signal-to-noise ratios (SNR) attainable by the individual constituting elements. Such techniques have also been implemented in the quantum regime, where a linear increase in the SNR has been achieved via using entangled states.

Motivations for obtaining advance provision of medication abortion in the United States: a qualitative study.

Background: Advance provision of medication abortion, or the prescription of mifepristone and misoprostol before pregnancy occurs, is an unexplored care model aimed at expanding abortion access. We examine motivations for obtaining advance provision from the online telemedicine service, Aid Access, which supports people in the United States.

Methods: Between May and November 2023, we conducted semi-structured, in-depth interviews with 39 people who obtained advance provision between January 2022 and April 2023.

Background: Substantial evidence suggests an association between obesity and sleep. However, research investigating sleep patterns in relation to novel anthropometric indices is limited. Therefore, we conducted a cross-sectional analysis of data from the National Health and Nutrition Examination Survey (NHANES) from 2007 to 2014 to examine the relationship between the body roundness index (BRI) and unhealthy sleep patterns.

Activation of caged functional RNAs by an oxidative transformation.

RNA exhibits remarkable capacity as a functional polymer, with broader catalytic and ligand-binding capability than previously thought. Despite this, the low side chain diversity present in nucleic acids (two purines and two pyrimidines) relative to proteins (20+ side chains of varied charge, polarity, and chemical functionality) limits the capacity of functional RNAs to act as environmentally responsive polymers, as is possible for peptide-based receptors and catalysts. Here we show that incorporation of the modified nucleobase 2-thiouridine (2sU) into functional (aptamer and ribozyme) RNAs produces functionally inactivated polymers that can be activated by oxidative treatment.

Objectives: This study aimed to evaluate the effect of 30 g prebreakfast and 30 g predinner supplementation of pistachios for 12 wk on glycated hemoglobin (HbA1c), other glycemic markers, anthropometry, and lipid profile of Asian Indians with prediabetes.

Methods: In a 12-wk parallel arm, randomized controlled trial, we recruited 120 participants with prediabetes based on American Diabetes Association criteria.

Background: Greater World Trade Center (WTC) exposure levels have been associated with chronic rhinosinusitis (CRS) diagnoses and symptoms. We aimed to determine whether self-reported CRS is elevated in WTC-exposed Fire Department of the City of New York (FDNY) firefighters when compared with non-WTC-exposed/non-FDNY firefighters and with the general population.

Methods: The study included male WTC-exposed (N = 7840) and non-WTC-exposed (N = 2771) firefighters who were employed on 9/11/2001 and later completed a health survey.

A bifunctional coumarin-based CD probe for chiral analysis of amino acids in aqueous solution.

Amino acids play important roles in human pathology and physiology and the qualitative and quantitative determination of chiral amino acids in humans and mammals also has important impacts on the life sciences. Therefore, the introduction of artificial probes to assess the concentrations and enantiomeric compositions [ee = ([D] - [L])/([D] + [L])] of amino acids in aqueous solution is necessary in understanding certain biological processes and diagnosing and treating diseases. Herein, a bifunctional achiral coumarin probe (Br-Coumarin) is reported to determine the absolute configuration, ee value, and concentration of 16 amino acids in THF/HO = 1/4 solution at micromolar concentrations.

Bilateral internal superior laryngeal nerve injections for unexplained chronic cough.

Objective: Internal Superior laryngeal nerve (iSLN) injections with steroids and anesthetic for Unexplained Chronic Cough (UCC) was initially described as a unilateral injection. This study reports the safety profile and patient-reported outcomes of concurrent bilateral iSLN injections for UCC.
