The hypothalamus plays a central role in regulating energy expenditure and maintaining energy homeostasis, crucial for an organism's survival. Located in the ventral diencephalon, it is a dynamic and adaptable brain region capable of rapid responses to environmental changes, exhibiting high anatomical and cellular plasticity and integrates a myriad of sensory information, internal physiological cues, and humoral factors to accurately interpret the nutritional state and adjust food intake, thermogenesis, and energy homeostasis. Key hypothalamic nuclei contain distinct neuron populations that respond to hormonal, nutrient, and neural inputs and communicate extensively with peripheral organs like the gastrointestinal tract, liver, pancreas, and adipose tissues to regulate energy production, storage, mobilization, and utilization.

The balance between food intake and energy expenditure is precisely regulated to maintain adipose stores. Leptin, which is produced in and released from adipose in direct proportion to its size, is a major contributor to this control and initiates its homeostatic responses largely via binding to leptin receptors (LepR) in the hypothalamus. Decreases in hypothalamic LepR binding signals starvation, leading to hunger and reduced energy expenditure, whereas increases in hypothalamic LepR binding can suppress food intake and increase energy expenditure.

The nonimage-forming effects of light are pivotal in regulating cognitive functions, including alertness, sustained attention, and higher-order cognitive processes. These cognitive domains are deeply influenced by the sleep-wake cycle, which are governed by two key processes: the homeostatic process, which builds sleep pressure during wakefulness, and the circadian process, which aligns with environmental light cues to regulate wakefulness and sleep. When these processes fall out of sync-a condition known as circadian misalignment-alertness, sustained attention, and cognitive performance can suffer significantly.

Introduction: Buprenorphine is a highly effective medication for opioid use disorder (MOUD; OUD), which can be prescribed alongside naloxone in the primary care setting as part of a harm reduction approach to OUD. Novel kappa opioid receptor (KOR) agonists that preferentially activate G-protein signaling versus β-arrestin-2 recruitment are described. Starting from a literature-reported phenol-containing diphenethylamine KOR agonist, structure-activity relationship (SAR) studies revealed replacement of the phenol with various non-hydroxylated bicyclic heteroaromatics led to tertiary diarylethylamines which retained KOR agonist activity and improved metabolic stability in human liver microsomes. Further optimizations produced compound 39, a potent activator of G-protein signaling (GTPγS EC = 14 nM, 83 % E) that did not elicit a β-arrestin-2 recruitment functional response (E < 10 %).

Trans-active response DNA-binding protein-43 (TDP-43) is the major pathological protein in motor neuron disease and TDP-43 pathology has been described in the brains of up to 50% of patients with Alzheimer disease (AD). Hippocampal sclerosis of aging (HS-A), an age-related neuropathology characterized by severe neuronal loss and gliosis in CA1 and/or subiculum, is found in ∼80% of cases that are positive for phosphorylated TDP-43. HS-A is seen as a co-pathology in cases with AD, limbic-predominant age-related TDP-43 encephalopathy neuropathologic changes (LATE-NC), and frontotemporal degeneration.

Despite the dire warnings and calls to action, US public opinion on the causes and solutions to climate change remain divided. In the global context, the US's split views are anomalous and may be an artifact of the US media's coverage of the climate crisis.

This study introduces two refined rainfall anomaly indices-the Modified Rainfall Anomaly Index (MRAI) and the Standardized Rainfall Anomaly Index (SRAI)-to address limitations in the traditional Rainfall Anomaly Index (RAI). The existing RAI struggles to effectively capture extreme wet and dry rainfall conditions and relies on a simplistic formulation. To evaluate these indices on a continental scale, data from the Integrated Multi-Satellite Retrievals for GPM (IMERG) was used for the Conterminous United States (CONUS), enabling scalability to ungaged locations and beyond.
