Article Synopsis
  • The study aimed to systematically review HIV-1 reservoirs in children and adolescents with perinatally acquired HIV, analyzing published data from various studies between 2002 and 2022.
  • A total of 49 studies involving over 2,300 participants were included, revealing a high prevalence of archived-drug resistance mutations (ADRMs) across different regions, with significant findings regarding early antiretroviral therapy (ART) initiation impacting HIV-1 DNA levels.
  • The conclusions emphasized that high ADRM prevalence and larger viral reservoirs are associated with late ART initiation and shorter periods of viral suppression, suggesting the need for early ART strategies in pediatric populations to improve treatment outcomes.

Vitamin B12 (VitB12) is a micronutrient and acts as a cofactor for fundamental biochemical reactions: the synthesis of succinyl-CoA from methylmalonyl-CoA and biotin, and the synthesis of methionine from folic acid and homocysteine. VitB12 deficiency can determine a wide range of diseases, including nervous system impairments. Although clinical evidence shows a direct role of VitB12 in neuronal homeostasis, the molecular mechanisms are yet to be characterized in depth.

Loss-of-function mutations in the GNAL gene are responsible for DYT-GNAL dystonia. However, how GNAL mutations contribute to synaptic dysfunction is still unclear. The GNAL gene encodes the Gα protein, an isoform of stimulatory Gα enriched in the striatum, with a key role in the regulation of cAMP signaling.

Diagnosing and treating the elderly individual with hypopituitarism.

Rev Endocr Metab Disord

June 2024

Division of Endocrinology, Department of Medicine, and Pituitary Center, Johns Hopkins University, Baltimore, MD, 2187, USA.

Hypopituitarism in the elderly is an underestimated condition mainly due to the non-specific presentation that can be attributed to the effects of aging and the presence of comorbidities. Diagnosis and treatment of hypopituitarism often represent a challenging task and this is even more significant in the elderly. Diagnosis can be insidious due to the physiological changes occurring with aging that complicate the interpretation of hormonal investigations, and the need to avoid some provocative tests that carry higher risks of side effects in this population.

Background & Aims: Sustained virological response (SVR) by direct-acting antivirals (DAAs) may reverse the hypercoagulable state of HCV cirrhosis and the portal vein thrombosis (PVT) risk. We evaluated the incidence and predictive factors of de novo, non-tumoral PVT in patients with cirrhosis after HCV eradication.

Methods: Patients with HCV-related cirrhosis, consecutively enrolled in the multi-center ongoing PITER cohort, who achieved the SVR using DAAs, were prospectively evaluated.

Department of Biomedicine and Prevention, University of Tor Vergata, Rome, Italy.

Therapy-related Myeloid Neoplasm (t-MN) represents one of the worst long-term consequences of cytotoxic therapy for primary tumors and autoimmune disease. Poor survival and refractoriness to current treatment strategies characterize affected patients from a clinical point of view. In our aging societies, where newer therapies and ameliorated cancer management protocols are improving the life expectancy of cancer patients, therapy-related Myeloid Neoplasms are an emerging problem.

Accumulating evidence suggests that oral supplementation with myo-Inositol (myo-Ins) is able to reduce the amount of gonadotropins and days of controlled ovarian hyperstimulation (COS) necessary to achieve adequate oocyte maturation in assisted reproduction technology (ART) protocols, particularly in women affected by polycystic ovary syndrome (PCOS). We used computational calculations based on simulation modellings. We simulated in vitro fertilization (IVF) procedures-with or without intracytoplasmic sperm injection (ICSI)-with 100,000 virtual patients, accounting for all the stages of the entire IVF procedure.

Article Synopsis
  • Glioblastoma (GBM) is one of the most aggressive human cancers, and there is a critical need for new treatment methods.
  • Research on the cyclic decapeptide uPAcyclin shows it specifically binds to GBM cells and hinders their movement and ability to invade surrounding tissues without affecting their growth.
  • uPAcyclin significantly reduces the formation of vascular-like structures in GBM cells, indicating its potential as a targeted therapy to prevent new blood vessel formation in treating GBM.
