Background: Empyema, characterized by the accumulation of pus in the pleural cavity, poses significant treatment challenges. While intrapleural tissue plasminogen activator (tPA) and deoxyribonuclease (DNase) therapy is effective for many patients, a substantial proportion require surgical intervention, such as video-assisted thoracoscopic surgery (VATS), following treatment failure. We identified a 5-fluoro-benzothiazole-containing small molecule, TKB272, through fluorine-scanning of the benzothiazole moiety, which more potently inhibits the enzymatic activity of SARS-CoV-2's main protease (M) and more effectively blocks the infectivity and replication of all SARS-CoV-2 strains examined including Omicron variants such as SARS-CoV-2 and SARS-CoV-2 than two M inhibitors: nirmatrelvir and ensitrelvir. Notably, the administration of ritonavir-boosted nirmatrelvir and ensitrelvir causes drug-drug interactions warranting cautions due to their CYP3A4 inhibition, thereby limiting their clinical utility. When orally administered, TKB272 blocked SARS-CoV-2 replication without ritonavir in B6.

What constitutes enough evidence to make a decision? While this is an important question across multiple domains, it takes on special importance in the US legal system, where jurors and judges are instructed to apply specific burdens of proof to render life-changing decisions. Civil trials use a preponderance of evidence (PoE) threshold to establish liability, while criminal trials require proof beyond a reasonable doubt (BaRD) to convict. It is still unclear, however, how laypeople interpret and apply these decision thresholds and how these standards compare to people's intuitive belief (IB) of what constitutes enough evidence.

The Simons Observatory (SO) is a cosmic microwave background (CMB) experiment located in the Atacama Desert in Chile that will make precise temperature and polarization measurements over six spectral bands ranging from 27 to 285 GHz. Three small aperture telescopes (SATs) and one large aperture telescope (LAT) will house 60,000 detectors and cover angular scales between one arcminute and tens of degrees. We present the performance of the dichroic, low-frequency (LF) lenslet-coupled sinuous antenna transition-edge sensor (TES) bolometer arrays with bands centered at 27 and 39 GHz.
