Owing to their ability to reliably detect even very rare antigen-specific B cells in cellular isolates such as peripheral blood mononuclear cells (PBMC), and doing so robustly in a high throughput-compatible manner, B cell ELISPOT/FluoroSpot (collectively "B cell ImmunoSpot") tests have become increasingly attractive for immune monitoring in regulated settings. Presently, there are no guidelines for the qualification and validation of B cell ImmunoSpot assay results. Here, we propose such guidelines, building on the experience acquired from T cell ImmunoSpot testing in an environment adhering to the requirements of regulatory bodies yet taking the unique features of B cell assays into account.

Air-pollution monitoring is sparse across most of the United States, so geostatistical models are important for reconstructing concentrations of fine particulate air pollution (PM) for use in health studies. We present XGBoost-IDW Synthesis (XIS), a daily high-resolution PM machine-learning model covering the contiguous US from 2003 through 2023. XIS uses aerosol optical depth from satellites and a parsimonious set of additional predictors to make predictions at arbitrary points, capturing near-roadway gradients and allowing the estimation of address-level exposures.

Despite significant advancements in knowledge, medicine, and technology, as well as the promotion of guidelines and the implementation of numerous global initiatives aimed at addressing health disparities and mitigating the progression of non-communicable diseases (NCDs) worldwide, substantial work remains to be undertaken particularly in addressing inequalities in physical activity. Achieving equitable access to health resources and parity in health outcomes remains a critical and unresolved challenge.

Sorghum proteins were used to prepare gels with concentrations of 15 %, 20 %, 25 %, 30 %, and 35 % (w/w) in 70 % ethanol. Numerous unregulated organic compounds (UOCs) including pharmaceuticals, opioids, and personal care products (PCPs) end up in wastewater. UOC presence in biosolids (a wastewater treatment byproduct), which are applied to soil for different reasons raises environmental and health risk concerns. In this study, two multi-class extraction methods were developed and validated to target 111 UOCs from 8 different major families simultaneously in biosolids and biosolids-impacted soil.

Scent marking through urine spraying is known to aid mate selection, territory marking and chemical communication in terrestrial, but not in aquatic mammals. We quantify an unusual aerial urination behaviour in botos (Inia geoffrensis) and discuss its potential functions. Between 2014 and 2018, we conducted land-based behavioural surveys on wild botos in central Brazil, recording the sequence, duration and social context of aerial urination.

Truncated hemoglobins (TrHbs) have an ancient origin and are widely distributed in microorganisms where they often serve roles other than dioxygen transport and storage. In extremophiles, these small heme proteins must have features that secure function under challenging conditions: at minimum, they must be folded, retain the heme group, allow substrates to access the heme cavity, and maintain their quaternary structure if present and essential. The genome of the obligate psychropiezophile Shewanella benthica strain KT99 harbors a gene for a TrHb belonging to a little-studied clade of globins (subgroup 2 of group N).

Somatic Y537S and D538G mutations within the estrogen receptor alpha ligand-binding domain (ERα-LBD) have been linked to enhanced cell proliferation, survival, and metastases in ER-positive breast cancers. Such mutations are thought to confer ligand-independent receptor activation by increasing the flexibility of helix 12 (H12), a segment within the ERα-LBD that acts as a dynamic regulator of ERα activity. We employed bipartite tetracysteine display coupled with the biarsenical profluorophore FlAsH-EDT to monitor ligand-independent structural transitions of H12 in ERα-LBDs that include Y537S or D538G mutations.
