Human citrate synthase (hCS) was kinetically characterized through full progress curve kinetic modelling using kinetic simulation, global fitting of the direct AcCoA to CoA transition, and a coupled thiol probe reaction to better determine the kinetics with low substrate concentration. Our analysis provides one of the most rigorous kinetic analyses of any citrate synthase ruling out the need to invoke complex cooperative mechanisms to explain progress curve data. Furthermore, we collected and modeled stopped-flow pH-dependent kinetic data with CoA and popular thiol probes such as Ellman's reagent (DTNB) and 4,4'-Dithiodipyridine (DPS), providing the opportunity for detailed kinetic simulations using these thiol probes with CoA producing enzymes.

We suggest that this mnemonic includes several improvements on previous pathophysiology-based acronyms and have informally validated this new mnemonic with two lists of common diseases.

This study compared maximum a posteriori (MAP), expected a posteriori (EAP), and Markov Chain Monte Carlo (MCMC) approaches to computing person scores from the Multi-Unidimensional Pairwise Preference Model. The MCMC approach used the No-U-Turn sampling (NUTS). Results suggested the EAP with fully crossed quadrature and the NUTS outperformed the others when there were fewer dimensions.

Background: More than 90% of in-hospital cardiac arrests involving children occur in an intensive care unit (ICU) with less than half surviving to discharge. We sought to assess the association of the display of risk scores of cardiovascular and respiratory instability with the incidence of cardiac arrest in a pediatric ICU.
Methods: Employing supervised machine learning, we previously developed predictive models of cardiovascular and respiratory instability, incorporating real-time physiologic and laboratory data, to display risk scores for potentially catastrophic clinical events in the subsequent 12 h.

In 2025, it will be 30 years since the initial clinical approval of pegylated liposomal doxorubicin (PLD) by the Food and Drug Administration. PLD predated the field of nanomedicine and became a model nanomedicine setting key pharmacological principles (prolonged circulation, slow drug release and the enhanced permeability and retention (EPR) effect) for clinical application of other nano-drugs in cancer therapy. The impressive reduction of cardiotoxicity conferred by PLD is the most valuable clinical asset.

Purpose: Radiation therapy is used in over 60% of cancer patients and can lead to radiation dermatitis, radiation induced fibrosis, hyperpigmentation, telangiectasias, fat necrosis, and poor wound healing. Deferoxamine (DFO) is an iron-chelating agent that has been used systemically to treat iron overload conditions and more recently been studied to treat radiation fibrosis. Through iron chelation, DFO stabilizes hypoxia inducible factor-1α, driving downstream upregulation of angiogenic factors, and reduces formation of reactive oxygen species, thereby offering a potential therapy for radiation associated chronic wounds.
