Viral variant and host vaccination status impact infection with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), yet how these factors shift cellular responses in the human nasal mucosa remains uncharacterized. We performed single-cell RNA sequencing (scRNA-seq) on nasopharyngeal swabs from vaccinated and unvaccinated adults with acute Delta and Omicron SARS-CoV-2 infections and integrated with data from acute infections with ancestral SARS-CoV-2. Patients with Delta and Omicron exhibited greater similarity in nasal cell composition driven by myeloid, T cell and SARS-CoV-2 cell subsets, which was distinct from that of ancestral cases.

Adenosine, a critical molecule regulating cellular function both inside and outside cells, is controlled by two human adenosine deaminases: ADA1 and ADA2. While ADA1 primarily resides in the cytoplasm, ADA2 can be transported to lysosomes within cells or secreted outside the cell. Patients with ADA2 deficiency (DADA2) often suffer from systemic vasculitis due to elevated levels of TNF-α in their blood.

Temporally and Spatially Controlled Age-Related Prostate Cancer Model in Mice.

Bio Protoc

January 2025

Department of Structural and Cellular Biology, Tulane University, New Orleans, LA, USA.

The initiation and progression of prostate cancer (PCa) are associated with aging. In the history of age-related PCa research, mice have become a more popular animal model option than any other species due to their short lifespan and rapid reproduction. However, PCa in mice is usually induced at a relatively young age, while it spontaneously develops in humans at an older age.

Immunotherapy, particularly that based on blocking checkpoint proteins in many tumors, including melanoma, Merkel cell carcinoma, non-small cell lung cancer (NSCLC), triple-negative breast (TNB cancer), renal cancer, and gastrointestinal and endometrial neoplasms, is a therapeutic alternative to chemotherapy. Immune checkpoint inhibitor (ICI)-based therapies have the potential to target different pathways leading to the destruction of cancer cells. Although ICIs are an effective treatment strategy for patients with highly immune-infiltrated cancers, the development of different adverse effects including cutaneous adverse effects during and after the treatment with ICIs is common.

Evaluating the impact of performance status in elderly patients with glioblastoma.

J Clin Neurosci

January 2025

Department of Radiation Medicine, Roswell Park Comprehensive Cancer Center, Buffalo, NY, USA; Department of Neurosurgery, Roswell Park Comprehensive Cancer Center, Buffalo, NY 14203, USA. Electronic address:

Background: Glioblastoma (GBM) is a common brain tumor with a poor prognosis. There is a paucity of knowledge regarding optimal treatment approaches for elderly patients with GBM who have a relatively good Karnofsky (KPS) or Eastern Cooperative Oncology Group (ECOG) performance status. This study compared treatment outcomes in older patients (≥65) with GBM based on their performance status, either high (KPS ≥ 70 and ECOG < 2) or low (KPS < 70 and ECOG ≥ 2), who underwent hypofractionated radiotherapy (HFRT) (40 Gy in 15 fractions) versus conventional fractionation (60 Gy in 30 fractions).

Transcriptional coupling of telomeric retrotransposons with the cell cycle.

Sci Adv

January 2025

Department of Biochemistry and Molecular Biology, Tulane University School of Medicine, Louisiana Cancer Research Center, 1700 Tulane Avenue, New Orleans, LA 70112, USA.

Article Synopsis
  • Many dipterans utilize telomere-specific retrotransposons (TRs) instead of telomerase for maintaining chromosome ends, but their transcription regulation is not well understood.
  • This study identifies key regulators like the Mediator complex, E2F1-Dp, and Scalloped/dTEAD that influence TR transcription, where reduced Mediator or Sd/dTEAD activity leads to increased TR expression and longer telomeres.
  • CUT&RUN analysis reveals that these regulatory factors directly bind to telomeric repeats, linking the regulation of TR transcription to the cell-cycle machinery, thereby playing a crucial role in the upkeep of chromosome ends.

Mutational signature analysis predicts bacterial hypermutation and multidrug resistance.

Nat Commun

January 2025

Department of Biochemistry and Molecular Biology, School of Medicine, Tulane University, New Orleans, LA, USA.

Article Synopsis
  • Pseudomonas aeruginosa, when losing DNA mismatch repair (MMR), becomes a hypermutator, leading to high rates of multidrug resistance (MDR), especially after exposure to antibiotics.
  • Hypermutated MMR-deficient P. aeruginosa has a specific mutational signature and quickly develops MDR through shared resistance mechanisms across different drug classes.
  • Analyzing mutational signatures of P. aeruginosa in patients shows many MMR-deficient strains are already MDR or likely to become resistant, indicating that this analysis could be a valuable diagnostic tool for predicting and managing MDR in clinical settings.

In Bayesian phylogenetic and phylodynamic studies it is common to summarise the posterior distribution of trees with a time-calibrated consensus phylogeny. While the maximum clade credibility (MCC) tree is often used for this purpose, we here show that a novel consensus tree method - the highest independent posterior subtree reconstruction, or HIPSTR - contains consistently higher supported clades over MCC. We also provide faster computational routines for estimating both consensus trees in an updated version of TreeAnnotator X, an open-source software program that summarizes the information from a sample of trees and returns many helpful statistics such as individual clade credibilities contained in the consensus tree.

Microphysiological systems (MPS) containing perfusable vascular beds unlock the ability to model tissue-scale elements of vascular physiology and disease in vitro. Access to inexpensive stereolithography (SLA) 3D printers now enables benchtop fabrication of polydimethylsiloxane (PDMS) organ chips, eliminating the need for cleanroom access and microfabrication expertise, and can facilitate broader adoption of MPS approaches in preclinical research. Rapid prototyping of organ chip mold designs accelerates the processes of design, testing, and iteration, but geometric distortion and surface roughness of SLA resin prints can impede the development of standardizable manufacturing workflows.
