Purpose: To test the hypothesis that lactate oxidation contributes to the C-bicarbonate signal observed in the awake human brain using hyperpolarized C MRI.

Methods: Healthy human volunteers (N = 6) were scanned twice using hyperpolarized C-MRI, with increased radiofrequency saturation of C-lactate on one set of scans. C-lactate, C-bicarbonate, and C-pyruvate signals for 132 brain regions across each set of scans were compared using a clustered Wilcoxon signed-rank test.

This exploratory study is a follow up to our previous investigation of immune response in the circulation of high-grade Gleason 9 prostate cancer patients treated with EBRT + BT compared to EBRT alone. Notably, EBRT + BT demonstrates the potential to elicit an effect on CD4/CD8 ratio which may have attributed to improved clinical response to therapy. Our findings show promise for leveraging circulating immune cells as predictive biomarkers for radiotherapy response.

The Clinical Utility of Mutation Testing in Acute Leukemia: A Canadian Consensus.

FMS-like tyrosine kinase 3 () mutations are detected in approximately 20-30% of patients with acute myeloid leukemia (AML), with the presence of a internal tandem duplication (-ITD) mutation being associated with an inferior outcome. Assessment of mutational status is now essential to define optimal upfront treatment in both newly diagnosed and relapsed AML, to support post-induction allogeneic hematopoietic stem cell transplantation (alloSCT) decision-making, and to evaluate treatment response via measurable (minimal) residual disease (MRD) evaluation. In view of its importance in AML diagnosis and management, the Canadian Leukemia Study Group/Groupe canadien d'étude sur la leucémie (CLSG/GCEL) undertook the development of a consensus statement on the clinical utility of mutation testing, as members reported considerable inter-center variability across Canada with respect to testing availability and timing of use, methodology, and interpretation.
