Background: Gastric cancer (GC) is a prevalent malignancy with a substantial health burden and high mortality rate, despite advances in prevention, early detection, and treatment. Compared with the global average, Asia, notably China, reports disproportionately high GC incidences. The disease often progresses asymptomatically in the early stages, leading to delayed diagnosis and compromised outcomes.

Inaticabtagene Autoleucel (CNCT19) in adult relapsed or refractory B-cell acute lymphoblastic leukemia.

Blood Adv

December 2024

1. State Key Laboratory of Experimental Hematology, National Clinical Research Center for Blood Diseases, Haihe Laboratory of Cell Ecosystem, Institute of Hematology and Blood Diseases Hospital, Chinese Academy of Medical Sciences and Peking Union Medical College, Tianjin 300020, China; 2. Tianjin Institutes of Health Science, Tianjin 301600, China., Tianjin, China.

Prior to November 2023, CD19 CAR-T therapies had not been approved in China for patients with relapsed or refractory B-cell acute lymphoblastic leukemia (r/r B-ALL), leaving a significant unmet need. In response, Inaticabtagene Autoleucel (Inati-cel), a novel CD19 CAR-T therapy with a distinct scFv (HI19α), was developed and showed promising efficacy in preliminary clinical research. We conducted a phase 2, single-arm, multicenter study of Inati-cel in adult CD19+ r/r B-ALL in China.

Discovery of novel NLRP3 inhibitors based on machine learning and physical methods.

BMC Chem

October 2024

School of Basic Medical Sciences, Anqing Medical College, No.1588, Jixian North Road,Yixiu District, Anqing City, Anhui Province, China.

The NLRP3 inflammasome plays a crucial role in inflammatory responses, particularly in alcohol-related liver disease (ALD). Given that NLRP3 has emerged as a potential therapeutic target for ALD, the development of effective inhibitors is of great importance. In this study, we trained 11 regression models, and the results showed that LightGBM, Random Forest, and XGBoost performed the best, achieving R² values of 0.

Article Synopsis
  • * A study reports that 32 patients with post-transplant relapsed B-ALL treated with donor-derived CAR-T therapy achieved promising outcomes, with many obtaining complete remission.
  • * Over a median follow-up of 42 months, the 2-year overall survival rate was 56.25%, and the treatment showed good long-term safety with no new adverse events, positioning donor-derived CAR-T as a viable option for these patients.

Article Synopsis
  • CAR-T therapy has improved treatment options for relapsed/refractory B-cell lymphoblastic leukemia, but it can cause serious side effects like cytokine release syndrome (CRS) and immune effector cell-associated neurotoxicity syndrome (ICANS).
  • A study involving 93 patients showed that while most (81.7%) experienced CRS and a few (5.3%) experienced ICANS, these conditions did not significantly impact treatment effectiveness or patient survival rates.
  • Ultimately, the findings suggest that experiencing CRS or ICANS does not lead to worse outcomes for patients receiving anti-CD19 CAR-T therapy.

Article Synopsis
  • The study develops a deep learning model using whole-slide images to predict the pathological stage of clear cell renal cell carcinoma (ccRCC) in patients.
  • Histopathological images from 513 patients were split into training and validation sets, with the model showing a strong predictive ability (AUC of 0.875 and accuracy of 0.809).
  • The study also creates a prognostic risk model based on deep learning features, revealing significant differences in survival rates between high-risk and low-risk groups, thus offering valuable insights for clinical diagnosis.

Objective: Diabetic osteoporosis (DOP) is a metabolic disease that occurs in patients with diabetes due to insufficient insulin secretion. This condition can lead to sensory neuropathy, nephropathy, retinopathy, and hypoglycemic events, which can increase the risk of fractures. This study aimed to assess the effectiveness of Empagliflozin, a sodium-glucose cotransporter-2 (SGLT-2) inhibitor, in treating diabetic osteoporosis (DOP) and preventing fractures.

Managing large B-cell lymphoma (LBCL) that is refractory to or relapsed after chimeric antigen receptor (CAR)-T therapy remains a significant challenge. Here we aimed to investigate the safety and efficacy of C-CAR066, an autologous fully human anti-CD20 specific CAR-T, for relapsed/refractory LBCL after failure of anti-CD19 CAR-T therapy. This first-in-human, single-arm, phase 1 study was conducted at two sites in China.

Objective: This study explores the effects of music therapy under the framework of positive emotions, engagement, relationships, meaning, and accomplishment (PERMA) on the negative emotions and quality of life (QOL) in patients with digestive diseases and mild-to-moderate depression in a general hospital.

Methods: From January 2020 to December 2021, 192 digestive diseases patients with mild-to-moderate depression who met the inclusion criteria were retrospectively analyzed and divided into the control group (routine nursing, n = 90) and the observation group (music therapy under PERMA, n = 102). Hamilton Depression Scale (HAMD-17), Hamilton Anxiety Scale (HAMA), General Well-being Scale, and a 36-item short-form health survey were used to evaluate the negative emotions and QOL of patients.

Late-onset Pompe disease (LOPD) is caused by a genetic deficiency of the lysosomal enzyme acid alpha-glucosidase (GAA), leading to progressive limb-girdle weakness and respiratory impairment. The insidious onset of non-specific early symptoms often prohibits timely diagnosis. This study aimed to validate the high-risk screening criteria for LOPD in the Chinese population.

Paroxysmal atrial fibrillation is a common arrhythmia, and its development process and prediction of the degree of atrial fibrosis are of great significance for treatment and management. Optical imaging technology provides a new means for non-invasive observation of atrial electrical activity. The aim of this study is to investigate the predictive effect of sinus node recovery time on the degree of atrial fibrosis in patients with paroxysmal atrial fibrillation, and to provide a basis for the application of optical imaging technology in the study of atrial fibrosis.

Background: Warm compresses are the routine treatment for Meibomian gland dysfunction (MGD) in daily life, but in order to achieve satisfactory efficacy, the treatment needs to be sustained over a long time, which can have an impact on the patient compliance. A more convenient warm compresses will help improve the patient compliance. Therefore, the purpose of the study is to investigate the efficacy and safety of the disposable eyelid warming masks for treatment of dry eye disease (DED) due to MGD.

The process of endometrial repair after injury involves the synergistic action of various cells including immune cells and stem cells. In this study, after combing Fibrinogen(Fg) with poly(L-lacticacid)-co-poly(ε-caprolactone)(P(LLA-CL)) by electrospinning, we placed Fg/P(LLA-CL) into the uterine cavity of endometrium-injured rats, and bioinformatic analysis revealed that Fg/P(LLA-CL) may affect inflammatory response and stem cell biological behavior. Therefore, we verified that Fg/P(LLA-CL) could inhibit the lipopolysaccharide (LPS)-stimulated macrophages from switching to the pro-inflammatory M1 phenotype in vitro.

Article Synopsis
  • - Esophageal cancer (EC) causes a blockage in the esophagus, and while stents can help, tumor growth often leads to re-blockage; a new stent design incorporating Ce6-SiO@MnO nanoparticles has been developed to improve treatment.
  • - This advanced stent produces oxygen in the tumor environment, boosting the effectiveness of photodynamic therapy (PDT) while also altering immune cells (tumor-associated macrophages) to enhance the body's natural anti-tumor response.
  • - The strategy not only modulates immune responses and downregulates certain proteins involved in tumor progression but also prevents the death of key immune cells, showcasing a promising approach for treating advanced esophageal cancer.
