The adriamycin magnetic microspheres (ADM-MAMs) were prepared by the heat-stabilized protein methods. Their physico-chemical properties were examined; their cytotoxicities against tumor cells in vitro were assayed by a modified MTT method, and their effects were observed on the implanted gastric tumor in Wistar rats given ADM-MAMs via alimentary canal at the presence of the external magnetic fields. To investigate the effects of L-tetrahydropalmatine (L-THP) on energy metabolism, endothelin-1 (ET-1) and NO during acute cerebral ischemia-reperfusion of rats, 24 Wistar rats were randomly divided into four groups, with 6 rats in each group: sham-operation group, simple ischemia group, ischemia-reperfusion group and treatment group (L-THP group). Cerebral ATP, lactate, ET-1 and NO levels were measured in all groups. Our results showed that treatment with L-THP could increase cerebral ATP levels, but decrease cerebral lactate, ET-1 and NO concentrations during ischemia-reperfusion in the treatment group.

Growth hormone releasing peptide (GHRP-2) is a synthetic hexapeptide which specifically stimulates secretion of growth hormone (GH) by fetal pituitary somatotrophs through a new membrane receptor, which is different from growth hormone releasing hormone (GHRH) and somatostatin (SMS) receptors. We used cell cultures of human fetal pituitary somatotroph cells to investigate the effect of GHRH, GHRP-2 and somatostatin on GH secretion. To understand the effect of rh-IFN-gamma on the ability of curcumin to kill HL-60 cells in vitro, the myeloid leukemic cell line HL-60 was studied by using cell culture. BrdU incorporation rate was examined by SABC, DNA content was determined by flow cytometry and apoptotic cell percentage was determined by TUNEL method. The results showed that curcumin inhibited proliferation of leukemic cells in a dose-dependent manner.

The changes of cell surface markers before and after activation by IL-2 were detected by flow cytometry (FCM) to establish a more convenient and precise criterion for the judgment of the activation of bone marrow. By using the measurement of the release of lactate dehydrogenase (LDH) the cytotoxicity of mononuclear cells (MNCs) from bone marrow, activated or inactivated, on tumor cell line K562 was evaluated, and at the same time the changes of surface markers on MNCs before and after activation were examined by using FCM. The results showed that the cytotoxicity of MNCs from bone marrow activated by IL-2 on tumor cell line K562 was increased obviously and the number of CD25+ and CD70+ positive cells in bone marrow MNCs was higher than before activation.

The multidrug resistance P-glycoprotein (P-gp) expression and function in hematopoietic stem/progenitor cells were studied to investigate whether the inhibition of hematopoietic cell P-gp function by multidrug resistance reversal agent increases the cytotoxicity of chemotherapy drugs on the hematopoietic cells. The expression of P-gp on the surface of CD34+ cells from healthy human marrow was examined by flow cytometry. The multidrug resistance reversal agent MS-209 was used to measure the effects of MS-209 on the Rhodamin-123 uptaking of CD34+ hematopoietic cells.

The effect of Sinomenine on IL-8, IL-6, IL-2 and mIL-2R produced by peripheral blood mononuclear cells was investigated by using cell culture, radioimmunoassay and flow cytometry. It was showed that production of IL-8 and mIL-2R was inhibited, but the levels of IL-6 were enhanced by Sinomenine. Our results also demonstrated that Sinomenine did not have any effect on the production of IL-2.

The inhibition of CH50, a recombinant polypeptide of human fibronectin, on the formation of tumor metastases in vivo was studied by inoculation of melanoma B16/F1 cells by hypodermic or intraperitoneal injection, and the size and amount of tumor nodes after therapy were measured. In the treatment of hypodermic tumor, local injection of CH50 produced much better efficacy than distance injection of CH50 did. The inhibitory effect of CH50 on the growth of tumor reached 50%.

Objectives: In an attempt to develop new method of treating the end- or mid-stage pancreatic cancer, we examined the effect of ischemic re-perfusion injury plus particle embolism on the pathology and cell apoptosis of pancreatic cancer in Sprague Dawely rats.
Methods: 9 mg dimethylbeneanthracine (DMBA) were implanted directly into the parenchyma of pancreatic tail of Sprague-Dawely rats. After establishment of tumor, the inferior splenic artery, a main supplying vessel to pancreatic tail was subjected to blockade and re-opening for 30 min separately, then embolism particles were infused via the artery.

Objective: To explore the association between the abnormal phosphorylation sites found in Alzheimer disease (AD) tau and the inhibition of its biological activity.
Methods: Ultracentrifugation, chromatography, manual Edman degradation and autosequence techniques were used to prepare and phosphorylate human recombinant tau, isolate and purify 32P tau peptides and determine phosphorylation sites.
Results: (1) Phosphorylation of tau by casein kinase-1 (CK-1), cyclic AMP-dependent protein kinase (PKA) and glycogen synthetase kinase-3 (GSK-3) differentially inhibited its biological activity, and the inhibition of this activity of tau by GSK-3 was significantly increased if tau was prephosphorylated by CK-1 or PKA.
