This study aimed to investigate the prevalence of daytime sleepiness (DS) and its impact on quality of life (QOL) in outpatients with schizophrenia in the maintenance phase, as well as to identify the factors associated with DS. A total of 191 outpatients with schizophrenia completed a self-administered questionnaire including questions on lifestyle, sleep habits, DS, QOL, and sleep disorders. Insomnia, DS, and QOL were evaluated by the Athens Insomnia Scale (AIS), the Epworth Sleepiness Scale (ESS), and the MOS 8-Item Short-Form Health Survey (SF-8), respectively.

Objective/background: The short-term efficacy and safety of daridorexant, a dual orexin receptor antagonist, has been demonstrated in Japanese patients with insomnia disorder. The objective of this study was to evaluate, in a non-overlapping patient population to the short-term study, the long-term safety and efficacy of daridorexant in Japanese patients with insomnia disorder.

Patients/methods: In this Phase 3 open-label study conducted in Japan, 154 patients with insomnia disorder were randomized to daridorexant 50 mg (n = 102) or 25 mg (n = 52) for 52 weeks.

Daridorexant in Japanese patients with insomnia disorder: A phase 3, randomized, double-blind, placebo-controlled study.

Sleep Med

October 2024

Department of Psychiatry, Nihon University School of Medicine, Itabashi, Tokyo, Japan; Tokyo Adachi Hospital, Adachi, Tokyo, Japan. Electronic address:

Objective: This Phase 3 double-blind, placebo-controlled study evaluated the efficacy and safety of daridorexant in Japanese patients with insomnia disorder.

Patients/methods: 490 patients with insomnia disorder from 95 sites in Japan were randomized to daridorexant 50 mg (n = 163), 25 mg (n = 163) or placebo (n = 164) for 4 weeks, followed by a 7-day placebo run-out and a 30-day safety follow-up. The primary efficacy endpoints, in hierarchical order, were change from baseline at Week 4 in subjective total sleep time (sTST) and subjective latency to sleep onset (sLSO), for daridorexant 50 mg vs placebo.

Article Synopsis
  • This study examines the effectiveness of combining cognitive behavioral therapy (CBT) with medication versus medication alone in improving coping strategies and reducing depressive symptoms in patients with major depression.
  • Researchers analyzed changes in stress coping strategies and depression severity over 16 weeks in two groups of patients: one receiving CBT and medication, and the other receiving only medication.
  • Results indicated that the CBT group developed better coping skills and experienced a significantly greater decrease in depressive symptoms compared to the pharmacotherapy group, suggesting the benefits of integrating coping strategies in treatment plans.

Article Synopsis
  • A global push is underway to create community-based mental health systems, and this study aims to understand the views of both users and non-users of mental health services in Japan regarding mental illness.
  • An internet survey with a newly developed questionnaire sampled 500 people with depression and 500 healthy individuals, revealing that many participants were unfamiliar with key facts about mental illness and available services.
  • The findings suggest that there is a critical need for increased awareness and education about mental health, especially targeting adolescents and young adults, along with establishing accessible support services in communities.

Article Synopsis
  • - Central hypersomnia (HS) and delayed sleep-wake phase disorder (DSWPD) are common in adolescents and significantly lower their quality of life and academic performance, despite being thought to have different causes.
  • - A study analyzed 89 teenagers to compare psychosocial factors and sleep test results between those with HS and DSWPD, noting distinct psychosocial characteristics in each group.
  • - Findings indicated that psychosocial factors, rather than sleep study outcomes, can help differentiate between HS and DSWPD, as sleep latency was the only notable sleep parameter difference.

Adiponectin is a cytokine secreted from adipocytes and regulates metabolism. Although serum adiponectin levels show diurnal variations, it is not clear if the effects of adiponectin are time-dependent. Therefore, this study conducted locomotor activity analyses and various metabolic studies using the adiponectin knockout (APN (-/-)) and the APN (+/+) mice to understand whether adiponectin regulates the circadian rhythm of glucose and lipid metabolism.

This study aimed to investigate the association between insomnia symptoms and non-restorative sleep (NRS) in individuals with Typus melancholicus, a personality trait linked to depression, in the general population. We analyzed data from a Japanese cross-sectional survey of 2519 randomly sampled adults. Typus melancholicus was evaluated using a modified version of Kasahara's Typus melancholicus inventory (modified-KTM).

Aims: A global movement, including in Asia, is seeking to establish integrated youth mental health services that provide early intervention in the continuum from mental health to mental illness. Clinical case management (CCM), in which a case manager becomes not only a coordinator of services but also a provider of psychosocial support, can establish a 'one-stop network' that supports youth in densely populated areas with various social resources. In 2019, we opened a community-based centre called 'SODA' in front of a metropolitan railway station, which was designed to be highly accessible, stigma-free and youth-friendly to provide CCM.

In patients with schizophrenia, motivation has been assumed to act as an intervening factor between cognitive function and social functioning. Motivation is thought to comprise three domains defined by their orientations: intrinsic motivation, extrinsic motivation, and amotivation. Although these orientations can confound each other, the effects of motivation domains on social functioning remain obscure.

Article Synopsis
  • Periocular skin warming has been shown to positively impact both perceived and measured sleep quality.
  • A study with 18 healthy participants found that using a warming eye mask improved the temperature gradient of the skin and led to faster sleep onset and deeper sleep stages compared to a sham mask.
  • The findings suggest that warming the skin around the eyes may help enhance heat loss in the body, promoting quicker and more restful sleep, similar to natural sleep patterns.

Objective: Functional capacity, which indicates one's ability to perform everyday living tasks, contributes to real-world functional performance in patients with schizophrenia. However, functional capacity is sometimes not comparable with functional performance in clinical settings. We hypothesized that specific personality traits are related to this capacity-performance discrepancy of functional outcome.

Background: While longitudinal studies have investigated the relationships between mild cognitive impairment (MCI) subtypes and dementia subtypes, the results have been contradictory. In addition, some research shows that depression accompanied by MCI might increase the risk of Alzheimer's disease (AD).

Objective: The aim of this study is to longitudinally investigate the relationships between MCI subtypes and dementia subtypes, with special attention to the effect of comorbid depressive symptoms in a Japanese rural community.
