Fluorinated multifunctional polymer vesicles for enhanced ocular surface penetration and synergistic treatment of dry eye disease.

J Control Release

January 2025

National Engineering Research Center of Ophthalmology and Optometry, School of Biomedical Engineering, School of Ophthalmology and Optometry, Eye Hospital, Wenzhou Medical University, Wenzhou 325027, China. Electronic address:

Current pharmacotherapy for DED is limited by a vicious inflammatory cycle in which reactive oxygen species (ROS) play a critical role. Additionally, topical eye drop therapy for DED often suffers from poor ocular availability due to multiple ocular surface barriers. Considering the key role of the ROS-NLRP3-IL-1β signaling axis in DED, in this investigation, fluorinated multifunctional polymer vesicles were developed for enhanced ocular surface penetration and synergistic DED therapy by combining ROS scavenging and immunomodulation.

State Key Laboratory of Experimental Hematology, National Clinical Research Center for Blood Diseases, Haihe Laboratory of Cell Ecosystem, Institute of Hematology & Blood Diseases Hospital, Chinese Academy of Medical Sciences & Peking Union Medical College, Tianjin, China.

Lactate dehydrogenase A (LDHA) is upregulated in multiple cancer types and contributes to the Warburg effect. Several studies have found that many tumor-related genes have subtypes and play important roles in promoting cancer development. Here, we identified a novel LDHA transcript, which produced a new protein 3 kDa larger than LDHA, which we named LDHAα.

Proliferative vitreoretinopathy (PVR) is a major cause of rhegmatogenous retinal detachment repair failure. Despite many attempts to find therapeutics for PVR, no pharmacotherapy has been proven effective. Steroids, as the epitome, show uncertain clinical effectiveness, which lacks an explanation and hints at unappreciated mechanisms of PVR.

3D printed biomimetic bilayer limbal implants for regeneration of the corneal structure in limbal stem cell deficiency.

Acta Biomater

January 2025

Beijing Institute of Ophthalmology, Beijing Tongren Eye Center, Beijing Tongren Hospital, Capital Medical University, Beijing 100005, China. Electronic address:

Limbal stem cell deficiency (LSCD) causes vision loss and is often treated by simple corneal epithelial cell transplantation with poor long-term efficiency. Here, we present a biomimetic bilayer limbal implant using digital light processing 3D printing technology with gelatin methacrylate (GelMA) and poly (ethylene glycol) diacrylate (PEGDA) bioinks containing corneal epithelial cells (CECs) and corneal stromal stem cells (CSSCs), which can transplant CECs and improve the limbal niche simultaneously. The GelMA/PEGDA hydrogel possessed robust mechanical properties to support surgical transplantation and had good transparency, suitable swelling and degradation rate as a corneal implant.

Sjögren's syndrome-related dry eye (SSDE) is a severe dry eye subtype characterized by significant immune cell attacks on the lacrimal gland. However, delivering immunosuppressive drugs to the lacrimal glands for SSDE therapy safely and sustainably poses significant challenges in clinical practice. Herein, a ROS-responsive microneedle patch with detachable functionality (CE-MN) is developed to enable straightforward and minimally invasive administration to the lacrimal gland area by penetrating the periocular skin.

Background: The increasing prevalence of myopia among Chinese children and adolescents, especially at younger ages, has emerged as a significant concern in recent years. Pre-myopia is a key period for myopia prevention and control in children and adolescents. Previous studies suggested auricular acupressure (AA) therapy might offer a viable approach to prevent and slow down myopia progression.

Hepatitis C virus (HCV) infection is a major cause of chronic liver disease. Although interferon-free direct-acting antivirals have led to significant advancements in the treatment of HCV infection, the high genetic variability of the virus and the emergence of acquired drug resistance pose potential threats to their effectiveness. In this study, we develop a broad-spectrum aptamer-based proteolysis targeting chimera, designated dNS5B, which effectively degrades both pan-genotypic NS5B polymerase and drug-resistant mutants through ubiquitin proteasome system.

The role of RGC degeneration in the pathogenesis of glaucoma.

Int J Biol Sci

January 2025

Department of Ophthalmology, The Second Hospital of Jilin University, Jilin University, Changchun 130000, Jilin, China.

Glaucoma is a neurodegenerative disorder marked by the loss of retinal ganglion cells (RGCs) and axonal degeneration, resulting in irreversible vision impairment. While intraocular pressure (IOP) is presently acknowledged as the sole modifiable risk factor, the sensitivity of RGCs to IOP varies among individuals. Consequently, progressive vision loss may ensue even when IOP is effectively managed.

Amblyopia is a common eye disease that seriously affects children's eye health. The Chinese Association for Pediatric Ophthalmology and Strabismus has carried out a lot of work for the prevention and treatment of amblyopia, but there are still many problems that need to be paid attention to and solved in the amblyopia screening and clinical diagnosis and treatment. In this article, we discuss three aspects of the amblyopia prevention and treatment, including the timing of prevention and treatment according to visual developmental patterns, the standardized treatment of amblyopia, overdiagnosis and commercialization, and the establishment of an efficient amblyopia screening system, for providing guidance for the standardization of clinical practice and improving the level of amblyopia prevention and treatment in China.
