The Glycopeptide PV-PS A1 Immunogen Elicits Both CD4+ and CD8+ Responses.

Vaccines (Basel)

December 2024

Department of Chemistry and Biochemistry and School of Green Chemistry and Engineering, University of Toledo, 2801 West Bancroft Street, Toledo, OH 43606, USA.

Background/objectives: The MHCII-dependent, CD4+ T-cell zwitterionic polysaccharide PS A1 has been investigated as a promising carrier for vaccine development because it can induce an MHCII-dependent CD4+ response towards a variety of tumor-associated carbohydrate antigens (TACAs). However, PS A1 cannot elicit cytotoxic T lymphocytes through MHCI, which may or may not hamper its potential clinical use in cancer, infectious and viral vaccine development. This paper addresses PS A1 MHCI independence through the introduction of an MHCI epitope, the poliovirus (PV) peptide, to establish an MHCI- and MHCII-dependent vaccine.

A Review of the Consequences of Gut Microbiota in Neurodegenerative Disorders and Aging.

Brain Sci

December 2024

Department of Medicinal and Biological Chemistry, College of Pharmacy and Pharmaceutical Sciences, The University of Toledo, Toledo, OH 43614, USA.

Age-associated alterations in the brain lead to cognitive deterioration and neurodegenerative disorders (NDDs). This review with a particular focus on Alzheimer's disease (AD), emphasizes the burgeoning significance of the gut microbiota (GMB) in neuroinflammation and its impact on the gut-brain axis (GBA), a communication conduit between the gut and the central nervous system (CNS). Changes in the gut microbiome, including diminished microbial diversity and the prevalence of pro-inflammatory bacteria, are associated with AD pathogenesis.

Injectable promethazine in clinics and hospitals: What's the alternative?

JAAPA

January 2025

At the University of Toledo in Toledo, Ohio, Andrew Overholser is an assistant professor in the PA program and practices in the Department of Family Medicine, Megan Sizemore is an assistant professor and clinical pharmacist in the Department of Family Medicine, and Eric Czech is an assistant professor in the PA program and practices in the Department of Family Medicine. The authors have disclosed no potential conflicts of interest, financial or otherwise.

The Institute for Safe Medication Practices and the American Society of Health-System Pharmacists have advocated for removing all injectable promethazine from inpatient and outpatient settings; however, this drug is still being used despite the risk for tissue necrosis, gangrene, and possible amputation when it inadvertently is given by the subcutaneous or intra-arterial route. This article describes alternative injectable medications that can be selected based on patient comorbidities, indications, and clinician experience.

Despite the Heterosexist Harassment, Rejection, and Discrimination Scale (HHRDS) being a routinely used measure of discrimination, there is no current consensus regarding its fundamental psychometric properties. This study sought to: (a) test the dimensionality of the scale using a classical test theory approach, and (b), if lacking a clear multidimensional structure, use an item response theory approach to develop a concise unidimensional measure and examine differential item and test functioning across various identity factors. In a sample of sexual minority adults ( = 1,243), evaluation of the HHRDS supported a reduced 11-item unidimensional measure.

Article Synopsis
  • Transcription factors (TFs) are crucial for regulating gene expression and influencing an organism's traits, and gene-regulatory networks (GRNs) are used to identify potential target genes for these TFs.
  • Researchers conducted experiments in maize using a reverse genetics approach to isolate mutant alleles of 22 TFs but found no significant physical changes in the plants.
  • Although no major morphological changes were observed, transcriptomic profiling revealed differences in gene expression and phenolic compound levels in some mutants, suggesting that while individual TFs may not show dramatic effects, they can still influence gene regulatory networks.
