Quinoline-based compounds can inhibit diverse enzymes that act on DNA.

Cell Chem Biol

December 2024

Department of Epigenetics and Molecular Carcinogenesis, University of Texas MD Anderson Cancer Center, Houston, TX 77030, USA. Electronic address:

DNA methylation, as exemplified by cytosine-C5 methylation in mammals and adenine-N6 methylation in bacteria, is a key epigenetic process. Developing non-nucleoside inhibitors to cause DNA hypomethylation is crucial for treating various conditions without the toxicities associated with existing cytidine-based hypomethylating agents. This study characterized fifteen quinoline-based analogs, particularly compounds with additions like a methylamine (9) or methylpiperazine (11), which demonstrate similar low micromolar inhibitory potency against human DNMT1 and Clostridioides difficile CamA.

Purpose Artificial intelligence (AI) has rapidly gained popularity with the growth of ChatGPT (OpenAI, San Francisco, USA) and other large-language model chatbots, and these programs have tremendous potential to impact medicine. One important area of consequence in medicine and public health is that patients may use these programs in search of answers to medical questions. Despite the increased utilization of AI chatbots by the public, there is little research to assess the reliability of ChatGPT and alternative programs when queried for medical information.

Maintenance methylation, of palindromic CpG dinucleotides at DNA replication forks, is crucial for the faithful mitotic inheritance of genomic 5-methylcytosine (5mC) methylation patterns. MBD proteins use two arginine residues to recognize symmetrically-positioned methyl groups in fully-methylated 5mCpG/5mCpG and 5mCpA/TpG dinucleotides. In contrast, C2H2 zinc finger (ZF) proteins recognize CpG and CpA, whether methylated or not, within longer specific sequences in a site- and strand-specific manner.

  • Cervical tuberculous lymphadenitis (CTL), also known as scrofula, arises from tuberculosis and is especially common in developing countries, but can also be found in developed nations.
  • Diagnosing CTL is challenging due to its similarity to other conditions, leading clinicians to favor traditional RIPE therapy (rifampin, isoniazid, pyrazinamide, ethambutol) over aggressive surgical options due to potential severe complications.
  • A rare side effect of isoniazid, a component of RIPE therapy, may be the development of acanthosis nigricans, as illustrated by a case of a young man from Nepal who experienced this skin condition likely as a result of the medication.

Uncontrolled angiogenesis underlies various pathological conditions such as cancer, age-related macular degeneration (AMD), and proliferative diabetic retinopathy (PDR). Hence, targeting pathological angiogenesis has become a promising strategy for the treatment of cancer and neovascular ocular diseases. However, current pharmacological treatments that target VEGF signaling have met with limited success either due to acquiring resistance against anti-VEGF therapies with serious side effects including nephrotoxicity and cardiovascular-related adverse effects in cancer patients or retinal vasculitis and intraocular inflammation after intravitreal injection in patients with AMD or PDR.
