The human genome contains over 4 million variant sites, as compared with the reference genome, including rare sequence variants, which have the potential to exert large phenotypic effects, such as susceptibility to drug toxicity. Purpose: Several simplifications used in clinical implementations of the convolution∕superposition (C∕S) method, specifically, density scaling of water kernels for heterogeneous media and use of a single polyenergetic kernel, lead to dose calculation inaccuracies. Although these weaknesses of the C∕S method are known, it is not well known which of these simplifications has the largest effect on dose calculation accuracy in clinical situations. The purpose of this study was to generate and characterize high-resolution, polyenergetic, and material-specific energy deposition kernels (EDKs), as well as to investigate the dosimetric impact of implementing spatially variant polyenergetic and material-specific kernels in a collapsed cone C∕S algorithm.

Purpose: The purpose of this project was to determine the spectrum and frequency of mutations in the small nuclear riboprotein 200 kDa gene (SNRNP200) that cause autosomal dominant retinitis pigmentosa (adRP).

Methods: A well-characterized adRP cohort of 251 families was tested for mutations in the exons and intron/exon junctions of SNRNP200 using fluorescent dideoxy sequencing. An additional 21 adRP families from the eyeGENE® Network were tested for possible mutations.

The response of the lymphatic system to inflammatory insult and infection is not completely understood. Using a near-infrared fluorescence (NIRF) imaging system to noninvasively document propulsive function, we noted the short-term cessation of murine lymphatic propulsion as early as 4h following LPS injection. Notably, the effects were systemic, displaying bilateral lymphatic pumping cessation after a unilateral insult.

We previously determined that the predicted risk of radiogenic second cancer in the bladder and rectum after proton arc therapy (PAT) was less than or equal to that after volumetric modulated arc therapy (VMAT) with photons, but we did not consider the impact of margin size on that risk. The current study was thus conducted to evaluate margin size's effect on the predicted risks of second cancer for the two modalities and the relative risk between them. Seven treatment plans with margins ranging from 0 mm in all directions to 6 mm posteriorly and 8 mm in all other directions were considered for both modalities.

Purpose: To calculate photon energy deposition kernels (EDKs) used for convolution/superposition dose calculation at a higher resolution than the original Mackie et al. 1988 kernels and to calculate material-specific kernels that describe how energy is transported and deposited by secondary particles when the incident photon interacts in a material other than water.

Methods: The high resolution EDKs for various incident photon energies were generated using the EGSnrc user-code EDKnrc, which forces incident photons to interact at the center of a 60 cm radius sphere of water.

Modulating cellular adhesion through nanotopography.

Cellular adhesion is a fundamental process in the development of scaffolds for tissue engineering; in the design of biosensors and in preparing antibacterial substrates. A theoretical model is presented for predicting the strength of cellular adhesion to originally inert surfaces as a function of the substrate topography, accounting for both specific (ligand-receptor) and non-specific interfacial interactions. Three regimes have been identified depending on the surface energy (gamma) of the substrate: for small gamma, any increase in roughness is detrimental to adhesion; for large gamma, an optimal roughness exists that maximizes adhesion; and for intermediate gamma, surface roughness has a minor effect on adhesion.

Purpose: The purpose of this study is to evaluate the impact of a promotores-led diabetes self-management program by comparing the outcomes (knowledge, beliefs, and HbA1c level) of Mexican American patients with type 2 diabetes who received usual diabetic care in a wait-list control group to those who received self-management education and follow-up by promotores in consultation with clinic providers and staff.

Methods: This randomized control study compared the results of 63 patients who attended a promotores-led culturally sensitive diabetes self-management course at a community clinic with 68 patients in a wait-list, usual-care control group. Participants were Mexican Americans with type 2 diabetes who were patients at the clinic and at least 18 years of age.
