View Article and Find Full Text PDFThis study proposes a sequential liquid dispensing method using a centrifugal microfluidic device operating at a constant rotational speed for the multiplexed genetic detection of nucleic acid targets across multiple samples in a single operation. A pair of passive valves integrated into each microchamber enabled the liquid to fill towards the center of rotation against the centrifugal force, facilitating the complete removal of air inside the microchamber. Liquid manipulation can be achievable without any surface coating of the device by exploiting the inherent hydrophobicity of the polymer.

Building on previous investigations, structural modifications to the neuronal calcium ion channel blocker MONIRO-1 and related compounds were conducted that included replacement of the amide linker with an aniline and isosteric sulfonamide moiety, and the previously used strategy of substitution of the guanidinium group with less hydrophilic amine functionalities. A comprehensive SAR study revealed a number of phenoxyaniline and sulfonamide compounds that were more potent or had similar potency for the Ca2.2 and Ca3.

Dual functional heterojunctions of tungsten oxide and bismuth vanadate (WO/BiVO) photoanodes are developed and their applications in photoelectrochemical (PEC) water splitting and mineralization of glycerol are demonstrated. The thin-film WO/BiVO photoelectrode was fabricated by a facile hydrothermal method. The morphology, chemical composition, crystalline structure, chemical state, and optical absorption properties of the WO/BiVO photoelectrodes were characterized systematically.
View Article and Find Full Text PDFAngiotensin-converting enzyme 2 (ACE2) is protective in cardiovascular disease, lung injury and diabetes yet paradoxically underlies our susceptibility to SARs-CoV2 infection and the fatal heart and lung disease it can induce. Furthermore, diabetic patients have chronic, systemic inflammation and altered ACE2 expression resulting in increased risk of severe COVID-19 and the associated mortality. A drug that could increase ACE2 activity and inhibit cellular uptake of severe acute respiratory syndrome coronavirus 2 (SARs-CoV2), thus decrease infection, would be of high relevance to cardiovascular disease, diabetes and SARs-CoV2 infection.

The dense glycan shield is an essential feature of the SARS-CoV-2 spike (S) architecture, key to immune evasion and to the activation of the prefusion conformation. Recent studies indicate that the occupancy and structures of the SARS-CoV-2 S glycans depend not only on the nature of the host cell, but also on the structural stability of the trimer; a point that raises important questions about the relative competence of different glycoforms. Moreover, the functional role of the glycan shield in the SARS-CoV-2 pathogenesis suggests that the evolution of the sites of glycosylation is potentially intertwined with the evolution of the protein sequence to affect optimal activity.

Although attributed to a combination of local and global human stressors, the lack of long-term data on Caribbean reef coral communities has prevented a clear understanding of the causes and consequences of coral declines. We integrated paleoecological, historical, and modern survey data to track the occurrence of major coral species and life-history groups throughout the Caribbean from the prehuman period to the present.

Precise manipulation of mode order in silicon waveguides plays a fundamental role in the on-chip all-optical interconnections and is still a tough task in design when the functional region is confined to a subwavelength footprint. In this paper, digital metamaterials consisting of silicon and air pixels are topologically designed by an efficient method combining 2D finite element method for optical simulations, density method for material description and method of moving asymptotes for optimization. Only around 150 iterations are required for searching satisfactory solutions.

Janus transition metal dichalcogenides with unique physical properties have recently attracted increasing research interest for their energy and catalytic applications. In this paper, we investigate the lithiation behavior of a square phase Janus MoSSe monolayer (1S-MoSSe) using first-principles calculations. Computational results show that a single Li atom energetically prefers to adsorb on the central site of the octagonal ring (O site) and on the S-layer side of 1S-MoSSe.

Objective: To investigate feasibility, safety, and tolerability of long-term (48 weeks) add-on treatment with triheptanoin (UX007), the triglyceride of heptanoate, in adults with drug-resistant epilepsy.
Methods: This extension study was offered to adult participants with drug-resistant epilepsy who completed a 12-week randomized controlled trial of add-on medium-chain triglycerides (MCT) vs triheptanoin. Participants were asked to titrate triheptanoin to their maximum tolerated dose over 3 weeks, followed by 48-week maintenance before tapering or treatment extension.

We aimed to study the genesis of seizures caused by mutations affecting K1 channels and searched for potential therapeutic targets.
Methods: We used a novel in silico model, the laminar cortex model (LCM), to examine changes in neuronal excitability and network dynamics associated with loss-of-function mutations in K1 channels.
