Despite the success of poly-ADP-ribose polymerase inhibitors (PARPi) in the clinic, high rates of resistance to PARPi presents a challenge in the treatment of ovarian cancer, thus it is imperative to find therapeutic strategies to combat PARPi resistance. Here, we demonstrate that inhibition of epigenetic modifiers euchromatic histone lysine methyltransferases 1/2 (EHMT1/2) reduces the growth of multiple PARPi-resistant ovarian cancer cell lines and tumor growth in a PARPi-resistant mouse model of ovarian cancer. We found that combinatory EHMT and PARP inhibition increases immunostimulatory double-stranded RNA formation and elicits several immune signaling pathways in vitro.

The widespread use of menthol can be attributed to its unique sensory characteristics; however, emerging evidence suggests that menthol also alters sensitivity to nicotine through modulation of nicotinic acetylcholine receptors (nAChRs). Euchromatic histone lysine methyltransferases 1 and 2 (EHMT1/2), which catalyze demethylation of histone H3 lysine 9 (H3K9me2), contribute to tumorigenesis and therapy resistance through unknown mechanisms of action. In ovarian cancer, EHMT1/2 and H3K9me2 are directly linked to acquired resistance to poly-ADP-ribose polymerase (PARP) inhibitors and are correlated with poor clinical outcomes. Using a combination of experimental and bioinformatic analyses in several PARP inhibitor resistant ovarian cancer models, we demonstrate that combinatory inhibition of EHMT and PARP is effective in treating PARP inhibitor resistant ovarian cancers.

The Atmospheric X-ray Imaging Spectrometer (AXIS) described in this work is a compact, wide field-of-view, hard x-ray imager. The AXIS instrument will fly onboard the Atmospheric Effects of Precipitation through Energetic X-rays (AEPEX) 6U CubeSat mission and will measure bremsstrahlung x-ray photons in the 50-240 keV range with cadmium-zinc-telluride (CZT) detectors using coded aperture optics. AXIS will measure photons generated by energetic particle precipitation for the purpose of determining the spatial scales of precipitation and estimating electron precipitation characteristics.

When voice pitch auditory feedback perturbations are applied in sentence versus sustained-vowel production, larger and faster vocal f responses are measured in sentence production. The synchronized operation of power generators is the foundation of electric power network stability and a key to the prevention of undesired power outages and blackouts. Here, we derive the conditions that guarantee synchronization in power networks with inherent generator heterogeneity when subjected to small perturbations, and perform a parametric sensitivity analysis to understand synchronization with varied types of generators. As inverter-based resources, which are the primary interfacing technology for many renewable sources of energy, have supplanted synchronous generators in ever growing numbers, the center of attention on associated integration challenges have resided primarily on the role of declining system inertia.

We present a method to determine local gravity fields for the Moon using Gravity Recovery and Interior Laboratory (GRAIL) data. We express gravity as gridded gravity anomalies on a sphere, and we estimate adjustments to a background global start model expressed in spherical harmonics. We processed GRAIL Ka-band range-rate data with a short-arc approach, using only data over the area of interest.

We designed, simulated, and experimentally demonstrated components for a microwave-frequency digital-to-analog converter based on single flux quantum (SFQ) circuits and an amplifier based on superconducting-quantum-interference-device (SQUID) stacks. These are key components for a self-calibrated programmable waveform reference for communications metrology capable of synthesizing high-frequency signals with quantum-based output accuracy. The amplifier is an SFQ voltage multiplier circuit that consists of a network of SFQ-splitters and SQUID transformers that provides output signals consisting of quantized pulses.

Electric vehicle (EV) adoption promises potential air pollutant and greenhouse gas (GHG) reduction co-benefits. As such, China has aggressively incentivized EV adoption, however much remains unknown with regard to EVs' mitigation potential, including optimal vehicle type prioritization, power generation contingencies, effects of Clean Air regulations, and the ability of EVs to reduce acute impacts of extreme air quality events. The Pacific Crest Trail (PCT) is a 4265-km hiking trail that extends from the US-Mexican border to the US-Canadian border through the mountain ranges of western North America. Individuals who hike the entire length of the trail in one season (4-6 months) perform long daily exercise durations while exposed to extreme environmental temperatures, high altitudes, intense solar radiation, and the consumption of calorie-rich, nutrient-poor diets.
