Bovine milk extracellular vesicles (EVs) attract research interest as carriers of biologically active cargo including miRNA from donor to recipient cells to facilitate intercellular communication. Since toxicity of edible milk seems to be negligible, milk EVs are applicable to use for therapeutics in human medicine. Casein separation is an important step in obtaining pure EVs from milk, and recent studies reported that adding hydrochloric acid (HCl) and acetic acid (AA) to milk accelerates casein aggregation and precipitation to facilitate EV isolation and purification; however, the effects of acidification on EVs remain unclear.

Fungal pneumonia is a common disease in bottlenose dolphins (Tursiops truncatus), including pregnant and lactating ones. Voriconazole (VRCZ) is commonly used to treat respiratory fungal infections in this species; however, it is unknown whether VRCZ is transferred via the placenta and breastmilk and whether its usage is safe in pregnant and lactating dolphins. We measured VRCZ concentrations in breastmilk and dams', umbilical cord, and calves' plasma samples from four dam-calf dolphin pairs in the Port of Nagoya Public Aquarium, Japan, treated with or without VRCZ.

Article Synopsis
  • CIRBP is a protein that helps protect organisms from cold temperatures and has various splicing variants found in different animals, including hamsters and mice.
  • In mice, several splicing variants of CIRBP mRNA were identified, and a specific short form variant became more prominent during induced hypothermia.
  • The study suggests that low temperatures significantly influence the alternative splicing of CIRBP, indicating that this regulatory mechanism exists in both hibernating and non-hibernating species.

Lipopolysaccharide and lipoteichoic acid enhance serum amyloid A3 mRNA expression in murine alveolar epithelial cells.

J Vet Med Sci

October 2019

Laboratory of Food and Environmental Hygiene, Cooperative Department of Veterinary Medicine, Gifu University, Gifu 501-1193, Japan.

Serum amyloid A (SAA) is an acute-phase protein indicative of inflammation. In murine colonic epithelial cells, lipopolysaccharide (LPS), a gram-negative bacterial antigen, strongly enhanced mRNA expression of SAA3, but not SAA1 or SAA2, suggesting that SAA3 might respond to bacterial infection in other epithelia. We examined SAA1/2 and SAA3 mRNA expression in murine alveolar epithelial cells exposed to LPS or the gram-positive bacterial antigen, lipoteichoic acid (LTA), using real-time PCR.

ATP-dependent potassium channels contribute to motor regulation of esophageal striated muscle in rats.

J Vet Med Sci

September 2019

Department of Basic Veterinary Science, Laboratory of Physiology, The United Graduate School of Veterinary Sciences, Gifu University, 1-1 Yanagaido, Gifu 501-1193, Japan.

Article Synopsis
  • The study focused on the role of ATP-dependent potassium channels (K channels) in controlling the movement of striated muscle in the esophagus.
  • In experiments, blocking K channels with glibenclamide led to stronger muscle contractions, while activating them with minoxidil weakened contractions.
  • The presence of K channel subunits was confirmed in esophageal tissue, suggesting they play a significant part in regulating muscle activity in this area.

Mild hypothermia causes a shift in the alternative splicing of cold-inducible RNA-binding protein transcripts in Syrian hamsters.

Am J Physiol Regul Integr Comp Physiol

August 2019

Laboratory of Physiology, Department of Basic Veterinary Science, The United Graduate School of Veterinary Sciences, Gifu University, Gifu , Japan.

Article Synopsis
  • Cold-shock proteins play a key role in the cold tolerance of hibernating animals, with a focus on an RNA-binding protein (CIRBP) that is rapidly induced in hamsters' hearts through alternative splicing.
  • While CIRBP mRNA is consistently expressed across various organs in nonhibernating hamsters, hibernating hamsters predominantly show a short variant of CIRBP necessary for function.
  • Maintaining hamsters in mild hypothermia for an adequate duration triggers a shift in alternative splicing, which mimics the gradual temperature decrease seen during natural hibernation.

Background: Degenerative myelopathy (DM) is a progressive neurodegenerative disease frequently found in Pembroke Welsh Corgis (PWCs). Most DM-affected PWCs are homozygous for the mutant superoxide dismutase 1 (SOD1) allele; however, the genetic examination for the SOD1 mutation does not exclusively detect symptomatic dogs. In order to identify novel biomarkers, the plasma microRNA (miRNA) profiles of PWCs with DM were investigated.

Article Synopsis
  • * The TgCatJpOk4 strain, isolated in Okinawa, shows high virulence in laboratory mice, similar to the highly virulent RH strain, but its effects on domestic animals were previously unknown.
  • * In a study using Microminipigs, four out of five infected pigs exhibited severe symptoms, with three succumbing, revealing that the TgCatJpOk4 strain also has significant virulence in pigs, causing interstitial pneumonia and liver necrosis.

Prevalence and pattern of thoracolumbar caudal articular process anomalies and intervertebral disk herniations in pugs.

J Vet Med Sci

June 2019

Joint Department of Veterinary Medicine, Faculty of Applied Biological Sciences, Gifu University, Gifu 501-1193, Japan.

Thoracolumbar intervertebral disk herniation (TL-IVDH) with caudal articular process anomalies has been reported in Pugs. It currently remains unclear whether congenital caudal articular process aplasia/hypoplasia predisposes to the development of TL-IVDH. However, there are difficulties in proving the causal relationship between caudal articular process anomalies and TL-IVDH.

Atlantoaxial instability (AAI)/subluxation commonly occurs in small breed dogs. Ventral stabilization techniques using screws and/or pins and a plate or, more commonly, polymethylmethacrylate are considered to provide the most favorable outcome. However, the implantation of screws of sufficient sizes for long-term stability becomes challenging in toy breed dogs (e.

The increase in antimicrobial-resistant (AMR) bacteria caused by antimicrobial usage is a public health problem. We investigated the proportion of cephalexin (LEX)-resistant bacteria in fresh feces obtained from antimicrobial-free broilers in three flocks at <15, 15-40, and> 40 days old. DHL agar plates containing 25 μg/mL LEX (DHL-L) showed LEX-resistant bacteria in all flocks at <15 days old and in one flock at > 40 days old.
