Introductory Analysis and Validation of CUT&RUN Sequencing Data.

The CUT&RUN technique facilitates detection of protein-DNA interactions across the genome. Typical applications of CUT&RUN include profiling changes in histone tail modifications or mapping transcription factor chromatin occupancy. Widespread adoption of CUT&RUN is driven, in part, by technical advantages over conventional ChIP-seq that include lower cell input requirements, lower sequencing depth requirements, and increased sensitivity with reduced background signal due to a lack of cross-linking agents that otherwise mask antibody epitopes.

Promoting Equitable Engagement in Attention-Deficit/Hyperactivity Disorder Care.

Attention-deficit/hyperactivity disorder (ADHD) is a treatable pediatric condition, but children with racial-ethnic minority backgrounds often do not receive timely or consistent treatment. Understanding how systemic racism impacts care and learning from families of color about their experiences can provide critical insights for improving clinical practice and engaging patients equitably in ADHD care. We interweave a mother's experience navigating ADHD care for her son with commentary from an interprofessional team about what clinicians can do for families to reduce the impact of systemic racism on care.

TeenGrowth: Individualized Estimations of Weight-Related Risk and Recovery Metrics for Young People With Eating Disorders.

Objective: While weight restoration and/or stabilization is crucial for successful treatment and sustained recovery from restrictive eating disorders (EDs), it is often challenging to define an individual's expected healthy body weight. This paper introduces the TeenGrowth package and its web-based application, designed to calculate and forecast predicted body mass index (BMI) and weight across adolescence.

Method: TeenGrowth includes functions for data cleaning, predicted BMI z-score and BMI calculations, and growth forecasting.

Flow-Through Flaps as a Robust Method for Advanced Free Flap Reconstruction in the Head and Neck: A Multi-Institutional Experience.

Background: Flow-through flaps (FTFs) are an advanced technique in which a free flap is anastomosed to the pedicle of another free flap to reconstruct extensive head and neck defects when recipient vessels are scarce.

Methods: A multi-institutional cohort of FTFs used for head and neck reconstruction were reviewed. For comparison, FTF outcomes were compared to free flaps that required vein grafts (VG) to reach distant recipient vessels.

Effects of implant status and breed type on feedlot performance, carcass characteristics, sera metabolites, and immunohistochemical responses in finishing steers.

The number of beef × dairy animals entering feedlots has increased, but the response of beef × dairy cattle to growth-promoting implants has not been well characterized. The objective of this study was to evaluate the effects of breed type and implant administration on live performance, carcass characteristics, sera metabolites, and immunohistochemical (IHC) outcomes. Forty-eight steers (average body weight [BW] = 417±22 kg) were sorted by breed into groups of predominantly Angus (B), black-hided beef × primarily Holstein (B×D), or Holstein (D), and half of the steers within each breed type were administered a steroidal implant.

The uses of right heart catheterization in cardio-pulmonary disease: State-of-the-art.

The right heart catheterization (RHC) remains an important diagnostic tool for a spectrum of cardiovascular disease processes including pulmonary hypertension (PH), shock, valvular heart disease, and unexplained dyspnea. While it gained widespread utilization after its introduction, the role of the RHC has evolved to provide valuable information for the management of advanced therapies in heart failure (HF) and cardiogenic shock (CS) to name a few. In this review, we provide a comprehensive overview on the indications, utilization, complications, interpretation, and calculations associated with RHC.

Trends in stroke-related mortality in atrial fibrillation patients in the United States: Insights from the CDC WONDER database.

Background: Stroke associated with atrial fibrillation (AF) is a significant cause of mortality. This study analyzed demographic trends and disparities in mortality rates due to stroke in AF patients aged ≥25 years.

Methods: A retrospective analysis was conducted to acquire death data using the Centers for Disease Control and Prevention database from 1999 to 2020.

Persistent microbial infections and idiopathic pulmonary fibrosis - an insight into pathogenesis.

Interstitial lung disease (ILD) is characterized by chronic inflammation and scarring of the lungs, of which idiopathic pulmonary fibrosis (IPF) is the most devastating pathologic form. Idiopathic pulmonary fibrosis pathogenesis leads to loss of lung function and eventual death in 50% of patients, making it the leading cause of ILD-associated mortality worldwide. Persistent and subclinical microbial infections are implicated in the acute exacerbation of chronic lung diseases.

Surface interactions of gelatin-sourced carbon quantum dots with a model globular protein: insights into carbon-based nanomaterials and biological systems.

Carbon nanomaterials (CNMs), such as carbon nanotubes (CNTs), graphene quantum dots (GQDs), and carbon quantum dots (CQDs), are prevalent in biological systems and have been widely utilized in applications like environmental sensing and biomedical fields. While their presence in human matrices is projected to increase, the interfacial interactions between carbon-based nanoscopic platforms and biomolecular systems continue to remain underexplored. In this study, we investigated the effect of gelatin-sourced CQDs on the globular milk protein beta-lactoglobulin (BLG).

Background: Identifying risk factors for local recurrence (LR) is pivotal in optimizing rectal cancer treatment. Total mesorectal excision (TME) and lateral lymph node dissection (LLND) are the standard treatment for advanced low rectal cancer in Japan. However, large-scale studies to evaluate risk factors for LR are limited.

Aerodigestive Sequelae and Triple Endoscopy after Congenital Tracheoesophageal Fistula Repair in Children.

Objective: To identify factors associated with positive findings on triple endoscopy following neonatal TEF repair.

Civic identity: media, belonging, & Latiné youth in the 2020 US presidential election.

We explore how Latiné immigrant-origin youth used and made sense of media to enact their civic identities during the 2020 US presidential election cycle. In a polarized national context, this project highlights the voices and experiences of the participants (n=39). We conducted and analysed semi-structured interviews to identify three comprehensive themes.

Activation of mechanoreceptor Piezo1 inhibits enteric neuronal growth and migration .

Introduction: Dysfunction of the enteric nervous system (ENS) is linked to a myriad of gastrointestinal (GI) disorders. Piezo1 is a mechanosensitive ion channel found throughout the GI tract, but its role in the ENS is largely unknown. We hypothesize that Piezo1 plays an important role in the growth and development of the ENS.

Carotid Body Tumor: Incidental Discovery and Diagnosis.

Carotid body tumors (CBTs), rare neuroendocrine neoplasms near the carotid bifurcation, are mostly asymptomatic but may cause discomfort and autonomic dysfunction. Computed tomography angiography (CTA) is used for diagnosis, eliminating the need for a biopsy to avoid the risk of hemorrhage. Surgical excision is the preferred treatment, while radiotherapy is an option when surgery is impractical.

Purple Tea (Camellia sinensis var. assamica) Leaves and Obesity Management: A Review of 1,2-Di-Galloyl-4,6-Hexahydroxydiphenoyl-β-D-Glucose's (GHG) Potential Health Benefits, and Future Prospects.

Purple tea ( var. ) is a distinct variety of  known for its bioactive compounds, including caffeine, catechins, and a unique compound called 1,2-di-Galloyl-4,6-Hexahydroxydiphenoyl-β-D-Glucose, (GHG) found predominantly in purple tea leaves, which shows potential in obesity management. Studies have indicated that these bioactive compounds play a significant role in reducing BMI and body weight among obese patients.

Traumatic Cystic Artery Pseudoaneurysm After Transjugular Liver Biopsy: A Case Report.

Cystic artery pseudoaneurysms are a rare but life-threatening entity that commonly occurs as a sequela to acute cholecystitis. We present a case of a 52-year-old male with a past medical history of decompensated alcoholic liver cirrhosis who underwent a transjugular liver biopsy (TJLB) after correction of his baseline coagulopathy. On post-operative day one, the patient had significant blood loss with an inappropriate response to blood transfusions and without an identifiable source of bleeding.

Single-Nucleotide Polymorphisms in Thrombotic Thrombocytopenic Purpura: A Genetic Predisposition to Immune Thrombotic Thrombocytopenic Purpura.

There are two main classifications for thrombotic thrombocytopenic purpura (TTP): immune and hereditary. The majority of TTP cases are immune in nature and are due to inhibitor autoantibodies against ADAMTS13. Hereditary TTP is caused by biallelic pathogenic variants in the ADAMTS13 gene.

Improving Advanced Care Planning Discussions at an Internal Medicine Clinic.

Objective: The project aimed to standardize advanced care planning (ACP) at an internal medicine clinic by initiating physician-patient communication regarding the patient's knowledge, understanding, and openness to pursuing advanced medical directives.

Methods: Data collection was conducted from February 1 to April 1, 2024, with the study concluding on April 24, 2024. ACP was facilitated through an initial standardized six-question pre-intervention survey in both English and Spanish.

Background: Uptake of COVID-19 vaccines has stalled in the U.S. Some studies suggest that medical mistrust may be a barrier, but evidence is limited due to cross-sectional designs or convenience sampling.

The caribou ( sspp.) is a keystone wildlife species in northern ecosystems that plays a central role in the culture, spirituality and food security of Indigenous People. The Arctic is currently experiencing an unprecedented rate of climate change, including warming temperatures and altered patterns of precipitation.

TClC effectively suppresses the growth and metastasis of NSCLC via polypharmacology.

Despite significant advances in targeted therapies and immunotherapies, non-small cell lung cancer (NSCLC) continues to present a global health challenge, with a modest five-year survival rate of 28 %, largely due to the emergence of treatment-resistant and metastatic tumors. In response, we synthesized a novel bioactive compound, ethyl 6-chlorocoumarin-3-carboxylyl L-theanine (TClC), which significantly inhibited NSCLC growth, epithelial mesenchymal transition (EMT), migration, and invasion and tumor growth and metastasis without inducing toxicity. TClC disrupts autocrine loops that promote tumor progression, particularly in stem-like CD133-positive NSCLC (CD133+ LC) cells, which are pivotal in tumor metastasis.
