Mutants can be generated by physical or chemical mutagens. The procedure includes mutagenesis treatment, post-treatment, planting of M, harvest of M, planting of M, phenotypic observation or mutation scanning in M plants. Here we describe the combined treatment of the rice seeds from the indica variety Tenuo with Cs gamma rays at 100 Gy and a chemical mutagen-ethyl methanesulfonate (EMS).

Step emulsification is a low-shear method to produce monodispersed microdroplets by spontaneous breakup of dispersed fluid at a spatial "step". As a semi-open microfluidic system, controllable coalescence of multiple components in step emulsification has not been achieved. Here, we use a low voltage to control the coalescence position of flow tips in the terrace.

Target Induced Local Lesions In Genomes (TILLING) is a strategy of reverse genetics for the high-throughput screening of induced mutations. However, the TILLING system has less applicability for insertion/deletion (Indel) detection and traditional TILLING needs more complex steps, like CEL I nuclease digestion and gel electrophoresis. To improve the throughput and selection efficiency, and to make the screening of both Indels and single base substitions (SBSs) possible, a new high-resolution melting (HRM)-based TILLING system is developed.
