Electronic address:

Background: The broad use of bedaquiline and pretomanid as the mainstay of new regimens to combat tuberculosis is a risk due to increasing bedaquiline resistance. We aimed to assess the safety, bactericidal activity, and pharmacokinetics of BTZ-043, a first-in-class DprE1 inhibitor with strong bactericidal activity in murine models.

Methods: This open-label, dose-expansion, randomised, controlled, phase 1b/2a trial was conducted in two specialised tuberculosis sites in Cape Town, South Africa.

From responsibility to responsibilization in stem cell research: An ethical framework.

Stem Cell Reports

January 2025

Department of Bioethics and Health Humanities, Julius Center for Health Sciences & Primary Care, University Medical Center Utrecht, PO Box 8500, 3508 GA Utrecht, the Netherlands.

Adopting a responsibilization approach can further improve the ethical conduct of stem cell (SC) research and applications. This approach helps align new and existing solutions for ethical implications by focusing on equipping SC researchers with the knowledge, skills, and organizational arrangements to take (co-)responsibility for the socio-ethical implications of their research.

An accurate and robust multicomponent quantitative analysis method: Molar mass coefficient method.

J Chromatogr A

December 2024

Institute of Chinese Materia Medica, China Academy of Chinese Medical Sciences, Beijing, 100700, China. Electronic address:

Multicomponent quantitative analysis (MCQA) is necessary for comprehensively characterizing the quality of complex samples, including medicines, foods. However, the limited supply of reference substances and the high costs associated with testing hinder the application of the MCQA using the external standard (ES) method. Here we propose a Molar Mass Coefficient (MMC) method for the quantification of multiple compounds with identical chromophore utilizing a single reference compound (SRC) by a UV detector.

Gut microbiome and immune checkpoint inhibitor toxicity.

Eur J Cancer

January 2025

Department of Medical Oncology, University Medical Center Utrecht, Utrecht University, PO box 85500, Utrecht 3584 CX, the Netherlands. Electronic address:

Background: Multiple studies have suggested that gut microbiome may influence immune checkpoint inhibitor (ICI) efficacy, but its association with immune-related adverse events (irAEs) is less well studied. In this prospective cohort study, we assessed whether gut microbiome composition at start, or changes during ICI, are associated with severe irAEs.

Methods: Stool samples of cancer patients treated with anti-PD-1 ± anti-CTLA-4 were analyzed using 16S rRNA gene sequencing and metagenomic shotgun sequencing.

Mitochondrial disorder diagnosis and management- what the pediatric neurologist wants to know.

Eur J Paediatr Neurol

December 2024

University Children's Hospital, Paracelsus Medical University (PMU), Salzburg, Austria; Amalia Children's Hospital, Department of Paediatrics, Radboudumc, Nijmegen, the Netherlands. Electronic address:

Childhood-onset mitochondrial disorders are rare genetic diseases that often manifest with neurological impairment due to altered mitochondrial structure or function. To date, pathogenic variants in 373 genes across the nuclear and mitochondrial genomes have been linked to mitochondrial disease, but the ensuing genetic and clinical complexity of these disorders poses considerable challenges to their diagnosis and management. Nevertheless, despite the current lack of curative treatment, recent advances in next generation sequencing and -omics technologies have laid the foundation for precision mitochondrial medicine through enhanced diagnostic accuracy and greater insight into pathomechanisms.

Amsterdam Institute for Life and Environment (A-LIFE), Section Systems Ecology, Vrije Universiteit Amsterdam, De Boelelaan 1085, 1081HV, Amsterdam, the Netherlands.

Lichens play important roles in habitat formation and community succession in polar and alpine ecosystems. Despite their significance, the ecological effects of lichen traits remain poorly researched. We propose a trait trade-off for managing light exposure based on climatic harshness.

Grey mullets (family Mugilidae) are widespread across coastal, brackish, and freshwater habitats, and have supported fisheries for millennia. Despite their global distribution and commercial value, little is known about their movement ecology and its role in the co-existence of sympatric mullet species. Gaps in knowledge about migratory behaviour, seasonal occurrence, and movement scales have also impeded effective management, highlighting the need for further research.

Background: Addressing the growing challenge of nurse retention requires coordinated actions at national and global levels to improve recruitment, retention policies, and investments in the nursing work environment. The nursing work environment, defined as the "organizational characteristics of a work setting that facilitate or constrain professional nursing practice", is critical in influencing whether nurses decide to leave their jobs. This study investigates the impact of differentiated nursing practices - which involved tailoring roles and responsibilities based on nurses' training, skills, and experience in Dutch hospitals - and investigated their impact on the nursing work environment and turnover intention (i.

Objectives: To expand a translational pharmacokinetic-pharmacodynamic (PKPD) modelling approach for assessing the combined effect of polymyxin B and minocycline against Klebsiella pneumoniae.

Methods: A PKPD model developed based on in vitro static time-kill experiments of one strain (ARU613) was first translated to characterize that of a more susceptible strain (ARU705), and thereafter to dynamic time-kill experiments (both strains) and to a murine thigh infection model (ARU705 only). The PKPD model was updated stepwise using accumulated data.

Agouti-related peptide (AgRP) is a well-established potent orexigenic peptide primarily expressed in hypothalamic neurons. Nevertheless, the expression and functional significance of extrahypothalamic AgRP remain poorly understood. In this study, utilizing histological and molecular biology techniques, we have identified a significant expression of Agrp mRNA and AgRP peptide production in glomus type I cells within the mouse carotid body (CB).

Department of Rehabilitation Medicine, Amsterdam UMC, University of Amsterdam, Meibergdreef 9 1105 AZ, Amsterdam, the Netherlands; Amsterdam Movement Sciences, Rehabilitation & Development, Amsterdam, the Netherlands.

The offloading effectiveness of custom-made footwear for people with diabetes is assessed using plantar pressure measurements. While such pressure data is multidimensional, it is mostly analyzed using a scalar - maximum peak plantar pressure (PMax). We aimed to investigate the associations between multiple peak plantar pressure parameters for footwear assessment and determine whether this assessment depends on the chosen parameter.
