Objective: Serum p53 protein overexpression was detected in population exposed to traffic exhaust gas to study the relation between traffic exhaust gas and the increased risk in p53 gene mutation.
Methods: Serum p53 protein expression was measured by enzyme-linked immunosorbent assay. Relationship between different types of job and serum p53 protein overexpression were studied by pearson Chi-square tests.

Objective: To observe the survival of hand allograft under the state of immunosuppression and the pathological changes of rejection in the recovery process.
Methods: The biopsies of the skin, nerve, muscle, tendon and bone tissue of hand allografts during different stages from 1 day to 7 months after operation were observed using routine histological technique.
Results: No significant changes due to rejection in skin, nerve, muscle and bone tissue were observed.

O-(3-[(18)F]fluoropropyl)-L-tyrosine (FPT), an analogue of O-(2-[(18)F]fluoroethyl)-L-tyrosine (FET) as an amino acid tracer for tumor imaging with positron emission tomography (PET), was synthesized and evaluated. FPT was prepared by [(18)F]fluoropropylation of L-tyrosine in a two-step procedure. Biodistribution of FPT was determined in normal mice.

The aim of this study was to explore the sensitivity and specificity of interphase fluorescence in situ hybridization (I-FISH) technique in monitoring the tumor load of chronic myeloid leukemia (CML) during treatment.
Methods: Ten cases of normal bone marrow cells were examined using I-FISH technique to define the normal cut-off value.

Objective: To investigate the contribution of angiotensin II towards the process of hepatic fibrosis that is largely due to hepatic stellate cell growth.
Methods: Adult rat hepatic stellate cells were cultured and checked for the expression of angiotensin II receptor 1a (AT(1a)) mRNA by RT-PCR and sequence analysis. The effects of angiotensin II were observed on stimulation of hepatic stellate cell growth detected by MTT assays, (3)H-thymidine incorporation and cell count, and collagen synthesis by (3)H-proline incorporation.

Recently, a new immunotherapy against cancer based on a novel tumor-associated antigen of the human telomerase reverse transcriptase (hTERT) is being investigated. Antigen peptides derived from hTERT are expressed and presented with major histocompatibility complex (MHC) class I molecules in tumor cells, and elicit ex vivo cytotoxic T-lymphocyte (CTL) responses in healthy individuals and cancer patients. Data from both human and murine systems demonstrate that TERT-special CTL kill TERT-positive tumor cells of various histological origins in a MHC-restricted fashion.

Background & Objective: Glivec was approved by Food & Drug Administration (FDA) in May 2001 as a gene target drug for treatment of chronic myeloid leukemia (CML) and showed a good curative effect for patients with chronic myeloid leukemia in chronic phase. But its effectiveness was poor in patients with CML blast phase treated with Glivec alone. Glivec was reported having synergetic effect with other chemical agents in vitro, but there is few report in clinical combined application.

Methods: (C57BL/6 x 615) F1 (H-2bk) mice which was inoculated with L615 cells to develop leukemic murine model was the recipient. The healthy C57BL/6 (H-2b) mice was the donor.

Results: Out of 16 CML patients, the results of FISH was not consistent with that of nested RT-PCR in 3 cases.

Objective: To study the recombinant human interleukin 11 (rhIL-11) in combination with granulocyte-colony stimulating factor (rhG-CSF) for mobilizing peripheral blood stem/progenitor cells in C57BL/6 mice.
Methods: rhIL-11,250 micro g x kg(-1) x d(-1) per mouse alone or in combination with rhG-CSF 250 micro g x kg(-1) x d(-1) per mouse was administered to C57BL/6 mice from day 1 to 7. The changes of peripheral white blood cell count (WBC), platelet counts (BPC) and hematopoietic stem/progenitor cells yields were observed.

To study the correct method for determining ABO blood types in infants and its influencing factors, blood types of 33 infants under 6 months old were determined by routine serological method, micro-column gel typing system and PCR-SSP genotyping method. Of the 33 cases with discrepant results of ABO blood type by different methods, the blood types of 32 cases were discrepant between red cell and serological typings in the routine serological method, and a false coincidence in 1 case was caused by bacterial infection resulting in B-like antigen. Correct blood typing was obtained in 27 cases with a correct rate of 84.

In order to observe the curative and side effects in malignant hematologic diseases treated with autologous peripheral blood stem cell transplantation (auto-PBSCT) combined with halotype lymphocyte infusion, auto-PBSCs were mobilized, harvested and stored at -196 degrees C from patients in first CR or PR with intensive chemotherapy (Ara-C 1.0 g/m(2) x 5 days or cyclophosphamide 60 mg/kg x 2 days) and G-CSF. Unpurged auto-PBSCs were infused when patients received the conditioning regimen with busulfan, total irradiation or cyclophosphamide.

To clarify the association between HLA-DPB1 alleles and chronic myelogenous leukemia (CML) in South Chinese, the allelic types of HLA-DPB1 were detected by sequence based typing (SBT) in 86 patients with CML and 82 healthy individuals from Southern China. The results showed that the frequencies of HLA-DPB1 * 1301 and DPB1 * 20011 were higher in patients with CML in comparison with those of healthy individuals. It is concluded that positive association may exist between certain HLA-DPB1 alleles and CML.

This study was designed to explore the influence of STI571, a tyrosine kinase inhibitor, on the expression of c-kit in the bone marrow cells from patients with acute non-lymphocytic leukemia (ANLL). The cells were exposed to various concentration of STI571 for 72 hours, the expression of c-kit mRNA and CD117 was assayed by RT-PCR and flow cytometry, respectively.
