[Ocular adnexal mucosa-associated lymphoid tissue lymphoma: a pathological and imaging study].

Purpose: To analyze the relationship of the pathological and clinical features of patients with ocular adnexal mucosa-associated lymphoid tissue (MALT) type lymphoma, and to provide a suggestion of preoperative diagnosis and treatment of MALT lymphoma.

Methods: To analyze the clinical, imaging and histopathologic features of 20 cases (25 eyes) of MALT lymphoma retrospectively in the department of ophthalmology.

Results: Among the 20 cases (25 eyes), 18 cases (21 eyes)were located in the orbit, and 2 cases (4 eyes) were at conjunctiva.

Cloning and expression and immunogenicity of Helicobacter pylori BabA2 gene.

Aim: To construct a recombinant strain which expresses BabA of Helicobacter pylori (H pylori) and to study the immunogenicity of BabA.

Methods: BabA2 DNA was amplified by PCR and inserted into the prokaryotie expression vector pET-22b (+) and expressed in the BL21 (DE3) E.coli strain.

Construction of attenuated Salmonella typhimurium Strain expressing Helicobacter pylori conservative region of adhesin antigen and its immunogenicity.

Aim: To construct a non-resistant and attenuated Salmonella typhimurium (S. typhimurium) strain which expresses conservative region of adhesion AB of Helicobacter pylori (H pylori) and evaluate its immunogenicity.

Methods: The AB gene amplified by PCR was inserted into the expression vector pYA248 containing asd gene and through two transformations introduced into the delta Cya, delta Crp, delta Asd attenuated Salmonella typhimurium strain, constructing balanced lethal attenuated Salmonella typhimurium strains X4072 (pYA248-AB).

[Efficacy of 3-dimensional conformal hypofractionated single high-dose radiotherapy combined with transcatheter arterial chemoembolization for portal vein tumor thrombus in patients with hepatocellular carcinoma].

Background & Objective: Hepatocellular carcinoma (HCC) often results in portal vein tumor thrombus (PVTT), and the prognosis of the patients is extremely poor. It has been reported that some oversea scholars achieved fine therapeutic effects in treatment of this disease by 3-dimensional conformal radiation therapy (3DCRT) in fractionated conventional dose. The aim of this study was to evaluate the efficacy of 3-dimensional conformal hypofractionated single high-dose radiotherapy combined with transcatheter arterial chemoembolization (TACE) for portal vein tumor thrombus in patients with hepatocellular carcinoma.

[Method of detection of soluble HLA-I and soluble HLA-I level alteration in storage blood].

Aim of this study was to develop the detection method of soluble human leukocyte antigens I (sHLA-I) and to explore sHLA-I level alteration in storage blood and its significance. sHLA-I level in sera of 60 Guangdong normal individuals and sHLA-I concentration in blood components from 20 donors quantitatively were detected by sandwich ELISA. The results showed that sensitivity of this assay was 2.

Assessment of regional myocardial blood flow with myocardial contrast echocardiography: an experimental study.

Objectives: The aim of this study was to verify the accuracy of using myocardial contrast echocardiography (MCE), to quantify regional myocardial blood flow (MBF), and to evaluate myocardial viability in comparison to that measured by radiolabeled microsphere and pathologic examination.

Methods: Epicardial MCE was obtained in five myocardial ischemic dogs with constant microbubble intravenous infusion. After the video intensity (VI, y) versus pulsing interval plots derived from each myocardial pixel were fitted to an exponential function: y = A(1 - e(-beta t)), the MBF was calculated as the product of A (microvascular cross-sectional area or myocardial blood volume) and beta (mean myocardial microbubble velocity).

[Cloning of mouse TLR-2 gene and its expression in Pichia pastroris].

Aim: To clone the mouse TLR-2 gene and to express it in Pichia pastroris.

Methods: Full-length gene encoding mouse Toll-like receptor 2 (mTLR-2) was amplified by RT-PCR, cloned into pUCm-T vector, and confirmed by sequencing. The target gene was then inserted into Pichia pastroris expression vector pPICZalphaC, which was transformed into Pichia pastroris.

[Quantitative study of diffusion weighted imaging on magnetic resonance imaging in focal hepatic lesions less than 3 cm].

Objective: To evaluate the characteristics of MR imaging of hepatic lesions using measurement of apparent diffusion coefficient (ADC) value of hepatic lesions on diffusion weighted imaging.

Methods: MR diffusion weighted images were obtained in patients with 97 hepatic lesions (22 hepatocellular carcinomas, 21 metastatic tumors, 28 hemangiomas, 26 cysts). ADC values were evaluated with different sequences.

[Evaluation for degree of malignancy and prognosis of patients with brain glioma using 99Tcm-MIBI brain SPECT].

Background & Objective: It is important and useful to evaluate the degree of malignancy for therapeutic scheme and prognosis evaluation. Traditional imaging methods such as CT and MRI play the important role in distinguishing glioma, but sometimes it was still difficult to diagnose accurately only having the aid of these anatomic information. Stereotaxic needle biopsy is an invasive method and indicates only local pathologic change, sometimes the error of sample collection will result in inaccurate grading.

[Study of gene expression profile of chronic myeloid leukemia in chronic and blastic phases].

Objective: To study the gene expression profile of chronic myeloid leukemia (CML) in chronic and blast phases for providing information about the potential mechanisms of blastic crisis.

Methods: The gene expression difference of bone marrow mononuclear cells between CML in chronic and in blastic phases was examined with DNA array.

Results: In the blastic phase, 68 genes were obviously up-regulated in the 1176 tested genes.

[Finger reconstruction with extended free second toe flap transfer].

Objective: To investigate an ideal method for finger reconstruction with extended the second toe flap transfer.

Methods: The second toe free flap was created, combined with an pedicled skin flap from the fibular side of the great toe inlaid in the ventral side of the second toe, a double-wing flap and the distal part of the metatarsal bone. The composite free flap was transferred by vascular anastomosis for finger reconstruction.

[High efficient generation of recombinant adenovirus containing human Fas gene using a method of homologous recombination in bacteria].

Objective: To generate recombinant adenovirus with human Fas gene and transfect the Fas gene into keloid-derived fibroblasts to take place of the dysfunctional Fas gene, reconstruct the blocked Fas signal.

Methods: We used a new bacterial homologous recombination system (Ad-Easy system) to construct recombinant adenovirus vector. Fas gene was cut down from the PMD-T-Fas plasmid and then transferred to track plasmid.

Hydrogen peroxide in the Burkitt's lymphoma cell line Raji provides protection against arsenic trioxide-induced apoptosis via the phosphoinositide-3 kinase signalling pathway.

Many anticarcinogenic drugs kill tumour cells by inducing apoptosis. We examined the effects of hydrogen peroxide (H(2)O(2)) on arsenic trioxide (As(2)O(3))-induced cell killing. Low concentrations of H(2)O(2) (200 micromol/l) inhibited the ability of As(2)O(3) to induce apoptosis in the Burkitt's lymphoma cell line Raji.

Phosphatidylinositol 3-kinase signaling is involved in neurogenesis during Xenopus embryonic development.

Phosphatidylinositol 3-kinase (PI3K) has numerous cellular functions, including cell survival and proliferation. In this study, we demonstrated that the expression of the active form of PI3K induced dorsal differentiation and axis duplication and strongly induced the expression of neural markers. In contrast, the inhibition of PI3K activity by its dominant negative mutant induced the phenotype of losing posterior structures and the expression of ventral markers.

Construction of a metastasis-associated gene subtracted cDNA library of human colorectal carcinoma by suppression subtraction hybridization.

Aim: To construct a differentially-expressed gene subtracted cDNA library from two colorectal carcinoma (CRC) cell lines with different metastatic phenotypes by suppression subtractive hybridization.

Methods: Two cell lines of human CRC from the same patient were used. SW620 cell line showing highly metastatic potential was regarded as tester in the forward subtractive hybridization, while SW480 cell line with lowly metastatic potential was treated as tester in the reverse hybridization.

[Clinical significance of neutralizing anti-interferon antibodies in chronic hepatitis B patients treated with recombinant interferon-alpha].

Objective: To investigate the clinical significance of neutralizing anti-interferon-alpha antibodies (NA) in chronic hepatitis B patients treated with recombinant interferon-alpha(rIFN-alpha).

Methods: One hundred and eighty-one patients (128 male and 53 female) with histological proven chronic hepatitis B were treated with 5 MU recombinant interferon-alpha 1b (rIFN-alpha 1b) subcutaneously thrice weekly for 6 to 37 (median 10) months. For each patient, Specific detection of serum HBV DNA level with fluorescent-quantitative PCR, HBeAg with enzymoimmunoassay and NA with an antiviral neutralizing biological assay were performed during therapy.

[Characteristic of distortion product otoacoustic emissions for preterm newborn].

Objective: To investigate the characteristic of distortion product otoacoustic emissions (DPOAE) amplitude, SNR and noise in preterm newborns.

Method: DPAOEs at the 2f1-f2 frequency were recorded from 59 ears of 31 preterm newborns and 35 ears of 19 healthy fullterm newborns in response to equilevel primary tones at postnatal 3 days to 1 week. Detailed test included the recording of DPAOEs, SNR and noise.

[Clinical efficacy and side effects of STI571 in treatment of patients with chronic myeloid leukemia].

Background & Objective: The aberrant regulation of the protein tyrosine kinase (PTK) activity of P210(BCR-ABL), which is the protein product of Bcr-Abl fusion gene leads to the pathogenesis of chronic myeloid leukemia (CML). Though STI571 can inhibit specifically the PTK activity of P210(Bcr-Abl) and greatly improve the clinic curative effect on CML in chronic phase, its effect on CML in accelerated phase and blast crisis is not clear. In this article, we attempted to analyze the clinic efficacy and side effect of STI571 treatment on CML patients in different phase.

[Testosterone regulation of androgen receptor protein expression in cultured vascular smooth muscle cells].

Objective: To investigate the effects of testosterone exposure on androgen receptor (AR) protein expression in vascular smooth muscle cells (VSMC) and the possible mechanisms mediating the effects.

Methods: VSMC were cultured from thoracic aorta of male Sprague-Dawley rats by using the explant method. Cytoplasmic and nuclear extracts were prepared by means of cell lysis and high salt extraction respectively, and subjected to western blotting analysis for determination of AR protein level.
