The high-dynamic, high-loading environment in the joint cavity puts urgent demands on the cartilage regenerative materials with shear responsiveness and lubrication. Here, a new type of injectable hydrogel composed of oxidized hyaluronic acid (OHA), adipic dihydrazide-grafted hyaluronic acid (HA-ADH), oxidized chondroitin sulfate (OChs), and decellularized extracellular matrix methacrylate (dECMMA) was fabricated. The aldehyde groups in OHA and OChs reacted with the amino groups in HA-ADH to form a dynamic hydrogel, which was then covalently crosslinked with dECMMA to create a dual-crosslinked hydrogel with sufficient mechanical strength.

Repair of infectious bone defects remains a serious problem in clinical practice owing to the high risk of infection and excessive reactive oxygen species (ROS) during the early stage, and the residual bacteria and delayed Osseo integrated interface in the later stage, which jointly creates a complex and dynamic microenvironment and leads to bone non-union. The melatonin carbon dots (MCDs) possess antibacterial and osteogenesis abilities, greatly simplifying the composition of a multifunctional material. Therefore, a multifunctional hydrogel containing MCDs (GH-MCD) is developed to meet the multi-stage and complex repair needs of infectious bone injury in this study.

Pralsetinib demonstrated impressive improvement of survival in non-small cell lung cancer (NSCLC) patients harbored de novo RET fusion. However, the efficacy in patients with acquired RET fusion after resistance to EGFR/ALK-TKIs has only been reported on a case-by-case basis, and the strategy for overcoming the acquired RET fusion has not been fully investigated. This study analyzed targeted sequencing data from 6530 tissue samples from patients with metastatic Chinese colorectal cancer (CRC) to identify low mutation frequency and subgroup-specific driver genes, using three algorithms for overall CRC as well as across different clinicopathological subgroups. We analyzed 425 cancer-related genes, identifying 101 potential driver genes, including 36 novel to CRC. Notably, some genes demonstrated subgroup specificity; for instance, ERBB4 was found as a male-specific driver gene and mutations of ERBB4 only influenced the prognosis of male patients with CRC.
