Background: Pancreatic ductal adenocarcinoma (PDAC) often presents with liver or peritoneal metastases at diagnosis. Despite similar treatment approaches, patient outcomes vary between these metastatic sites. To improve targeted therapies for metastatic PDAC, a comprehensive analysis of the genetic profiles and evolutionary patterns at these distinct metastatic locations is essential.

Self-propelled nanomotors possess strong propulsion and penetration abilities, which can increase the efficiency of cellular uptake of nanoparticles and enhance their cytotoxicity against tumor cells, opening a new path for treating major diseases. In this study, the concept of driving nanomotors by alternately stretching and contracting a temperature-sensitive polymer (TS-P) chain is proposed. The TS-Ps are successfully linked to one side of CuSe@Au (CS@Au) nanoparticles to form a Janus structure, which is designated as CuSe@Au-polymer (CS@Au-P) nanomotors.

Article Synopsis
  • - The renin-angiotensin system (RAS) and the sympathetic nervous system (SNS) work together to regulate blood pressure, with prenatal exposure to lipopolysaccharide (LPS) driving increased RAS activity and sympathetic outflow in the paraventricular nucleus (PVN) of the brain.
  • - This exposure leads to an imbalance of inhibitory and excitatory neurons, increased reactive oxygen species (ROS), and ultimately contributes to hypertension in offspring rats.
  • - Melatonin treatment during pregnancy shows potential in reducing RAS activity and blood pressure, indicating it may help protect against hypertension caused by prenatal LPS exposure.

Nat Med

September 2024

Section of Esophageal and Mediastinal Oncology, Department of Thoracic Surgery, National Cancer Center/National Clinical Research Center for Cancer/Cancer Hospital, Chinese Academy of Medical Sciences and Peking Union Medical College, Beijing, China.

Recent single-arm studies involving neoadjuvant camrelizumab, a PD-1 inhibitor, plus chemotherapy for resectable locally advanced esophageal squamous cell carcinoma (LA-ESCC) have shown promising results. This multicenter, randomized, open-label phase 3 trial aimed to further assess the efficacy and safety of neoadjuvant camrelizumab plus chemotherapy followed by adjuvant camrelizumab, compared to neoadjuvant chemotherapy alone. A total of 391 patients with resectable thoracic LA-ESCC (T1b-3N1-3M0 or T3N0M0) were stratified by clinical stage (I/II, III or IVA) and randomized in a 1:1:1 ratio to undergo two cycles of neoadjuvant therapy.

Innate immunity is the first line of defense in the human body, and it plays an important role in defending against viral infection. Viruses are identified by different pattern-recognition receptors (PRRs) that activate the mitochondrial antiviral signaling protein (MAVS) or transmembrane protein 173 (STING), which trigger multiple signaling cascades that cause nuclear factor-κB (NF-κB) and interferon regulatory factor 3 (IRF3) to produce inflammatory factors and interferons (IFNs). PRRs play a pivotal role as the first step in pathogen induction of interferon production.

Multimodal mechanisms of pathogenic variants in the signal peptide of FIX leading to hemophilia B.

Blood Adv

August 2024

Henan International Joint Laboratory of Thrombosis and Hemostasis, College of Basic Medicine and Forensic Medicine, Henan University of Science and Technology, Luoyang, People's Republic of China.

Article Synopsis
  • Signal peptide (SP) is crucial for the secretion of factor IX (FIX), and variants in this region can lead to hemophilia B (HB), though the exact mechanism behind the genotype-phenotype correlation is not well understood.
  • In a study of 13 pathogenic point variants in the SP, researchers found that these mutations contribute to HB mainly through missense mutations affecting protein function and abnormal pre-mRNA splicing.
  • The study revealed that variants in specific areas of the SP disrupt its translocation and cleavage processes, leading to FIX deficiency, while vitamin K availability was shown to influence the severity of bleeding in patients with the same genetic variant.
