Background: Hepatocellular Carcinoma (HCC) is the most common type of primary liver cancer, accounting for the majority of liver cancer cases. Hepatocellular Carcinoma not only exhibits high heterogeneity but also possesses an immune-suppressive tumor microenvironment that promotes tumor evasion, posing substantial difficulties for efficient therapy. Our aim is to utilize single-cell RNA transcriptome data to investigate the dynamic changes in the tumor microenvironment during the malignant progression of HCC, the communication among immune cells, and the marker genes associated with patient prognosis.

Article Synopsis
  • Hypertrophic cardiomyopathy (HCM) is a hereditary condition characterized by thickening of the heart muscle, contributing significantly to cardiac-related deaths, with ongoing research exploring the role of immune inflammation in its development.
  • Five machine learning algorithms were used to identify five new biomarkers (DARS2, GATM, MGST1, SDSL, ARG2) linked to HCM, with GATM and MGST1 showing strong diagnostic potential based on their elevated expression in HCM tissues compared to normal tissues.
  • A new risk assessment model utilizing GATM and MGST1 was developed and validated, demonstrating effective performance in distinguishing HCM patients, while also indicating these markers have prognostic relevance in both HCM and various cancers.

Neutrophil and neutrophil extracellular traps (NETs) were reported to be associated with tumor development, but the exact role and concrete mechanisms are still poorly understood, especially in triple negative breast cancer (TNBC). In this study, our results exhibited that NETs formation in TNBC tissues was higher than that in non-TNBC tissues, and NETs formation was distinctly correlated with tumor size, ki67 level and lymph node metastasis in TNBC patients. Subsequent in vivo experiments demonstrated that NETs inhibition could suppress TNBC tumor growth and lung metastasis.
