Owing to the "cold" tumor immune microenvironment of prostate cancer, immune-targeting agents have shown limited efficacy in patients with advanced prostate cancer, highlighting the need for new therapies with novel mechanisms of action. In this context, T-cell engagers (TCEs), which induce T-cell-mediated killing of cancer cells by binding the CD3 receptor on T cells and a specific tumor antigen expressed on malignant cells, represent a promising therapeutic option. Multiple studies have explored the use of TCEs in previously treated patients with metastatic castration-resistant prostate cancer, and several ongoing trials are currently assessing novel TCEs either as single agents or in combinatorial regimens with molecules with a distinct mechanism of action (eg, androgen receptor pathway inhibitors and other immune-targeting agents).

Delayed inflammatory nodules (DIN) after soft-tissue filler injection may occur after infections, vaccinations, or procedures. We report a DIN from predominantly low-molecular-weight HA (PLMW-HA) filler secondary to COVID-19 infection that was resistant to conservative management, requiring intralesional combination therapy with triamcinolone, 5-fluorouracil, and recombinant human hyaluronidase for resolution.

This updated Heart Valve Collaboratory framework addresses the growing concern for transcatheter valve failure (TVF) following transcatheter aortic valve replacement (TAVR). With the increasing volume of redo-TAV and surgical TAV explantation, there is a critical need for standardized pathways and protocols for evaluating TVF using echocardiography and cardiac computed tomography (CT) angiography. This document clarifies prior definitions of bioprosthetic valve deterioration and bioprosthetic valve failure in a practical, imaging directed context for TAVR.

Weddell seal (Leptonychotes weddellii) females lose substantial body mass across an intensive, nutritionally restricted lactation period and then must rapidly recover mass during the short Antarctic summer. In this study, we examined endocrine dynamics associated with mass loss across lactation and subsequent realimentation in Weddell seals, comparing patterns between seals that recently gave birth and demographically similar non-reproductive females (skip females) in McMurdo Sound, Antarctica. Postpartum seals near weaning (∼35 days postpartum, n = 64) and skip females (n = 32) were handled during early austral summer (November/December) and rehandled in late summer (January/February).

The polyamine spermine is a flexible linear teraamine found in bacteria and eukaryotes, and in all known cases is synthesized from triamine spermidine by addition of an aminopropyl group acquired from decarboxylated S-adenosylmethionine (dcAdoMet). We have now identified in bacteria a second biosynthetic route for spermine based on the formation of carboxyspermine from spermidine, dependent on aspartate β-semialdehyde (ASA). This route also produces thermospermine from spermidine via carboxythermospermine.

Classically, the insula is considered an associative multisensory cortex where emotional awareness emerges through the integration of interoceptive and exteroceptive information, along with autonomic regulation. However, since early intracortical microstimulation (ICMS) studies, the insular cortex has also been conceived as a mosaic of anatomo-functional sectors processing various types of sensory information to generate specific overt behaviors. Based on this, the insula has been subdivided into distinct functional fields: an anterior field associated with oroalimentary behaviors, a middle field involved dorsally in hand movements and ventrally in emotional reactions, and a posterior field engaged in axial and proximal movements.

The etiology of Alzheimer's disease (AD), a progressive neurodegenerative illness marked by memory loss and cognitive decline, may be linked to dysregulated citrate transport, according to recent research. The bi-functional enzyme FicD catalyzes AMPylation and deAMPylation of the endoplasmic reticulum chaperone BiP to modulate ER homeostasis and the unfolded protein response (UPR). Human hFicD with an arginine-to-serine mutation disrupts FicD deAMPylation activity resulting in severe neonatal diabetes. We generated the mFicD mutation in mice to create a pre-clinical murine model for neonatal diabetes.

In mammals, tissues other than liver and intestine are known to possess functional MTTP (microsomal triglyceride transfer protein) and apoB (apolipoprotein B) capable of VLDL (very low-density lipoprotein) assembly. Birds are oviparous and possess unique capabilities in lipid biology to accommodate yolk formation through massive deposition of hepatically assembled yolk-targeted VLDLy into ovarian follicles. Following identifications of MTTP and ApoB expression within chicken ovarian stroma, granulosa, theca, and epithelial cells of various classes of follicles, we sought to define the functionality of ovarian MTTP and ApoB in VLDL assembly.
