[Interaction of annexin VI with actin].

Ukr Biokhim Zh (1999)

January 2004

Taras Shevchenko Kyiv National University, Ukraine.

Actin polymerization was investigated using fluorescence probe N-(1-pyrenyl)iodoacetamide, which was bound covalently to reactive sulfhydryl group, Cys-373. Labeled actin in the bulk was 0.5 to 1% of total actin concentration.

Platelet aggregation was decreased under action of ADP and collagen in patients with myelodysplastic syndrome. The decrease in aggregation started from 3-rd minutes and decreased in 4 and 5 minutes after action of ADP. The study of Ca(2+)-ATPase and Na+, K(+)-ATPase membrane activities showed the decrease in Ca(2+)-ATPase and increase in Na+, K(+)-ATPase activity in the patients with myelodysplastic syndrome.

[Particulars on the regulation of Ca(2+) in the cell nucleus].

Ukr Biokhim Zh (1999)

December 2003

Taras Shevchenko Kyiv National University, Ukraine.

The data of literature on the structural and functional features of nuclear pore complexes, the role of receptor-activated Ca(2+)-channels and Ca(2+)-ATPase of nuclear membranes are presented. The possible ways of Ca(2+)-signal transmission into the nucleus and the role of nuclear Ca2+ in apoptosis are discussed.

A solid-phase reagent based on 1-(4-adamantyl-2-thiasolylazo)-2-naphthol adsorbed onto silica gel was prepared for Co(II) recovery and preconcentration prior to its sorption-spectroscopic detection. The immobilized reagent was applied to the determination of free cobalt ions in natural water. The solid-phase reagent and chemiluminescent method coupled with membrane filtration, gel-permeation and ion-exchange chromatography were applied to the study of the speciation of iron and cobalt in water from the Dnieper reservoirs and lakes of Kyiv City; their predominant forms were complexes of Fe(III) and Co(II) with dissolved organic matter and fulvic acids play a main role in their complexation.

It has been shown, that the total X-ray irradiation in the dozes of 0.5 and 1 Gy influences on the content of lipid peroxidation products and enzymatic activity of antioxidant system in rat spleen and thymus cells. The influence of preparations "AMMIVIT" and "Ceruloplasmin" on these processes is investigated also.

Immunosuppressive activity of culture liquid substrate (CFS) of highly virulent strain Salmonella typhimurium has been studied. A model of delayed hypersensitivity (DHS) to nonbacterial antigen in mice, a method of gel-filtration through the sephadex column G-200, immunosorbents were used. Three components with immunosuppressive activity: thermolabile component and thermostable one with direct immunosuppressive action and the third thermolabile component which manifested inductive immunosuppressive activity only after redox treatment have been revealed in the strain CFS.

Electrophoretic patterns of cell wall protein of three industrial strains, that were used for production of lysin, and eight collection strains from the genus Corynevacterium were studied to analyze their similarity as well as to estimate an opportunity of using this parameter as an additional criterion for identification and classification of corynebacteria. Similarity coefficient of cell wall overall and main protein electrophoretic patterns were determined by a specially created computer program. Electrophoretic analysis showed that every specie had an individual protein profile.

Programmed cell death (PCD) is a characteristic of all living beings in process of their development and in response to biotic and abiotic factors effects. On the pattern of the growing old 7-8th leaves of the sugar-beet (Beta vulgaris L.) of the first year vegetation of Belocerkovskaya singl-seed-45 sort character of the degradation and state of the methylation GTCGAC- and CTGCAG-sequences of the DNA were investigated.

Present day scientific data about the Ca(2+)-phospholipid-dependent protein kinases structure and molecular mechanisms of their activity are summarized and analyzed in this review. Ca(2+)-phospholipid-dependent protein kinases family is well known to include a whole series of enzymes which are homologous by their structure. They play an important role in cell differentiation, growth and proliferation as well as signal transduction through the cytoplasmic membrane.

The effect of staphylococcus active substances--protein A (PA) and peptidoglican (PG) at concentrations 10(-6)-10(-2) mg/ml on the ATPase activity of pig stomach natural actomyosin and myosin was studied. It was shown that PA and PG at direct contact with smooth muscle contractile proteins caused the activation and inhibition of ATPase activity, respectively. On the basis of this investigation it was assumed that staphylococcal active substances were able to modify of the ATPase activity smooth muscle contractile proteins perhaps via direct action on the myosin molecule, which could be accompanied by conformational changes of the active center of myosin ATPase.

The influence of C27-steroids--vitamin D3 and 20-hydroxyecdysone in free-radical reactions on lipid peroxidation in the mitochondrial fraction of rat's liver has been studied. Antioxidant activity of superoxide dismutase and catalase and the content of lipid components--cholesterol and phospholipids has been determined. The assumption was put forward that antioxidant action of steroids was possible as a result of to the modification of mitochondrial membrane through its lipid content and changing of its cholesterol: phospholipids content.

[Specific interaction of biological molecules. A new method of registration].

Ukr Biokhim Zh (1999)

January 2002

Taras Shevchenko Kyiv National University, Ukraine.

The experimental results on influence of some biological molecules and their specific immune complexes on the current-voltage characteristic of surface-barrier contact structures (Ni-Si or PorSiSi) are presented. It was shown that the deposition of the myoglobin and specific to its monoclonal antibodies or its specific immune complex caused the modification of the current-voltage characteristic. Moreover, the structures more essentially react to the formed specific immune complex deposition.

The paper describes AFM methodic approaches in the investigation of viruses (TMV, phage T4). A possibility is shown to use this method to obtain high-quality three-dimensional image of objects without additional treatment with salts of heavy metals. The phage T4 was also a model to study the object density, using the phase shift of the AFM.

Antibiotic drugs (biseptol, polymyxin, canamycin) as well as their combinations with probiotics biosporin and subalin have been studied for their effect on mice intestine microflora. It has been established that under peroral administration of the above drugs dysbacteriosis of the 2-3rd degrees arises in animals; under their combined use with probiotics one can prevent the development of dysbacteriosis to considerable extent. It has been shown that dysbacteriosis of the 2-3rd degrees can be liquidated for 5-7 days, using probiotics biosporin and subalin.
