Purpose: The enucleation rate for retinoblastoma has dropped from over 95% to under 10% in the past 10 years as a result of improvements in therapy. This reduces access to tumor tissue for molecular profiling, especially in unilateral retinoblastoma, and hinders the confirmation of somatic RB1 mutations necessary for genetic counseling. Plasma cell-free DNA (cfDNA) has provided a platform for noninvasive molecular profiling in cancer, but its applicability in low tumor burden retinoblastoma has not been shown.

EGFR-mutant NSCLCs frequently respond to EGFR tyrosine kinase inhibitors (TKIs). However, the responses are not durable, and the magnitude of tumor regression is variable, suggesting the existence of genetic modifiers of EGFR dependency. Here, we applied a genome-wide CRISPR-Cas9 screening to identify genetic determinants of EGFR TKI sensitivity and uncovered putative candidates.

Bronchial asthma (BA) is a chronic inflammatory disease with a marked heterogeneity in pathophysiology and etiology. The heterogeneity of BA may be related to the inducing mechanism(s) (allergic vs non-allergic), the histopathological background (eosinophilic vs non-eosinophilic), and the clinical manifestations, particularly in terms of severity and frequency of exacerbations. Asthma can be divided into at least two different endotypes based on the degree of Th2 inflammation (T2 'high' and T2 'low').

The B cell survival factor (TNFSF13B/BAFF) is often elevated in autoimmune diseases and is targeted in the clinic for the treatment of systemic lupus erythematosus. BAFF contains a loop region designated the flap, which is dispensable for receptor binding. Here we show that the flap of BAFF has two functions.

Zika virus (ZIKV) is a mosquito-borne flavivirus that emerged recently as a global health threat, causing a pandemic in the Americas. ZIKV infection mostly causes mild disease, but is linked to devastating congenital birth defects and Guillain-Barré syndrome in adults. The high level of cross-reactivity among flaviviruses and their cocirculation has complicated serological approaches to differentially detect ZIKV and dengue virus (DENV) infections, accentuating the urgent need for a specific and sensitive serological test.

Ideal Cardiovascular Health and Subclinical Markers of Carotid Structure and Function: The Paris Prospective Study III.

Arterioscler Thromb Vasc Biol

October 2016

From the INSERM, U970, Paris Cardiovascular Research Center, Department of Epidemiology, France (B.G., S.L., M.C.P., C.G., M.T., P.B., X.J., J.P.E.); Université Paris Descartes, Sorbonne Paris Cité, Faculté de Médecine, France (B.G., S.L., M.C.P., C.G., M.T., P.B., X.J., J.P.E.); APHP, Georges Pompidou European Hospital, Pharmacology Departments, Paris, France (S.L., P.B.); Preventive and Clinical Investigation Center, Paris, France (F.T., B.P.); APHP, Georges Pompidou European Hospital, Cardiology Department, Paris, France (X.J.); and Department of Cardiology, University Hospital of Geneva, Switzerland (H.M.).

Objective: We hypothesized that subclinical markers of vascular structure and function, which are independent predictors of cardiovascular disease, would be less frequent in subjects with ideal than poor cardiovascular health (CVH) as defined by the American Heart Association (AHA).

Approach And Results: Carotid parameters were measured using high-precision echotracking device in 9155 nonreferred participants attending a health checkup in a large health center in Paris (France) between 2008 and 2012. According to the AHA, participants with 0 to 2, 3 to 4, and 5 to 7 metrics (smoking, physical activity, body mass index, diet, blood glucose and total cholesterol, blood pressure) at the ideal level were categorized as having poor, intermediate, and ideal CVH.

Microorganisms are of uttermost importance, yet in the eyes of the general public they are often associated with dirt and diseases. At the same time, microbiologists have access to and comprehensive knowledge of just a tiny minority of the microbial diversity existing in nature. In this commentary, we present these issues of public misconception and scientific limitations and their possible consequences, and propose ways to overcome them.

Background: Breastfeeding is best for infants and the World Health Organization recommends exclusive breastfeeding for at least the first 6 months of life. For those who are unable to be breastfed, previous studies demonstrate that feeding high-risk infants with hydrolyzed formulas instead of cow's milk formula (CMF) may decrease the risk of atopic dermatitis (AD).

Objective: To estimate the economic impact of feeding high-risk, not exclusively breastfed, urban Malaysian infants with partiallyhydrolyzed whey-based formula (PHF-W) instead of CMF for the first 17 weeks of life as an AD risk reduction strategy.

Decision analysis for the genotype designation in low-template-DNA profiles.

Forensic Sci Int Genet

March 2014

University of Lausanne, Ecole des Sciences Criminelles, Institut de Police Scientifique, le Batochime, 1015 Lausanne, Switzerland.

What genotype should the scientist specify for conducting a database search to try to find the source of a low-template-DNA (lt-DNA) trace? When the scientist answers this question, he or she makes a decision. Here, we approach this decision problem from a normative point of view by defining a decision-theoretic framework for answering this question for one locus. This framework combines the probability distribution describing the uncertainty over the trace's donor's possible genotypes with a loss function describing the scientist's preferences concerning false exclusions and false inclusions that may result from the database search.

Aim Of The Study: We use the IDM model to test for over- and underuse of plant taxa as source for medicine. In contrast to the Bayes approach, which only considers the uncertainty around the data of medicinal plant surveys, the IDM model also takes the uncertainty around the inventory of the flora into account, which is used for the comparison between medicinal and local floras.

Materials And Methods: Statistical analysis of the medicinal flora of Campania (Italy) and of the medicinal flora used by the Sierra Popoluca (Mexico) was performed with the IDM model and the Bayes approach.

The functional psychoses schizophrenia, schizoaffective disorder, and bipolar illness represent complex clinical syndromes that are characterized by phenotypic heterogeneity. Yet evidence from numerous studies suggests that (1) the prevalence of schizophrenia and bipolar illness is with 1% very similar across ethnicities, and (2) a strong genetic component is involved in the disorders' pathogenesis. Using data from different US-American ethnicities (77 families with a total of 17 unaffected and 170 affected sib pairs; 276 marker loci), we searched for ethnicity-independent oligogenic susceptibility loci for which the between-sib genetic similarity in affected sib pairs deviated from the expected values.
