The Warburg effect, which describes the fermentation of glucose to lactate even in the presence of oxygen, is ubiquitous in proliferative mammalian cells, including cancer cells, but poses challenges for biopharmaceutical production as lactate accumulation inhibits cell growth and protein production. Previous efforts to eliminate lactate production in cells for bioprocessing have failed as lactate dehydrogenase is essential for cell growth. Here, we effectively eliminate lactate production in Chinese hamster ovary and in the human embryonic kidney cell line HEK293 by simultaneous knockout of lactate dehydrogenases and pyruvate dehydrogenase kinases, thereby removing a negative feedback loop that typically inhibits pyruvate conversion to acetyl-CoA.

Objectives: To study neutrophil gelatinase-associated lipocalin (NGAL) levels in peripheral blood in SLE, and to propose a mechanism by which neutrophils secrete NGAL on stimulation with immune complexes (IC).

Methods: NGAL was measured by ELISA in two independent Swedish SLE cohorts acting as exploratory and validation cohort (n=124 and n=308, respectively), disease controls (n=38) and healthy controls (n=77). NGAL levels were measured in supernatant from IC-stimulated neutrophils in the presence or absence of a toll-like receptor 8 inhibitor (TLR8i).

Vertebrate brain function is particularly sensitive to the effects of hypoxia, with even brief periods of oxygen deprivation causing significant brain damage and impaired cognitive abilities. This study is the first to investigate the cognitive consequences of hypoxia in fish, specifically induced by exhaustive exercise and air exposure, conditions commonly encountered during catch-and-release (C&R) practices in recreational fishing. Angling exerts substantial pressure on inland fish populations, underscoring the need for sustainable practices like C&R.

Reactivation of cell division is crucial for the regeneration of damaged tissues, which is a fundamental process across all multicellular organisms. However, the mechanisms underlying the activation of cell division in plants during regeneration remain poorly understood. Reservoir computing advances the intriguing idea that a nonlinear recurrent neural circuit-the reservoir-can encode spatio-temporal input signals to enable efficient ways to perform tasks like classification or regression. However, recently the idea of a monolithic reservoir network that simultaneously buffers input signals and expands them into nonlinear features has been challenged. A representation scheme in which memory buffer and expansion into higher-order polynomial features can be configured separately has been shown to significantly outperform traditional reservoir computing in prediction of multivariate time-series.

Purpose: In locations where the proton energy spectrum is broad, lineal energy spectrum-based proton biological effects models may be more accurate than dose-averaged linear energy transfer (LET) based models. However, the development of microdosimetric spectrum-based biological effects models is hampered by the extreme computational difficulty of calculating microdosimetric spectra. Given a precomputed library of lineal energy spectra for monoenergetic protons, a weighted summation can be performed which yields the lineal energy spectrum of an arbitrary polyenergetic beam.

Proteins containing the PWWP domain are typically found in chromatin, where they bind to DNA and histones in nucleosomes, facilitating chromatin-associated functions.
