Rev Med Suisse

January 2025

Service de rhumatologie, HFR Fribourg, 1708 Fribourg.

In 2024, CD19-CAR T cells are ubiquitous in rheumatology, with incredible therapeutic results in cases of severe and refractory disease. This major advance confirms the interest of the B lymphocyte as a therapeutic target, and also suggests the real possibility of a drug-free remission, at the price of minor and minimal side effects for the time being. However, the necessary perspective is still lacking for a therapy that remains out of reach because of its price.

Joint hypermobility is a common feature in children and adolescents, often physiological and benign, but it is also a manifestation of hereditary connective tissue disorders. In pediatrics, there is a real need to differentiate Ehlers-Danlos syndrome (EDS) hypermobile type (hEDS) from benign hypermobility, in the absence of a genetic marker specific to this form of EDS. In 2023, the International Ehlers-Danlos Consortium revised the diagnostic criteria for children and adolescents, introducing the concept of hypermobility spectrum disorder.

Understanding a small molecule's mode of action (MoA) is essential to guide the selection, optimization and clinical development of lead compounds. In this study, we used high-throughput non-targeted metabolomics to profile changes in 2,269 putative metabolites induced by 1,520 drugs in A549 lung cancer cells. Although only 26% of the drugs inhibited cell growth, 86% caused intracellular metabolic changes, which were largely conserved in two additional cancer cell lines.

Hypothesis: Monodisperse phospholipid-coated microbubbles, with a size and resonance frequency tuned to the ultrasound driving frequency, have strong potential to enhance sensitivity, efficiency, and control in emerging diagnostic and therapeutic applications involving bubbles and ultrasound. A key requirement is that they retain their gas volume and shell material during physiologic pressure changes and withstand the overpressure during intravenous injection. The shell typically comprises a mixture of a phospholipid (e.
