Sixteen patient-derived xenografts (PDXs) were analyzed using a mass spectrometry (MS)-based kinase inhibitor pull-down assay (KIPA), leading to the observation that death-associated protein kinase 3 (DAPK3) is significantly and specifically overexpressed in the triple-negative breast cancer (TNBC) models. Validation studies confirmed enrichment of DAPK3 protein, in both TNBC cell lines and tumors, independent of mRNA levels. Genomic knockout of in TNBC cell lines inhibited in vitro migration and invasion, along with down-regulation of an epithelial-mesenchymal transition (EMT) signature, which was confirmed in vivo.

The authors' aim is to build "more biologically plausible learning algorithms" that work in naturalistic environments. Given that, first, human learning and memory are inextricable, and, second, that much human learning is unconscious, can the authors' first research question of how people improve their learning abilities over time be answered without addressing these two issues? I argue that it cannot.

In recent years, proposals have been advanced to redefine or reclassify Lewy body disorders by merging the long-established entities of Parkinson's disease (PD), Parkinson's disease dementia (PDD) and dementia with Lewy bodies (DLB). These proposals reject the International DLB Consortium classification system that has evolved over three decades of consensus collaborations between neurologists, neuropsychologists and neuropathologists. While the Consortium's "one year rule" for separating PD and DLB has been criticized as arbitrary, it has been a pragmatic and effective tool for splitting the continuum between the two entities.

Bcl-xL is translocated to the nucleus via CtBP2 to epigenetically promote metastasis.

Cancer Lett

November 2024

Department of Pathology and Laboratory Medicine, Weill Cornell Medicine, New York, NY, 10065, USA. Electronic address:

Article Synopsis
  • Nuclear Bcl-xL promotes cancer metastasis independently of its role in preventing cell death, but how it enters the nucleus and alters gene regulation remains unclear.
  • The study found that C-terminal Binding Protein 2 (CtBP2) is crucial for transporting Bcl-xL into the nucleus, and silencing CtBP2 reduces Bcl-xL levels and reverses its effects on cancer spread in mouse models.
  • Additionally, Bcl-xL's interactions with CtBP2 and MLL1 enhance histone modifications linked to promoting cancer-related genes, suggesting new treatment approaches for cancers with high Bcl-xL levels.
