Background: Opium use has been associated with an increased risk of cancers of the lung, oesophagus, and pancreas, and it was recently classified by the International Agency for Cancer Research as carcinogenic to humans. It is not clear whether opium also increases the risk of colorectal cancer (CRC). The aim of our study was to assess the association between various metrics of opium use and the risk of CRC.

Epigenetic regulation of breast cancer metastasis.

Cancer Metastasis Rev

June 2024

Stony Brook Cancer Center, Renaissance School of Medicine, Stony Brook University, Lauterbur Drive, Stony Brook, NY, 11794, USA.

Breast cancer is the most frequently diagnosed malignancy and the second leading cause of cancer-related mortality among women worldwide. Recurrent metastasis is associated with poor patient outcomes and poses a significant challenge in breast cancer therapies. Cancer cells adapting to a new tissue microenvironment is the key event in distant metastasis development, where the disseminating tumor cells are likely to acquire genetic and epigenetic alterations during the process of metastatic colonization.

Article Synopsis
  • * The Golestan Cohort Study began in 2004 and tracked cases of esophageal squamous cell carcinoma, matching them with controls based on several factors, and analyzed urinary biomarkers related to cancer exposure.
  • * Significant associations were found between certain metabolites and esophageal cancer, with higher odds ratios especially noted among tobacco users, suggesting both tobacco and some non-tobacco chemicals contribute to cancer risk.

Cancers attributable to overweight and obesity in Italy.

Cancer Epidemiol

December 2023

Department of Clinical Sciences and Community Health, University of Milan, Milan, Italy. Electronic address:

Background & Aims: Overweight and obesity are associated with multiple cancers. We quantified the burden of cancer attributable to overweight and obesity in Italy.

Methods: We estimated sex- and cancer site-specific population attributable fractions (PAFs) combining relative risks (from recent meta-analyses) with national obesity prevalence data (from a large sample survey conducted in 2005, to account for a 15-year lag period).

Incident cancers attributable to using opium and smoking cigarettes in the Golestan cohort study.

EClinicalMedicine

October 2023

Genomic Epidemiology Branch, International Agency for Research on Cancer (IARC - WHO), Lyon, France.

Background: Opium consumption has recently been identified as a carcinogen, but the impact of opium use on cancer burden is unknown. We aimed to evaluate the fraction of cancers that could be attributed to opium use alone and in combination with cigarette smoking in a region where opium is widely used.

Methods: 50,045 Iranian adults were recruited to this prospective cohort study between 2004 and 2008 and were followed through January 2022.

Introduction: It is unclear whether differences in health outcomes by racial and ethnic groups among World Trade Center (WTC) rescue and recovery workers reflect those of the population of New York State (NYS) or show distinct patterns. We assessed cancer incidence in WTC workers by self-reported race and ethnicity, and compared it to population figures for NYS.

Methods: A total of 61,031 WTC workers enrolled between September 11, 2001 and January 10, 2012 were followed to December 31, 2015.

Background: Poor oral health has been identified as a prognostic factor potentially affecting the survival of patients with head and neck squamous cell carcinoma. However, evidence to date supporting this association has emanated from studies based on single cohorts with small-to-modest sample sizes.

Methods: Pooled analysis of 2449 head and neck squamous cell carcinoma participants from 4 studies of the International Head and Neck Cancer Epidemiology Consortium included data on periodontal disease, tooth brushing frequency, mouthwash use, numbers of natural teeth, and dental visits over the 10 years prior to diagnosis.

The heat shock protein Hsp27 controls mitochondrial function by modulating ceramide generation.

Cell Rep

September 2023

Department of Biochemistry and Molecular Biology, Virginia Commonwealth University School of Medicine, Richmond, VA 23398, USA; Massey Cancer Center, Virginia Commonwealth University, Richmond, VA 23398, USA. Electronic address:

Article Synopsis
  • Sphingolipids are essential for membrane structure and cellular signaling, with ceramide being a key molecule in this process generated by specific enzymes called ceramide synthases (CerS).
  • The small heat shock protein 27 (Hsp27) specifically interacts with and inhibits CerS1, affecting ceramide production; this interaction is crucial for understanding CerS regulation.
  • Hsp27's activity influences mitochondrial function and mitophagy, highlighting its role in the cellular response to stress and its significance in managing CerS1-related processes.

Article Synopsis
  • - The study investigated the links between sleep duration, stress levels, and gastric cancer (GC) by analyzing data from five case-control studies, which included 1,293 GC cases and 4,439 controls.
  • - Findings revealed that sleeping 9 hours or more is associated with a higher risk of GC (OR = 1.57) and that stress levels also increase the risk of developing GC (OR = 1.33), particularly for noncardia GC.
  • - The correlations between stress and GC risk varied based on factors like smoking and socioeconomic status, indicating that long sleep duration and heightened stress are important risk factors for different types of gastric cancer.

Cancer mortality associated with low education in Italy.

J Public Health (Oxf)

November 2023

Department of Pathophysiology and Transplantation, Università degli Studi di Milano, 20122 Milano, Italy.

Article Synopsis
  • * It found that 29.1% of cancer deaths in men and 13.3% in women in 2019 were attributable to low education levels, translating to 22,271 deaths in men and 7,456 in women.
  • * The research suggests that reducing educational disparities could potentially prevent about a quarter of cancer deaths in Italy by addressing socioeconomic factors.

ORCHESTRA ("Connecting European Cohorts to Increase Common and Effective Response To SARS-CoV-2 Pandemic") is an EU-funded project which aims to help rapidly advance the knowledge related to the prevention of the SARS-CoV-2 infection and the management of COVID-19 and its long-term sequelae. Here, we describe the early results of this project, focusing on the strengths of multiple, international, historical and prospective cohort studies and highlighting those results which are of potential relevance for vaccination strategies, such as the necessity of a vaccine booster dose after a primary vaccination course in hematologic cancer patients and in solid organ transplant recipients to elicit a higher antibody titer, and the protective effect of vaccination on severe COVID-19 clinical manifestation and on the emergence of post-COVID-19 conditions. Valuable data regarding epidemiological variations, risk factors of SARS-CoV-2 infection and its sequelae, and vaccination efficacy in different subpopulations can support further defining public health vaccination policies.

ΔNp63 Regulates Homeostasis, Stemness, and Suppression of Inflammation in the Adult Epidermis.

J Invest Dermatol

January 2024

Department of Pathology, Renaissance School of Medicine, Stony Brook University, Stony Brook, New York, USA; Stony Brook Cancer Center, Stony Brook, New York, USA. Electronic address:

The p63 transcription factor is critical for epidermis formation in embryonic development, but its role in the adult epidermis is poorly understood. In this study, we show that acute genetic ablation of ΔNp63, the main p63 isoform, in adult epidermis disrupts keratinocyte proliferation and self-maintenance and, unexpectedly, triggers an inflammatory psoriasis-like condition. Mechanistically, single-cell RNA sequencing revealed the downregulation of cell cycle genes, upregulation of differentiation markers, and induction of several proinflammatory pathways in ΔNp63-ablated keratinocytes.
