Cystic fibrosis (CF) is characterized by abnormal mucus hydration due to a defective CF Transmembrane Regulator (CFTR) protein, leading to the production of difficult-to-clear mucus. This causes airflow obstruction, recurrent infections, and respiratory complications. Chronic lung infections are the leading cause of death for CF patients and inhaled tobramycin is the first-in-line antibiotic treatment against these infections, mainly caused by Pseudomonas aeruginosa in adult patients.

Compounding medications in pharmacies is a common practice for patients with prescriptions that are not available commercially, but it is a laborious and error-prone task. The incorporation of emerging technologies to prepare personalised medication, such as 3D printing, has been delayed in smaller pharmacies due to concerns about potential workflow disruptions and learning curves associated with novel technologies. This study examines the use in a community pharmacy of a pharmaceutical 3D printer to auto-fill capsules and blisters using semisolid extrusion, incorporating an integrated quality control system.

The original study was based on Australian and Belgian population, but significant variation in CPAK distribution exists between different geographic areas. The primary objective is to evaluate knee phenotypes of osteoarthritic Spanish population based on the CPAK system.

Atrial fibrillation (AF) and atrial myopathy are recognized contributors to cardiovascular morbidity, particularly ischemic stroke. AF poses an elevated risk of thrombogenesis due to irregular heart rhythm leading to blood stasis and clot formation. Atrial myopathy, marked by structural and functional alterations in the atria, is emerging as a crucial factor influencing thromboembolic events, independently of AF.

The age-specific incidence of traumatic brain injury in older adults is rising in high-income countries, mainly due to an increase in the incidence of falls. The severity of traumatic brain injury in older adults can be underestimated because of a delay in the development of mass effect and symptoms of intracranial haemorrhage. Management and rehabilitation in older adults must consider comorbidities and frailty, the treatment of pre-existing disorders, the reduced potential for recovery, the likelihood of cognitive decline, and the avoidance of future falls.

Safety and efficacy of nipocalimab in adults with generalised myasthenia gravis (Vivacity-MG3): a phase 3, randomised, double-blind, placebo-controlled study.

Background: Given burdensome side-effects and long latency for efficacy with conventional agents, there is a continued need for generalised myasthenia gravis treatments that are safe and provide consistently sustained, long-term disease control. Nipocalimab, a neonatal Fc receptor blocker, was associated with dose-dependent reductions in total IgG and anti-acetylcholine receptor (AChR) antibodies and clinically meaningful improvements in the Myasthenia Gravis Activities of Daily Living (MG-ADL) scale in patients with generalised myasthenia gravis in a phase 2 study. A forward genetic screen identifies potassium channel essentiality in SHH medulloblastoma maintenance.

Distinguishing tumor maintenance genes from initiation, progression, and passenger genes is critical for developing effective therapies. We employed a functional genomic approach using the Lazy Piggy transposon to identify tumor maintenance genes in vivo and applied this to sonic hedgehog (SHH) medulloblastoma (MB). Combining Lazy Piggy screening in mice and transcriptomic profiling of human MB, we identified the voltage-gated potassium channel KCNB2 as a candidate maintenance driver.

Green solutions for treating groundwater polluted with nitrates, pesticides, antibiotics, and antibiotic resistance genes for drinking water production.

The present study evaluates for the first time the seasonal performance of an innovative green groundwater treatment. The pilot plant combines microalgae-bacteria treatment and a cork-wood biofilter to reduce nitrates, pesticides, antibiotics (ABs), and antibiotic resistance genes (ARGs) from groundwater. Groundwater had nitrate concentrations ranging from 220 to 410 mg/L, while ABs (sulfonamides and fluoroquinolones) and pesticides (triazines) were detected at concentrations ranging from a few ng/L to 150 ng/L.

The new Urban Wastewater Treatment Directive (UWWTD) in European Union requires implementation of quaternary wastewater treatment processes to limit the loads of pharmaceuticals reaching water bodies.

The presence of cells in urine and in particular White Blood Cells (WBCs) is often associated with Urinary Tract Infections (UTIs) and other diseases. Non-invasive screening of WBCs requires the development of cost-effective point of care diagnostic tools. Infrared (IR) spectroscopy has the potential to identify and quantify cells in urine.

Background: Isolated subsegmental pulmonary embolism (issPE) is a commonly encountered diagnosis. Although the International Classification of Diseases (ICD)-10 codes are used for research, their validity for identifying issPE is unknown. Moreover, issPE diagnosis is challenging, and the findings from radiology reports may conflict with those from expert radiologists.

Revisiting secondary model features for describing the shoulder and lag parameters of microbial inactivation and growth models.

The Baranyi and Geeraerd models are two of the most reliable models for the description of, respectively, microbial growth and inactivation. They are defined as a system of differential equations, whose algebraic solution can describe the microbial response during isothermal conditions, especially when combined with suitable secondary models. However, there are still large uncertainties regarding the best functions to use as secondary models for the lag phase duration (λ) and the shoulder length (S).

Lung function-associated exposome profile in the era of climate change: Pooled analysis of 8 population-based European cohorts within the EXPANSE project.

Background: The independent and interrelated long-term effects of the exposome such as air pollution, greenness, and ambient temperature on lung function are not well understood, yet relevant in the light of climate change.

Methods: Pre-bronchodilation FEV1 from five mature birth cohorts (N = 4724) and three adult cohorts (N = 6052) from five European countries were used to assess cross-sectional associations with air pollution, greenness, and ambient temperature, assigned to their residential address. All two-way interactions and square terms were a priori included in building the final elastic net regression model.

Background: Dravet syndrome (DS) is a severe developmental and epileptic encephalopathy associated with loss-of-function variants in the SCN1A gene. Although predominantly expressed in the central nervous system, SCN1A is also expressed in the heart, suggesting a potential link between neuronal and cardiac channelopathies. Additionally, DS carries a high risk of sudden unexpected death in epilepsy (SUDEP).

Objectives: The aim of this study was to explore the association of fruit, vegetable, and pulses consumption with all-cause, cardiovascular, and cancer mortality.

Study Design: This prospective study included 66,933 individuals from three Spanish health surveys linked to the national death registry up to December 2022.

Methods: Adjusted Poisson regression models were used to analyze the data, categorizing fruit, vegetable and pulses intake according to Spanish dietary recommendations and using splines to examine non-linear relationships.

Background: Adult disclosure of child sexual abuse (CSA) is often addressed to partners and friends. The risk of negative reactions is high due to the stigma associated with CSA, however receiving social support is a protective factor. Therefore, educating the environment on how to face disclosure becomes crucial.

Research on nanoparticle (NP) release and potential exposure can be assessed through experimental field campaigns, laboratory simulations, and prediction models. However, risk assessment models are typically designed for manufactured NP (MNP) and have not been adapted for incidental NP (INP) properties. A notable research gap is identifying NP sources and their chemical, physical, and toxicological properties, especially in real-world settings.

Bacterial and fungal diversity and species interactions inversely affect ecosystem functions under drought in a semi-arid grassland.

Extreme climatic events, such as drought, can significantly alter belowground microbial diversity and species interactions, leading to unknown consequences for ecosystem functioning. Here, we simulated a drought gradient by removing 30 %, 50 %, and 70 % of precipitation in a semi-arid grassland over five years. We assessed the effects of drought on bacterial and fungal diversity, as well as on their species interactions.

Chloramphenicol glycoside derivative: A way to overcome its antimicrobial resistance and toxicity.

Triggered by the urgent need to tackle the global crisis of multidrug-resistant bacterial infections, in this work, we present a way to overcome chloramphenicol resistance by introducing modifications based on the glycosylation of its hydroxyl groups. The synthesized derivatives demonstrate complete resistance to the action of recombinant chloramphenicol acetyltransferase (CAT) from Escherichia coli and efficacy against methicillin-resistant Staphylococcus aureus (MRSA), Escherichia coli ESBL, and Pseudomonas aeruginosa ATCC 27853. Glycosylation gives chloramphenicol an additional advantage - the stable glycosidic form is less toxic to human dermal fibroblasts and has significantly better water solubility than non-glycosylated chloramphenicol.

Parenthood, gender, and turning points to crime for young people in Latin America.

In very different societal contexts, parenthood has been identified as a critical turning point in life course trajectories. In this qualitative study, we explore parenthood as a turning point for 40 young women and 40 young men in prisons across Latin America. We study the impact of parenthood on criminal trajectories, identify gender differences, and analyze the different mechanisms at work.

Direct analysis of engineered iron nanotubes and platinum nanorods: A challenge for single particle inductively coupled plasma mass spectrometry.

The use of inductively coupled plasma mass spectrometry in single particle mode (SP-ICP-MS) for the characterization of micro and nanostructured materials is a growing field of research. In this work, the possibility of expanding the boundaries to anisotropic structures including solid Pt-nanorods and hollowed FeO-nanotubes is presented. The obtained structures are evaluated by scanning electron microscopy (SEM), high-resolution electron microscopy (HR-TEM) and SP-ICP-MS techniques.
