Heterozygotic mutations are responsible for various congenital diseases in the heart, pancreas, liver, and other organs in humans. However, there is lack of an animal that can comprehensively model these diseases since GATA6 is essential for early embryogenesis. Here, we report the establishment of a knockout zebrafish which recapitulates most of the symptoms in patients with mutations, including cardiac outflow tract defects, pancreatic hypoplasia/agenesis, gallbladder agenesis, and various liver diseases.

Despite being a groundbreaking approach to treating colorectal cancer (CRC), the efficacy of immunotherapy is significantly compromised by the immunosuppressive tumor microenvironment and dysbiotic intestinal microbiota. Here, leveraging the superior carrying capacity and innate immunity-stimulating property of living bacteria, a nanomedicine-engineered bacterium, LR-S-CD/CpG@LNP, with optical responsiveness, immune-stimulating activity, and the ability to regulate microbiota metabolome is developed. Immunoadjuvant (CpG) and carbon dot (CD) co-loaded plant lipid nanoparticles (CD/CpG@LNPs) are constructed and conjugated to the surface of Limosilactobacillus reuteri (LR) via reactive oxygen species (ROS)-responsive linkers.

The Q-matrix is one of the core components of cognitive diagnostic assessment, which is a matrix describing the relationship between items and the attributes being assessed. Numerous studies have shown that inaccuracies in defining the Q-matrix can degrade parameter estimation and model fitting results. Currently, Q-matrix validation often involves exhaustive search algorithms (ESA), which traverse through all possible -vectors and determine the optimal -vector for items based on indicators or criteria corresponding to different validation methods.

A pot experiment was conducted on arsenic (As) and cadmium (Cd) co-contaminated soil to discern the influence of varying proportions of pig manure compost (PM) vis-à-vis chemical fertilizers (NPK) on the mitigation of Cd and As absorption by rice. Our findings illustrated that by increasing the PM proportions from 25 to 100%, it manifested a statistically significant reduction in the mobilized fractions of Cd, accounting for up to 77% reduction in soil CaCl-Cd concentrations. Conversely, the NaHCO-As reactions were contingent on the distinct PM application rates.

When the traditional random forest (RF) algorithm is used to select feature elements in biostatistical data, a large amount of noise data and parameters can affect the importance of the selected feature elements, making the control of feature selection difficult. Therefore, it is a challenge for the traditional RF algorithm to preserve the accuracy of algorithm results in the presence of noise data. Generally, directly removing noise data can result in significant bias in the results.

Bio-microfluidic technologies offer promising applications in diagnostics and therapy, yet they face significant technical challenges, particularly in the need for external power sources, which limits their practicality and user-friendliness. Recent advancements have explored innovative methods utilizing body fluids, motion, and heat to power these devices, addressing the power supply issue effectively. Among these, body-motion and body-heat-powered systems stand out for their potential to create self-sustaining, wearable, and implantable devices.

The biological carbon pump (BCP) associated with aquatic photosynthesis in karst surface waters converts dissolved inorganic carbon (DIC) into organic carbon. In the context of global climate change, BCP could be an important carbon sink mechanism, ultimately regulating atmospheric carbon dioxide (CO) and mitigating climate change. Because of the high DIC and pH, and low dissolved CO [CO (aq)], the hydrochemical characteristics of karst surface water bodies cause C limitation in BCP efficiency.

The concentration variation of luteinizing hormone (LH) regulates the cell cycle of oocyte meiosis and significantly affect the whole reproductive cycle. Sensitively quantifying the LH biomarker therefore plays an important role for reproductive disease diagnosis. By coupling a new low background catalytic redox recycling strategy with hybridization chain reaction (HCR), we propose a highly sensitive bio-electrochemical aptamer LH sensing method.

The traditional gelatin extraction methods (acid-base) may hinder to their green applications due to mass energy consumption and pollution. Herein, we constructed a clean and sustainable gelatin extraction method, investigated the molecular mechanism of microwave treatment (0-360 min) and freeze-thaw on the gelatin extraction from the perspective of the crosslinking degree and hydrogen bonds. Microwave (0-60 min) can improve the hydrolysis degree (DH) and expose more enzyme cleavage sites of collagen by destroying the intramolecular and intermolecular covalent crosslinking.

An accurate understanding of the spatial distribution of soil heavy metals (HMs) is crucial for the effective prevention of soil pollution and remediation strategies. Traditional machine learning models often overlook the spatially stratified heterogeneity inherent to environmental data, which can impair predictive accuracy. Therefore, we combined the Geodetector model (GDM) with machine learning models.

Traditional wound closure methods often present several issues, including additional puncture wounds, adverse effects from anesthesia, and noticeable scarring. Inspired by embryonic wound healing, a Janus hydrogel (PG/Au-Asp@PCM) is designed to manipulate non-invasive wound closure by photothermal-responsive self-contraction of PG/Au-Asp@PCM, which is attributed to the shape memory behavior of PG/Au-Asp@PCM under near-infrared (NIR). Wherein, gelatin acts as a thermally reversible "switch" and polyacrylamide creates stable and cross-linked "net-points".

Rab GTPases are a class of small GTP-binding proteins, play crucial roles in the membrane transport machinery with in eukaryotic cells. They dynamically regulate the precise targeting and tethering of transport vesicles to specific compartments by transitioning between active and inactive states. In plants, Rab GTPases are classified into eight distinct subfamilies: Rab1/D, Rab2/B, Rab5/F, Rab6/H, Rab7/G, Rab8/E, Rab11/A, and Rab18/C.

In criminal investigations, distinguishing between impact spatters and fly spots presents a challenge due to their morphological similarities. Traditional methods of bloodstain pattern analysis (BPA) rely significantly on the expertise of professional examiners, which can result in limitations including low identification efficiency, high misjudgment rates, and susceptibility to external disturbances. To enhance the accuracy and scientific rigor of identifying impact spatters and fly spots, this study employed artificial intelligence techniques in image recognition and transfer learning.
