Article Synopsis
  • Inebilizumab is an anti-CD19 antibody effective for treating neuromyelitis optica spectrum disorder (NMOSD) in adults with aquaporin-4 autoantibodies, showing significant long-term benefits in reducing disease activity.
  • In the N-MOmentum study, treatment led to quick and sustained depletion of B-cells and plasma-cells, with deeper depletion linked to better clinical outcomes, including lower attack rates and fewer new MRI lesions.
  • The study concludes that while inebilizumab rapidly depletes B-cells, the extent of this depletion correlates with improved stability and reduced severity of NMOSD symptoms over time.

Fabry disease (FD) is an X-linked inherited lysosomal metabolism disorder in which globotriaosylceramide (Gb3) accumulates in various organs resulting from a deficiency in alpha-galactosidase A. The clinical features of FD include progressive impairments of the renal, cardiac, and peripheral nervous systems. In addition, patients with FD often develop neuropsychiatric symptoms, such as depression and dementia, which are believed to be induced by the cellular injury of cerebrovascular and partially neuronal cells due to Gb3 accumulation.

Objective: Psychosis is an important comorbidity in epilepsy, but its pathophysiology is still unknown. The imaging modality F-fluorodeoxyglucose-positron emission tomography ( F-FDG PET) is widely used to measure brain glucose metabolism, and we speculated that F-FDG PET may detect characteristic alteration patterns in individuals with temporal lobe epilepsy (TLE) and psychosis.

Methods: We enrolled 13 patients with TLE and interictal psychosis (TLE-P) and 21 patients with TLE without psychosis (TLE-N).

This study surveyed Japanese anesthesiologists' knowledge of the cost of medicine and their attitudes toward cost containment to determine how these factors may affect their choice of medication and provide insight into reducing healthcare expenditures. In this cross-sectional study, Japanese anesthesiologists' knowledge of medicine prices and their attitudes toward cost containment were surveyed to identify barriers in lowering the cost of anesthesia. The proportion of participants who correctly guessed the cost of the five most frequently used types of drugs within 25% of the actual price was determined, and their attitudes regarding drug prices and barriers to achieving cost containment were analyzed.

The misuse of stimulant drugs such as methamphetamine is a global public health issue. One important neurochemical mechanism of methamphetamine use disorder may be altered dopaminergic neurotransmission. For instance, previous studies using positron emission tomography (PET) have consistently shown that striatal dopamine D2-type receptor availability (quantified as binding potential; BP) is lower in methamphetamine use disorder.

Objective: Although previous studies have investigated age and gender effects on striatal subregional dopamine transporter (DaT) binding, these studies were mostly based on a conventional regions of interest-based analysis. Here, we investigated age and gender effects on striatal DaT binding at the voxel level, using a multicenter database of [()I] N-omega-fluoropropyl-2beta-carbomethoxy-3beta-{4-iodophenyl}nortropane ([()I] FP-CIT)-single photon emission computed tomography (SPECT) scans in 256 healthy Japanese adults.

Methods: We used the Southampton method to calculate the specific binding ratios (SBRs) of each subject's striatum and then converted the [I] FP-CIT SPECT images to quantitative SBRs images.

Software development for quantitative analysis of brain amyloid PET.

Brain Behav

March 2022

Drug Discovery and Cyclotron Research Center, Southern Tohoku Research Institute for Neuroscience, Koriyama, Fukushima, Japan.

Introduction: Centiloid (CL) scaling has become a standard quantitative measure in amyloid PET because it allows the direct comparison of results across sites, even when different analytical methods or PET tracers are used.

Methods: In the present study, we developed new standalone software to easily handle a pipeline for accurate calculation of the CL scale for the five currently available amyloid PET tracers- C-PiB, F-florbetapir, F-flutemetamol, F-florbetaben, and F-NAV4694. This pipeline requires reorientation and coregistration of PET and MRI, anatomic standardization of coregistered PET to a standardized space using a warping parameter for coregistered MRI, application of standard volumes of interest (VOIs) to the warped PET, calculation of the standardized uptake value ratio (SUVR) for the target VOIs, and finally conversion of the SUVR to the CL scale.
