Objective: To investigate the molecular alterations related to the carcinogenesis of esophageal squamous cell carcinoma (SCC), and also to find some molecular markers for the early detection of this cancer.

Methods: The resected tumor specimens and dysplasia tissues from 34 patients with esophageal cancer as well as their matched blood DNAs were analyzed for loss of heterozygosity (LOH) at 16 microsatellites by using PCR and fluorescence-based DNA sequencing technology. Gene chip methods are applied to the study of gene expression. The differentially expressed genes in different specimens may be detected with parallel analysis by gene chip, which has greatly improved the traditional experiments in that only a single or several gene expressions need to be observed for each test, thereby speeding up the identification of differentially expressed genes and the construction of differential expression profiles. Many studies have applied this technology for Barrett's metaplasia and adenocarcinoma, and identified a number of candidate genes useful as biomarkers in cancer staging, prediction of recurrence, prognosis and treatment selection.

The electronic nursing documentation is not unified in Finland and it doesn't connect with nationally recommended multiprofessional core documentation of the patient history. The aims are to develop a nationally unified and standardized nursing documentation by the year 2007, to use the standardized nursing data to manage and assess the quality of the nursing process and to integrate the nursing documentation into the multiprofessional patient record. The nationally defined structured nursing documentation is piloted in 23 health care organizations during 2005-2007 by the support of the Ministry of Social Affairs and Health.

Methods: Eighty circular full-thickness wounds were produced on both sides of the back in 20 male Wistar rats labeled with BrdU 60 days previously (4 wounds in each rat). BrdU, beta1 integrin and keratin 19 (K19) were employed to determine the epidermal stem cells with SP immunohistochemical methods, and the epithelialization was determined with routine histological methods of HE staining on the 3rd, 7th, 14th, and 21st days after operation.

Retinitis pigmentosa is a very heterogeneous group of retinal degenerations, with multiple genes identified in each mode of inheritance. For autosomal dominant retinitis pigmentosa (ADRP), the most common gene is the rhodopsin (RHO) gene, mutations in which contribute to about 25% of ADRP in Caucasian population. To investigate the frequency and pattern of RHO point mutations in Chinese patients with ADRP, we have screened the five coding exons and splice sites of the RHO gene in 50 unrelated probands from Chinese ADRP families and 100 normal controls to identify disease-associated mutations, using conformation sensitive gel electrophoresis (CSGE) and direct DNA sequencing.

Background: Retinoblastoma, an embryonic neoplasm of retinal origin, is the most common and severe intra-ocular tumor affecting infants and young children.

Methods: Loss of heterozygosity (LOH) on chromosome 13 was investigated in 16 Chinese sporadic RB patients, using 14 microsatellite markers spanning the complete chromosome 13, to determine whether those alterations different from the alteration of RB1 gene on 13q14 may play a role in the development of RB. Loss of RB1 allele is commonly encountered in sporadic RB.

The aim is to investigate biological features of porcine endogenous retrovirus (PERV) of pigs in China and to provide basic parameters for evaluation of biological safety of xenotransplantation from pig to human. In this study, basic biologic features of PERV of DNA of peripheral blood white cells of 12 species of domestic pigs in China were examined by polymerase chain reaction; Analysis of difference of PERV gene sequence was investigated by sliver-stained single stranded conformational polymorphism and restriction fragment length polymorphism; Difference of PERV mRNA from a variety of tissue of two species of mini-pig was analysized by reverse transcriptase-PCR and semi-quantitative analysis. These results showed that Sequences of PERV-A, -B genes are ubiquitous in DNA genome of peripheral blood white cells in the examined pigs; There was no single stranded conformational polymorphism in PERV-A, -B gene sequences.

To explore the application value of bcr-abl fusion gene deterction microarray in diagnosis, typing, choosing of treatment variant and prognosis judgment, probe for fusion gene detection was designed, oligonucleotide microarray was prepared; total RNA was extracted, reverse-transcripted and labeled by fluorescence, then cDNA was hybridized with microarray in order to detect bcr-abl fusion gene in leukemia cells. The results showed that better reaction conditions were gained by exploration of hybridizotion temperature and elution conditions, bcr-abl fusion gene in leukemia cells was detected by prepared miccroarray. In conclusion, oligonucleotide microarray is effective in detecting the fusion gene and has some unique advantages and certain clinical application value, but has some deficiency too.
