The Role of MicroRNAs and Stem cells in Rheumatoid Arthritis: A Therapeutic and Diagnostic Approach.

Curr Rheumatol Rev

September 2024

Departments of Histology and Cell Biology (Genetics Unit), Faculty of Medicine, Suez Canal University, 41522, Ismailia, Egypt.

In autoimmune illnesses like Rheumatoid Arthritis (RA), the synovium is constantly inflamed, and joints are ruptured. It is becoming increasingly clear that stem cells, especially notably mesenchymal stem cells (MSCs) and microRNAs (miRNAs), play important roles in the onset and amelioration of RA. There is mounting evidence from both animal and human studies that suggests a potentially safe and effective method to treat RA and other intractable diseases by reducing chronic inflammation and spurring tissue regeneration through the transplantation of multipotent adult stem cells, such as mesenchymal stromal/stem cells.

  • Tramadol, a widely used painkiller, may negatively affect male reproductive health, as shown in an observational study of 30 tramadol addicts compared to 30 healthy individuals.
  • The study found that tramadol abuse led to decreased sperm quality, lower testosterone levels, and higher prolactin levels, along with increased oxidative stress markers in semen.
  • Additionally, correlations were found linking higher tramadol levels in seminal plasma with poorer sperm motility and vitality, as well as lower zinc and selenium levels, suggesting that tramadol may impair male fertility through oxidative damage and hormonal changes.

Article Synopsis
  • - A new series of thiazole-based chalcones was tested for anticancer properties, showing effective inhibition of cancer cell growth, particularly in Ovar-3 and MDA-MB-468 cells, at concentrations between 1.55 and 2.95 μM.
  • - One compound notably inhibited tubulin polymerization with an IC value of 7.78 μM, which is less effective than the reference drug Combretastatin-A4 (CA-4) at 4.93 μM.
  • - Molecular docking studies indicated that these compounds effectively bind to the same site on tubulin as CA-4, along with predictions suggesting they have good oral bioavailability and potential as chemotherapy drugs.

  • Sparsentan has shown positive results in treating IgA nephropathy and focal glomerulosclerosis (FSGS), but a pooled analysis of recent trials is needed to confirm its efficacy and safety compared to irbesartan.
  • A systematic review of three studies involving 884 patients found that sparsentan significantly improves proteinuria levels and remission rates, but both medications have similar effects on kidney function.
  • Although sparsentan is generally effective and safe, it does have a higher incidence of hypotension, and further research with larger sample sizes is necessary for more definitive conclusions.

Article Synopsis
  • - The World Health Organization reported that about 970 million people globally had a mental disorder in 2019, highlighting the significant burden of mental health issues on individuals and healthcare systems.
  • - A survey of 1,394 people in Iraq found that 54.73% had positive attitudes towards mental health, but men and older individuals (46 years and up) were more likely to have negative views.
  • - The study indicates that stigma around mental health problems in the Middle East affects treatment and recovery, and emphasizes the need for health education initiatives to challenge these misconceptions, while suggesting further research for better understanding in Iraq.

This research addresses the challenge of analyzing pregabalin, a primary amine in a zwitterionic structure, which is difficult to evaluate due to its lack of chromatophore. The study introduces a derivatization assessment using Hantzsch's multicomponent organic reaction to enhance the detectability of pregabalin by forming a highly fluorescent dihydropyridine derivative. This process involves the condensation of pregabalin with acetylacetone and formaldehyde, yielding a yellowish-green compound measurable both colorimetrically at 338 nm and fluorimetrically at an emission wavelength of 486 nm (λ = 408 nm).

Heavy metal contamination threatens the aquatic environment and human health. Different physical and chemical procedures have been adopted in many regions; however, their adoption is usually limited since they take longer time, are more expensive, and are ineffective in polluted areas with high heavy metal contents. Thus, biological remediation is considered a suitable applicable method for treating contaminates due to its aquatic-friendly features.

M-polarized, tumor-associated macrophages (TAMs) produce pro-tumorigenic and angiogenic mediators, such as interleukin-8 (IL-8) and IL-10. Leucine-rich repeat-containing protein 8 members (LRRC8s) form volume-regulated anion channels and play an important role in macrophage functions by regulating cytokine and chemokine production. We herein examined the role of LRRC8A in IL-8 and IL-10 expression in THP-1-differentiated M-like macrophages (M-MACs), which are a useful tool for investigating TAMs.
