Background: Up to 65% of patients with chronic myeloid leukemia (CML) who are treated with imatinib do not achieve sustained deep molecular response, which is required to attempt treatment-free remission. Asciminib is the only approved BCR::ABL1 inhibitor that Specifically Targets the ABL Myristoyl Pocket. This unique mechanism of action allows asciminib to be combined with adenosine triphosphate-competitive tyrosine kinase inhibitors to prevent resistance and enhance efficacy.

Astrocytes contribute to toll-like receptor 2-mediated neurodegeneration and alpha-synuclein pathology in a human midbrain Parkinson's model.

Transl Neurodegener

December 2024

School of Medical Sciences, Faculty of Medicine and Health and the Brain and Mind Centre, University of Sydney, Camperdown, NSW, 2050, Australia.

Background: Parkinson's disease (PD) is characterised by degeneration of ventral midbrain dopaminergic (DA) neurons and abnormal deposition of α-synuclein (α-syn) in neurons. Activation of the innate immune pathogen recognition receptor toll-like receptor 2 (TLR2) is associated with exacerbation of α-syn pathology. TLR2 is increased on neurons in the PD brain, and its activation results in the accumulation and propagation of α-syn through autophagy inhibition in neurons.

School of Psychological Sciences and Turner Institute for Brain and Mental Health, Monash University, Victoria, 3800, Australia.

Secondary motor cortical regions, such as the supplementary motor area (SMA) are involved in planning and learning motor sequences, however the neurophysiological mechanisms across these secondary cortical networks remain poorly understood. In primary motor cortex, changes in excitatory and inhibitory neurotransmission (E:I balance) accompany motor sequence learning. In particular, there is an early reduction in inhibition (i.

Australian parents' attitudes, perceptions and supply of alcohol to adolescents: a national cross-sectional survey.

Health Promot Int

December 2024

Centre for Alcohol Policy Research, School of Psychology and Public Health, La Trobe University, Kingsbury Drive, Bundoora, Victoria 3086, Australia.

Parental supply of alcohol to adolescents is associated with increased risk of subsequent adolescent alcohol use and harms, so identifying factors associated with parents' decision-making is critical. This study examined how parental supply is associated with attitudes toward adolescent alcohol use, perceived norms of parental supply, perceived behavioural control and perceived acceptable age to drink alcohol. A total of 1197 Australian parents with children aged 12-17 years completed an online cross-sectional survey assessing their parental supply behaviours, attitudes and perceptions in April 2022.

Article Synopsis
  • * A systematic search yielded 62 relevant trials, revealing that 87% did not factor in multiple births for sample size calculations, and almost half ignored the clustering effect of multiple births in their analyses.
  • * The findings highlight ongoing issues in trial designs that overlook the complexities of multiple births, signaling a need for trialists to better incorporate these considerations and for readers to critically assess the methodologies in neonatal research.

The impact of fibre and prebiotic interventions on outcomes in cancer and haematopoietic stem cell transplantation: A systematic review.

Clin Nutr

November 2024

Section of Nutrition, Department of Metabolism, Digestion and Reproduction, Faculty of Medicine, Imperial College London, London, W12 0NN, UK. Electronic address:

Background & Aims: Cancer therapy is associated with a range of toxicities that severely impact patient well-being and a range of clinical outcomes. Dietary fibre/prebiotics characteristically improve the gastrointestinal microenvironment, which consequently elicits beneficial downstream effects that could be relevant to the prevention and management of treatment-related toxicities. Despite the compelling theoretical scientific rationale there has been limited effort to synthesise the available evidence to conclude such scientific underpinning to the clinical use of fibre/prebiotics in cancer patients.

Novel loci and biomedical consequences of iron homoeostasis variation.

Commun Biol

December 2024

BHF Cardiovascular Epidemiology Unit, Department of Public Health and Primary Care, University of Cambridge, Cambridge, UK.

Iron homoeostasis is tightly regulated, with hepcidin and soluble transferrin receptor (sTfR) playing significant roles. However, the genetic determinants of these traits and the biomedical consequences of iron homoeostasis variation are unclear. In a meta-analysis of 12 cohorts involving 91,675 participants, we found 43 genomic loci associated with either hepcidin or sTfR concentration, of which 15 previously unreported.
