We show that a non-Hermitian lattice with a disclination can host topological disclination states that are induced by on-site gain and loss. The disclination states are inherently non-Hermitian as they do not exist in the limit of zero gain or loss. They arise from charge fractionalization in the non-Hermitian lattice, which we establish using non-Hermitian Wilson loops calculated with biorthogonal products.

A Self-Reinforced "Microglia Energy Modulator" for Synergistic Amyloid-β Clearance in Alzheimer's Disease Model.

Angew Chem Int Ed Engl

December 2024

Nanyang Technological University, School of Chemistry, Chemical Engineering and Biotechnology, 21 Nanyang Link, 637371, Singapore, SINGAPORE.

Microglial phagocytosis is a highly energy-consuming process that plays critical roles in clearing neurotoxic amyloid-β (Aβ) in Alzheimer's disease (AD). However, microglial metabolism is defective overall in AD, thereby undermining microglial phagocytic functions. Herein, we repurpose the existing antineoplastic drug lonidamine (LND) conjugated with hollow mesoporous Prussian blue (HMPB) as a "microglial energy modulator" (termed LND@HMPB-T7) for safe and synergistic Aβ clearance.

Limited by the two mutually exclusive physicochemical processes of separation and recombination of photogenerated carriers, achieving photoluminescence and photocatalysis simultaneously is extremely challenging but essential for ever-growing complex issues and specialized scenarios. Here we proposed a biomimetic isolation-conduction strategy induced by an arene-perfluoroarene (A-P) interaction for enabling photoluminescence and photocatalytic hydrogen evolution reaction (HER) activity in the co-assembly of aromatic monomers and octafluoronapthalene (OFN). Inspired by the isolation-conduction effect of periodic isolation of myelin sheaths on the axons of vertebrate nerve fibers by node of Ranvier, we use OFN as a molecular isolator embedded in the aromatic monomers array to block the singlet-to-triplet pathway, while the enlarged intermolecular dipoles resulting from the A-P interactions facilitate the conduction of photogenerated carriers in the isolated regions.

Asymmetric synthesis relies on seamless transmission of stereochemical information from a chiral reagent/catalyst to a prochiral substrate. The disruption by substrates' structural changes presents a hurdle in innovating generality-oriented asymmetric catalysis. Here, we report a strategy for substrate adaptability by exploiting a fundamental physicochemical phenomenon-ion hydration, in developing remote desymmetrization to access P-stereogenic triarylphosphine oxides and sulfides.

December 2024

Department of Diagnostic Radiology, Yong Loo Lin School of Medicine, National University of Singapore, Singapore, 119074, Singapore.

The unprecedented success of mRNA vaccines against COVID-19 has inspired scientists to develop mRNA vaccines for cancer immunotherapy. However, using nucleoside modified mRNA as vaccine, though evading innate immune toxicity, diminishes its therapeutic efficacy for cancers. Here, we report a polyvalent stimulator of interferon genes (STING) activating polymer (termed as PD) to bolster the immunogenicity of mRNA vaccine.

Article Synopsis
  • The alkaline oxygen evolution reaction (OER) involves four key reactions and poses challenges in balancing Gibbs free energies of intermediates like HO*, O*, and HOO* at active sites.
  • A new high-entropy metal-organic framework using Fe, Ni, Co, Cu, and Y is developed via electrodeposition, resulting in the FeCoNiCuYP/C composite, which features an amorphous structure with several active sites.
  • This composite shows enhanced OER performance in alkaline solutions, with a low overpotential of 316 mV at 100 mA/cm², attributed to the optimized electronic structure and binding strengths facilitated by Co/Ni and Fe atom interactions.

HClO-Activated Near-Infrared Chemiluminescent Probes with a Malononitrile Group for In-Vivo Imaging.

Adv Mater

December 2024

Joint School of National University of Singapore and Tianjin University, International Campus of Tianjin University, Binhai New City, Fuzhou, 350207, China.

Chemiluminescence (CL) imaging has emerged as a powerful approach to molecular imaging that allows exceptional sensitivity with virtually no background interference because of its unique capacity to emit photons without an external excitation source. Despite its high potential, the application of this nascent technique faces challenges because the current chemiluminescent agents have limited reactive sites, require complex synthesis, are insufficiently bright, and lack near-infrared emission. Herein, a series of HClO-activated chemiluminescent probes that exhibit robust near-infrared emission are studied.

Imaging the film formation process of waterborne barrier coatings in situ with nanoscopic resolution is very challenging, which limits the understanding of the underlying mechanisms and rational design of the materials. Here this challenge is tackled using in situ cryogenic transmission electron microscopy (cryoTEM) in combination with electron tomography (cryoET), which allows 3D imaging of the process with <1 nm resolution. By monitoring the film formation process of poly(ethylene-co-methacrylic acid) (EMAA) ionomer dispersion, onion-like nano-aggregates are captured.

The [5,5]-sigmatropic rearrangement is a less-studied reaction and may be strategically utilized to devise unique synthetic processes. Herein, we document a diboron-enabled [5,5]-sigmatropic rearrangement for practical synthesis of BINAM derivatives. Mechanistically, a concerted activation of azonaphthalenes by diboron creates a unique ten-membered transition state, which subsequently triggers a [5,5]-sigmatropic rearrangement.

Metals are an emerging topic in cancer immunotherapy that have shown great potential in modulating cancer immunity cycle and promoting antitumor immunity by activating the intrinsic immunostimulatory mechanisms which have been identified in recent years. The main challenge of metal-assisted immunotherapy lies in the fact that the free metals as ion forms are easily cleared during circulation, and even cause systemic metal toxicity due to the off-target effects. With the rapid development of nanomedicine, metal-based smart nanosystems (MSNs) with unique controllable structure become one of the most promising delivery carriers to solve the issue, owing to their various endogenous/external stimuli-responsiveness to release free metal ions for metalloimmunotherapy.
