Sexually dimorphic dopaminergic circuits determine sex preference.

Science

January 2025

Department of Neurology, the First Affiliated Hospital, Neuroscience Research Center, Key Laboratory of Biomedical Information Engineering of Ministry of Education, School of Life Science and Technology, Xi'an Jiaotong University, Xi'an, China.

Sociosexual preference is critical for reproduction and survival. However, neural mechanisms encoding social decisions on sex preference remain unclear. In this study, we show that both male and female mice exhibit female preference but shift to male preference when facing survival threats; their preference is mediated by the dimorphic changes in the excitability of ventral tegmental area dopaminergic (VTA) neurons.

Network-Based Methods for Deciphering the Oxidizability of Complex Leachate DOM with OH/O via Molecular Signatures.

Environ Sci Technol

January 2025

School of Environmental Science and Engineering, Shanghai Engineering Research Center of Solid Waste Treatment and Resource Recovery, Shanghai Jiao Tong University, Shanghai 200240, China.

In landfill leachates containing complex dissolved organic matter (DOM), the link between individual DOM constituents and their inherent oxidizability is unclear. Here, we resolved the molecular signatures of DOM oxidized by OH/O using FT-ICR MS, thereby elucidating their oxidizability and resistance in concentrated leachates. The comprehensive gradual fragmentation of complex leachate DOM was then revealed through a modified machine-learning framework based on 43 key pathways during ozonation.

A dual-mode detection platform utilizing colorimetric and Raman was developed based on the exponential amplification reaction (EXPAR) strategy and a "core-satellite" structure constructed by bimetallic nanozymes to detect chloramphenicol (CAP). Initially, DNA-gated metal-organic frameworks (MOFs) incorporating cascaded amplification were used to be nanocarriers for the colorimetric and Raman reporter molecules (3,3',5,5'-tetramethylbiphenyl; TMB). Subsequently, assembled DNA served as gatekeepers to create a stimulus-responsive DNA-gated MOF (TMB@DNA/MOF).

Vibrio parahaemolyticus propels itself through liquids using a polar flagellum and efficiently swarms across surfaces or viscous environments with the aid of lateral flagella. H-NS plays a negative role in the swarming motility of V. parahaemolyticus by directly repressing the transcription of the lateral flagellin gene lafA.

We evaluated the prognostic and therapeutic significance of measurable residual disease (MRD) during remission induction in pediatric acute lymphoblastic leukemia (ALL) patients. In the CCCG-ALL-2015 protocol, 7640 patients were categorized into low-, intermediate-, or high-risk groups based on clinical and genetic features. Final risk classification was determined by MRD assessed via flow cytometry on Days 19 and 46 of remission induction, with additional intensified chemotherapy for Day 19 MRD ≥1%.

Introduction: We aimed to explore the difference in choroidal vascular parameters using swept-source optical coherence tomography (SS-OCT) in patients with idiopathic intracranial hypertension (IIH) compared to controls. We also explored the ability of the choroidal parameters to reflect elevated intracranial pressure (ICP) in patients with IIH.

Methods: This observational study recruited patients diagnosed with IIH and healthy controls.

Background: Giant hydronephrosis as an rare condition is often caused by chronic ureteral obstruction. Nephroplication is a crucial procedure to improve urinary drainage in the kidney-sparing surgery for patients with giant hydronephrosis. However, traditional nephroplication via suturing kidney has technical difficulty and many potential risks.

Alzheimers Dement

December 2024

West China Hospital of Sichuan University, Chengdu, China.

Background: Alzheimer's disease (AD) and Frontotemporal dementia (FTD) are the two most common causes of early-onset dementia. Differentiating between AD and FTD can be challenging due to overlapping cognitive and behavioral manifestations. However, evidence on non-invasive and early-stage biomarkers for differentiating AD and FTD is still limited.

RNF7 (Ring Finger Protein 7) is a key component of CRLs (Cullin-RING-type E3 ubiquitin ligases) and has been found to possess intrinsic anti-ROS capabilities. Aberrant expression of RNF7 has been observed in various tumor types and is known to significantly influence tumor initiation and progression. However, the specific role of RNF7 in glioblastoma remains unclear.

Alzheimers Dement

December 2024

Innovation Center for Neurological Disorders, Xuanwu Hospital, Capital Medical University, Beijing, China;, Beijing, China.

Background: There is an urgent need for accurately predicting amyloid-beta (Aβ) pathology in Alzheimer's disease (AD) using non-invasive and readily available methods, particularly in the early stages of the disease. While blood-based biomarkers have shown promising consistency with brain Aβ pathology, there remains a scarcity of data predicting Aβ pathology using these biomarkers across different clinical settings in the Chinese population.

Method: We retrospectively selected 278 participants from five hospitals across China, comprising 127 patients with mild cognitive impairment (MCI) and 151 with dementia.

Macrophage Membrane-Cloaked ROS-Responsive Albumin Nanoplatforms for Targeted Delivery of Curcumin to Alleviate Acute Liver Injury.

Mol Pharm

January 2025

Sichuan Engineering Research Center for Biomimetic Synthesis of Natural Drugs, School of Life Science and Engineering, Southwest Jiaotong University, Chengdu 610031, China.

Developing low-toxicity, high-efficacy, and fast-acting strategies to manage acute liver injury (ALI) is critical due to its rapid progression and potential for severe outcomes. Curcumin (CUR) has shown promise in ALI therapy due to its ability to modulate the inflammatory microenvironment by scavenging reactive oxygen species (ROS). Nevertheless, CUR is highly hydrophobic limiting its bioavailability and effective in vivo transport, which hinders its further application.

Regeneration of diabetic bone defects remains a formidable challenge due to the chronic hyperglycemic state, which triggers the accumulation of advanced glycation end products (AGEs) and reactive oxygen species (ROS). To address this issue, we have engineered a bimetallic metal-organic framework-derived Mn@CoO@Pt nanoenzyme loaded with alendronate and Mg ions (termed MCPtA) to regulate the hyperglycemic microenvironment and recover the osteogenesis/osteoclast homeostasis. Notably, the Mn atom substitution in the CoO nanocrystalline structure could modulate the electronic structure and significantly improve the SOD/CAT catalytic activity for ROS scavenging.

Simultaneous Blockade of CD209 and CD209L by Monoclonal Antibody Does Not Provide Sufficient Protection Against Multiple Viral Infections In Vivo.

Immunology

January 2025

The Key Laboratory for Human Disease Gene Study of Sichuan Province, Department of Laboratory Medicine, Sichuan Provincial People's Hospital, University of Electronic Science and Technology of China, Chengdu, China.

Many virus species, including Ebola virus, Marburg virus, SARS-CoV-2, dengue virus (DENV) and Zika virus (ZIKV), exploit CD209 and CD209L as alternative or attachment receptors for viral cis- or trans-infection. Thus, CD209 and CD209L may be critical targets for the development of therapeutic monoclonal blocking antibody drugs to disrupt the infection process caused by multiple viruses. Here, we produced a human chimeric monoclonal blocking antibody that simultaneously blocks CD209 and CD209L, namely 7-H7-B1.
