is a widely distributed nosocomial pathogen that causes various acute and chronic infections, particularly in immunocompromised patients. In this study, the activities of the K9-specific virulent phage AM24 and phage-encoded depolymerase DepAPK09 were assessed using in vivo mouse sepsis and burn skin infection models. In the mouse sepsis model, in the case of prevention or early treatment, a single K9-specific phage or recombinant depolymerase injection was able to protect 100% of the mice after parenteral infection with a lethal dose of of the K9-type, with complete eradication of the pathogen.

Alzheimer's disease (AD) pathogenesis is correlated with the membrane content of various lipid species, including cholesterol, whose interactions with amyloid precursor protein (APP) have been extensively explored. Amyloid-β peptides triggering AD are products of APP cleavage by secretases, which differ depending on the APP and secretase location relative to ordered or disordered membrane microdomains. Once an exotic add-on to fluorescence microscopy for life science research, fluorescence lifetime imaging (FLIm) has become a powerful and increasingly utilised technique owing to its self-calibration nature, which affords superior quantification over conventional steady-state fluorescence imaging. This review focuses on the state-of-the-art implementation of FLIm related to the formulation, release, dosage, and mechanism of action of drugs aimed for innovative diagnostics and therapy. Quantitative measurements using FLIm have appeared instrumental for encapsulated drug delivery design, pharmacokinetics and pharmacodynamics, pathological investigations, early disease diagnosis, and evaluation of therapeutic efficacy.

The Orange Carotenoid Protein (OCP) is a unique water-soluble photoactive protein that plays a critical role in regulating the balance between light harvesting and photoprotective responses in cyanobacteria. The challenge in understanding OCP´s photoactivation mechanism stems from the heterogeneity of the initial configurations of its embedded ketocarotenoid, which in the dark-adapted state can form up to two hydrogen bonds to critical amino acids in the protein's C-terminal domain, and the extremely low quantum yield of primary photoproduct formation. While a series of experiments involving point mutations within these contacts helped us to identify these challenges, they did not resolve them.

A total chemical synthesis of spacer-armed Forssman pentasaccharide is reported. The choice of the 2(donor) + 3(acceptor) block scheme, the optimal combination of a limited number of simple protecting groups and the sequence of deprotection steps allowed to achieve the high yield and stereoselectivity of glycosylation and to avoid losses during deprotection. The target pentasaccharide was obtained in a 10-mg scale.

Comparison of the reactivity of sialyl chlorides and bromides based on -acetylneuraminic acid (Neu5Ac) and its deaminated analogue (KDN) in reactions with MeOH and -PrOH without a promoter revealed that the acetoxy group at C-5 in a molecule of a sialic acid glycosyl donor can destabilize the corresponding glycosyl cation making the S1-like reaction pathway unfavorable. A change to the S2-like reaction pathway ensures preferential formation of the α-glycoside.
