Gastric cancer is a malignant tumor associated with a high mortality rate. Recently, emerging evidence has shown that ferroptosis, a regulated form of cell death induced by iron (Fe)-dependent lipid peroxidation. Nuclear factor E2 related factor 2 (NRF2) is a key regulator of intracellular oxidation homeostasis that plays a pivotal role in controlling lipid peroxidation, which is closely related to the process of ferroptosis.

Article Synopsis
  • Gastric cancer (GC) is a common cancer affecting the digestive system, and Weifuchun (WFC), a traditional Chinese herbal treatment, is used for stomach issues but its effects on GC are not well-studied.
  • In a clinical trial with GC patients, combining WFC with adjuvant chemotherapy significantly lowered the rates of cancer recurrence and metastasis compared to chemotherapy alone over a 6-month period.
  • WFC was found to have multiple active ingredients that influence key signaling pathways related to cell growth and death, particularly affecting the AMPK and NFKB pathways, suggesting it may help inhibit GC cell proliferation and promote apoptosis.

Article Synopsis
  • This study explores how cholangiocarcinoma (CCA) relies on protein homeostasis for growth and how disrupting this balance with celastrol may induce stress in the cancer cells.
  • Researchers found that the effectiveness of celastrol varies with the PTEN protein status in CCA cells, where PTEN-positive cells showed resistance and PTEN-negative cells displayed increased sensitivity to celastrol.
  • The study identified celastrol's mechanism of action as disrupting proteasomal function by targeting PSMB5 and suggested potential combination therapies with autophagy inhibitors to enhance treatment effectiveness in resistant cancers.

Ethnopharmacological Relevance: Chemotherapy tolerance weakened efficacy of chemotherapy drugs in the treating gastric cancer (GC). Banxiaxiexin decoction (BXXXD) was widely used in digestive diseases for thousands of years in Traditional Chinese medicine (TCM). In order to better treat GC, three other herbs were added to BXXXD to create a new prescription named Modified Banxiaxiexin decoction (MBXXXD).

Polyphyllin VII induces CTC anoikis to inhibit lung cancer metastasis through EGFR pathway regulation.

Int J Biol Sci

November 2023

Institute of Oncology, Shanghai Municipal Hospital of Traditional Chinese Medicine, Shanghai University of Traditional Chinese Medicine (TCM), Shanghai, 200071, China.

Circulating tumor cells (CTCs) are cells that detach from the primary tumor and enter the bloodstream, playing a crucial role in the metastasis of lung cancer. Unfortunately, there is currently a lack of drugs specifically designed to target CTCs and prevent tumor metastasis. In this study, we present evidence that polyphyllin VII, a potent anticancer compound, effectively inhibits the metastasis of lung cancer by inducing a process called anoikis in CTCs.

To use metagenomic sequencing to compare the differences in intestinal microbiota species and metabolic pathways in patients with hepatitis B cirrhosis with or without ascites and further explore the correlation between the differential microbiota and clinical indicators and metabolic pathways. 20 hepatitis B cirrhosis cases [10 without ascites (HBLC-WOA), 10 with ascites (HBLC-WA), and 5 healthy controls (HC)] were selected from the previously studied 16S rRNA samples. Metagenome sequencing was performed on the intestinal microbiota samples.

Background: Electromyography of the cricopharyngeal muscle (CP-EMG) is one of many assessment tools for dysphagia. The key to performing EMG and BTX injections is to precisely locate the cricopharyngeal muscle with an electrode. One of the main difficulties of electrode insertion is the fact that the CP muscle is located deep within the neck.

Article Synopsis
  • Cholestatic liver fibrosis (CLF) develops due to damage to bile ducts and involves the Notch signaling pathway, with reduced Numb protein potentially influencing disease progression.
  • This study examined how manipulating the Numb gene in stem cells affected liver repair in rat models of CLF, revealing that Numb overexpression promoted healthy liver cell (hepatocyte) formation, while its knockdown led to harmful epithelial cell growth.
  • The research suggests that enhancing Numb function could be a potential therapeutic strategy for treating CLF by guiding the differentiation of stem cells towards beneficial liver cell types.

Cancer Cell Int

November 2022

Institute of Oncology, Shanghai Municipal Hospital of TCM, Shanghai University of Traditional Chinese Medicine (TCM), Shanghai, 200071, People's Republic of China.

Metastasis is the primary cause of death in lung cancer patients. However, until now, effective drugs and intervention strategies for treating lung cancer metastasis have been lacking. This hypothesis focuses on circulating tumour cells (CTCs) to develop a new antimetastatic therapeutic strategy for lung cancer.

The oral cavity and the intestine are the main distribution locations of human digestive bacteria. Exploring the relationships between the tongue coating and gut microbiota, the influence of the diurnal variations of the tongue coating microbiota on the intestinal microbiota can provide a reference for the development of the disease diagnosis and monitoring, as well as the medication time. In this work, a total of 39 healthy college students were recruited.

Background: Metastasis and/or recurrence can decrease the survival time of gastric cancer patients undergoing radical operation. Among them, those with stage IIIb and IIIc are especially at a high risk of metastasis and recurrence. The traditional Chinese medicine collaborative model (TCMCM) has been used in the treatment of cancer; however, its effects have not been systematically evaluated.

Rationale: Gardner syndrome is a rare autosomal dominant disorder with a high degree of penetrance, which is characterized by intestinal polyposis, osteomas, and dental abnormalities. Majority of patients with Gardner syndrome will develop colorectal cancer by the age of 40 to 50 years. Mutations in the adenomatous polyposis coli gene are supposed to be responsible for the initiation of Gardner syndrome.

The aim of this study is to study the relationship between programmed cell death-1 ligand (PD-L1) and human epidermal growth receptor 2 (HER2) and the clinical-pathological features of gastric cancer (GC) and its predictive effect on the prognosis of gastric cancer (GC) patients. A retrospective analysis was performed on 113 patients undergoing GC surgery. The expression of PD-L1 and HER2 in GC and paired adjacent nontumor tissues was detected by immunohistochemistry or fluorescence hybridization, and the relationships between PD-L1 and HER2 expression and clinical-pathological features and survival were analyzed by chi-square analysis, Pearson analysis, logistic regression analysis, Kaplan-Meier analysis, and Cox regression model.

Article Synopsis
  • Numb is identified as a negative regulator of the Notch signaling pathway, which plays a crucial role in the differentiation of hepatic progenitor cells into cholangiocytes during cholestatic liver fibrosis (CLF).
  • A study using CLF rats with knocked down Numb gene showed that treatment with Huang Qi Decoction (HQD) in combination with bone marrow-derived mesenchymal stem cells (BMSC) effectively reduced liver fibrosis by inhibiting the Notch pathway and altering protein and gene expressions associated with liver damage.
  • In vitro results indicated that HQD diminishes the differentiation of WB-F344 cells into cholangiocytes, and this effect is lessened when the Numb gene is knocked down, suggesting that targeting Numb

Histone deacetylase 4 (HDAC4) plays a vital role in chondrocyte hypertrophy and bone formation. To investigate the function of HDAC4 in postnatal skeletal development, the present study developed lineage‑specific HDAC4‑knockout mice [collagen type 2α1 (Col2α1)‑Cre, HDAC4d/d mice] by crossing transgenic mice expressing Cre recombinase. Thus, a specific ablation of HDAC4 was performed in Col2α1‑expressing mice cells.
