Milk serves as an important dietary source of bioactive peptides, offering notable benefits to individuals. Among the antioxidant short peptides (di- and tripeptides) generated from gastrointestinal digestion are characterized by enhanced bioavailability and bioaccessibility, while assessing them individually presents a labor-intensive and expensive challenge. Based on 4 distinct types of AA descriptors (physicochemical, 3-dimensional structural, quantum, and topological attributes) and genetic algorithms for feature selection, 1 and 4 machine learning-predicted models separately for di- and tripeptides with 2,2-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt radical scavenging capacity exhibited excellent fitting and prediction ability with random forest regression as machine learning algorithm.

The spectral matching strategy of MS2 fragment spectrograms serves as a ubiquitous method for compound characterization within the matrix. Nevertheless, challenges arise due to the deficiency of distinctions in spectra across instruments caused by coelution peak-derived fragments and incompleteness of the current spectral reference database, leading to dilemma of multidimensional omics annotation. The spectral database-based mass spectrometry (MS) matching strategy is versatile for structural annotating in ingredient fluctuation profiling mediated by external interferences. However, the systematic variability of MS pool attributable to aliasing peaks and inadequacy of present spectral database resulted in a substantial metabolic feature depletion. Complete self-assembly and reassembly behavior of bitter peptide-protein necessitates multilevel theories that encompass phenomena ranging from the self-assembly of recombinant complex to atomic trajectories. An extension to the level of mechanism method was put forth, involves limited enzymatic digestion and bottom-up proteomics to dissect inherent heterogeneity within β-LG and β-LG-PPGLPDKY complex and uncover conformational and dynamic alterations occurring in specific local regions of the model protein. Bitter peptide PPGLPDKY spontaneously bound to IIAEKTK, IDALNENK, and YLLFCMENSAEPEQSLACQCLVR regions of β-LG in a 1:1 stoichiometric ratio to mask bitterness perception.

Scandal of detecting 1,2-propanediol (PD) in milk brought a crisis to the trust of consumers in dairy industry, and investigations focused effect of PD on digestive behavior of milk were still restricted. Long short-term memory amalgamated to quasi-targeted lipidomics was applied to monitor dynamics changes of lipids during digestion and the pseudo-first-order kinetic model elucidated that PD elevated the digestibility of lipid with the degradation rate (S) ranged from 4440.31 to 5665.
