To investigate the effect of arginine vasopressin (AVP) in the peripheral circulation on burn shock in the rat, AVP and its nonselective V1/V2 receptor antagonist d(CH2)5Tyr (Et)VAVP were administered intravenously in burn shocked rats. Cardiovascular parameters were recorded including left ventricular systolic pressure (LVSP), +/- dP/dt max, total area of the cardiac force loop (Lo), mean arterial blood pressure (MAP), heart rate (HR) and electrocardiogram (ECG). Our results showed that AVP failed to increase MAP in burned rats whereas it elicited a greater fall in LVSP, +/- dP/dt max and Lo and MAP than seen in control burned rats and hastened the onset of the decompensatory phase of burn shock resulting in the early death of burn shocked animals.

This paper shows that the alcoholic extract from Polygonatum odoratum and Cuscuta australis increases the percentage of phagocytosis and the index of phagocytosis of macrophages as well as hemolysin and proliferation of lymphocytes up to normal level in mice with II+ burn injury covering 5% body surface. It follows that both Polygonatum odoratum and Cuscuta australis may serve as a kind of immunopotentiator for mammals.

[The selection of treatment for giant-cell tumor of bone].

Zhonghua Zhong Liu Za Zhi

July 1990

Changhai Hospital, Second Military Medical College, Shanghai.

Eighty-two patients with giant-cell tumor of the bone were operated with 4 different methods depending on the site of tumor, extent of local invasion and degree of malignancy. Curettage plus filling operation was performed for Grade I tumors and localized lesions. Resection of the tumor segment and replacement of the joint with allograft or artificial joint were done for Grade II tumors involving the joint but with clear borders.

Plasma fibronectin concentrations were measured in 78 patients with traumatic fracture and 80 healthy people as a control. The results showed that plasma fibronectin concentrations of the patients without complicated infection were significantly decreased 5 to 48 hours after fracture. As comparison with the control, the mean decrease was 56 (16.

The random migration and chemotaxis of polymorphonuclear leukocytes (PMN) in agarose were studied in 21 patients with psoriasis and 10 healthy controls. Different concentrations of LTB4 were used as chemoattractants. The random migration and chemotaxis towards lower concentrations of LTB4 were enhanced and that towards higher concentrations of LTB4 reduced in psoriasis.

[Effect of radix Paeoniae rubrae extract on mesenteric microcirculation].

Zhonghua Zheng Xing Shao Shang Wai Ke Za Zhi

June 1990

Dept of Pharmacology, Second Military Medical College.

The effect of radix Paeoniae rubrae extract on mesenteric microcirculation was studied in 20 scalded SD rats. It was found that this drug effectively prevented some disturbances of mesenteric microcirculation induced by burn. It inhibited the contraction of arterioles, decreased the adhesion of leucocytes on the venous wall, reduced and delayed the aggregation of erythrocytes in microvessels.

Hypothalamic slices were prepared with a vibratome. The spontaneous discharge of 46 paraventricular neurons was recorded by glass microelectrode from 20 such slices, and effects of serotonin on spontaneous discharge were observed. After perfusing the brain slices with artificial cerebro-spinal fluid containing serotonin (10(-6) mol/L) for 3 min, discharge rate of 16 units is significantly increased with latency 1.

This experimental hepatic artery embolization with Camptothecin (CPT) albumin microspheres was performed in normal rats and rats with liver tumor. The basic properties of the CPT albumin microspheres and the effects of target treatment on rats with liver tumor were studied. The results showed that the concentration of the drug released into hepatic vein was almost stable for over 3 hours after hepatic arterial infusion of CPT albumin microspheres, the partial and complete recovery of hepatic artery inflow occurred within 50-70 days after embolization.

The effects of intraventricular injection (ICV) of oxytocin (OT) and antioxytocin serum (AOTS) on the pain threshold and electroacupuncture (EA) analgesia in rats were investigated in this study. The potassium iontophoresis induced tailflick was used to measure the pain threshold. An increase of 20-38% in the pain threshold was observed within 80 min after OT injection (50 ng), while the OT administration (50 ng) in combination with EA produced a dramatic increase of 139-234% in the pain threshold, which was much higher than that in the saline-EA group (P less than 0.

Dynamic observation on thyroid function in severe head injury.

Chin Med J (Engl)

April 1990

Department of Neurosurgery, Changhai Hospital, Second Military Medical College, Shanghai.

The dynamic changes of thyroid hormone levels in patients with severe head injury were observed. The results showed that the serum thyroid hormone levels changed rapidly within several hours after the injury and the mean levels of serum total T3 and T4 (TT3, TT4) decreased while high reverse triiodothyronine (rT3) increased significantly. Serum TT3, TT4 and rT3 levels returned to normal gradually in 2 to 3 weeks after the injury in the surviving group, whereas serum total T4 gradually declined and rT3 steadily increased in the fatal group.

The concentration of glucocorticoids (GC) in plasma was maintained at stress level, 20-40 micrograms/dl, for 3 days by subcutaneous injection of hydrocortisone (F) in polyvinyl alcohol (PVA) into rats, and the specific binding of [3H]Dexamethasone (Dex) in liver, spleen and brain was determined before and after injection. The binding capacity of glucocorticoid receptor (GR) in liver and spleen was decreased significantly 1 h after injection and maintained at low level for several days after the concentration of GC in plasma had returned to the normal level. The Kd was not altered.

In order to investigate the pathological changes and stages of development of degenerative arthritis, a model of degenerative arthritis of patello-femoral joint was established in 30 rabbits by shortening the patellar tendon by 4 mm. The animals were killed 1,2,4,6,8,12 weeks after operation, and the results showed that the compression force between patella and femoral condyle increased 1/4 and a typical change of degenerative arthritis was a observed grossly and histologically and ultrastructurally. The authors consider that this kind of model may be used for further study of prevention and treatment of degenerative arthritis.

1) There exists a CR of GR in human leukocytes, PMN, and monocytes with the peak values from 0400 to 0800 hr and the trough values between 2300 and 0000 hr. The difference between them was significant statistically. 2) The FI of the chemotactic migration rate of PMN by cortisol also showed diurnal changes which were synchronous with that of GR.

In an effort to study the structure-activity relationship of analogs of baclofen and to search for more potent and less toxic muscle relaxants and analgesics, fifteen 3-substituted pheny-4-aminobutyric acids and their derivatives were synthesized. Preliminary pharmacological tests in albino mice showed that some of the target compounds exhibited muscle relaxant and analgesic effects. Compounds Ib, IIb, IIIa, IIIc and compounds Ib, IIb, IIIa, IIIb were, respectively, more potent in hot plate tests and morphine-induced straub tail reaction tests than other analogs, but they were still less potent than baclofen.
