Mechanism of action of arginine vasopressin on acute ischemic brain edema.

The purpose of this experiment is to study the role of arginine vasopressin (AVP) in acute cerebral ischemic edema in Mongolian gerbils. The results show that intracerebroventricular injection (ICV) of AVP exacerbates acute ischemic brain edema, while ICV of AVP antiserum significantly decreases the ischemic brain edema. Nimodipine (calcium antagonist) cannot block this role of AVP in brain edema.

The number of estrogen receptors (ER) in human peripheral leucocytes in 12 women with menopausal type II diabetes was measured with radio-ligand binding method. The results were compared with those of 12 menopausal women without diabetes and 12 normal women of childbearing age. It was found that the number of ER in the patients was significantly decreased.

Study Objective: Two selective V1 and V2 receptor antagonists of arginine vasopressin, d(CH2)5Tyr(Me)AVP and d(CH2)5[D-Ile2, Ile4, Ala9-NH2]AVP, were given intravenously in burn shocked rats to investigate the respective effects of V1 and V2 receptor blockade on the haemodynamic variables in burn shock.

Design: Computer assisted on line real time measurements of mean arterial blood pressure, diastolic blood pressure, left ventricular systolic pressure, dP/dtmax, and heart rate were used to study the effects of the receptor antagonists during burn shock. In addition, the radioactive microsphere method was used to measure the changes of cardiac output and regional blood flows to heart, kidney, and liver in response to the antagonists during burn shock.

Hyperthyroxinemia model was made by giving thyroid tablet suspension to Wistar rats and hypothyroxinemia model was made by thyroidectomy. We measured serum thyroid hormone levels by RIA and the parameters of triiodothyronine receptors in rat hepatic cell nucleus by radio-ligand binding assay: Maximal binding capacity (Bmax) and Dissociation constant (Kd). Twenty-six cases of hepatocellular carcinoma were divided randomly into 3 groups, treated by transcatheter hepatic arterial chemo-embolizations with agents of MMC 20 mg, MMC 20 mg plus lipiodol 10 ml, and MMC 20 mg plus MTXmc 150 mg, respectively, 2 to 3 weeks before surgery. Pathologically, all main tumors in the resected specimens were necrosed to a certain extent, with extensive necrosis in the MMC-MTXmc group, whereas there was no necrosis of cancer cells in tumor capsules, daughter nodules, and intraportal vein emboli. We consider that the transcatheter arterial chemoembolization is effective in reducing tumor burden, but not enough in eradicating all cancer cells, so surgical resection should be carried out whenever resection is possible.

Circadian rhythm in glucocorticoid receptor (GR) was studied in the rat liver and human peripheral leukocytes. For rats exposed to a natural environmental photic cycle or a 12L:12D artificial light regime, peak values of hepatic GR were detected between 23:00 and 02:00 h. Except for a 4-hour advancement of the peak, a similar circadian rhythm of hepatic GR was detected in rats reared under a reversed lighting regimen (12D:12L; lights on between 18:30 and 06:30 h).

The action of glucocorticoids (GC) on neuronal cell membrane was studied in isolated and superfused guinea pig coeliac ganglia by the intracellular recording technique. Cortisol succinate (F) hyperpolarized the membrane potential of 47 of 179 cells and changed the cell's input resistance with a latency of less than 2 min in vitro. The effect persisted under low Ca2+/high Mg2+ superfusing condition and could be blocked by RU 38486, a competitive antagonist of GC cytosolic receptor.

Anti-tumor activity of cisplatin-capsulated microspheres with ethylcellulose (CDDPmc) was studied in Wistar rat bearing transplantable hepatocellular cancer. Seven days after inoculation into the rat's liver, normal saline, conventional cisplatin, placebo ethylcellulose microsphere and CDDPmc at comparable doses were infused into the proper hepatic artery. The results indicated that the rats treated with CDDPmc had a significantly slower tumor growth, more extensive tumor necrosis and longer survival as compared with the three other groups.

The photosensitizing effect of sulfonated aluminum phthalocyanine (ALSPC), a new photosensitizer, on human hepatoma cells (SMMC-7721) was studied in vitro. It was found that ALSPC was not cytotoxic even at a concentration of 100 micrograms/ml. In the photosensitizing experiments with different irradiation wavebands, the red waveband had the most marked killing effect, which coincided with the peak absorption band of ALSPC.

Burned rabbits' limbs, caused by electricity, were early debrided and covered by vascularized free flaps. This method could obviously shorten the healing time and improve local conditions. Blood vessels, 3 cm beyond the margin of the wound, can be used for anastomosis if they have the following characteristics, normal elasticity of vessel wall, intact endothelium with no separation from the media, and good arterial bleeding.

Using glutamate plus malate as substrates, the respiratory control ratio of liver mitochondria from burned male Sprague-Dawley rats with full skin thickness burns covering 20 per cent of the body surface area were increased at 45, 60, 75 and 90 min and using alpha-ketoglutarate, were increased at 60 and 75 min after burning, the peak being at 75 min postburn. The ADP/O ratio and the rate of ATP formation were also increased at about 75 min postburn and the ATP content in liver was increased at 120 min following burning. The ATP and creatine phosphate contents in heart tissue were increased at 120 min and at 105, 120 and 135 min postburn respectively.

ATPase-stained normal and topical nitrogen mustard treated Langerhans cells (LC) in guinea pig epidermis were studied quantitatively and morphologically by computerized image analysis. Significant correlation and linear regression were found between the perimeter and square root of its area for both cytoplasm and nucleus; a theoretical basis was thus given for morphological identification of LC. The directly measured and derived geometric parameters indicated not only substantial alteration of cytoplasm morphology of post-treated LC such as cytoplasmatosis and dendrites reduction but also a potential change of nuclear size and functional status.

Unipolar demand pacemaker myopotential inhibition by 5 different provocative maneuvers was studied in 48 patients with stable pacing rhythm out of 82 patients with unipolar pacemakers from 3 manufacturers implanted over the right pectoralis major to evaluate the incidence of inhibiting phenomenon and its clinical significance. Transient pacing inhibition occurred in 19 (39.6%) of the 48 patients.
