TurboID-mediated proximity labeling for screening interacting proteins of FIP37 in .

Plant Direct

December 2023

Shenzhen Branch, Guangdong Laboratory for Lingnan Modern Agriculture, Genome Analysis Laboratory of the Ministry of Agriculture, Agricultural Genomics Institute at Shenzhen Chinese Academy of Agricultural Science Shenzhen China.

Proximity labeling was recently developed to detect protein-protein interactions and members of subcellular multiprotein structures in living cells. Proximity labeling is conducted by fusing an engineered enzyme with catalytic activity, such as biotin ligase, to a protein of interest (bait protein) to biotinylate adjacent proteins. The biotinylated protein can be purified by streptavidin beads, and identified by mass spectrometry (MS).

Article Synopsis
  • SUMO (small ubiquitin-like modifier) plays a key role in plant development and stress responses by attaching to various proteins, but its secondary modifications through SUMOylation and ubiquitylation are not well understood.
  • Researchers created transgenic plants that lacked specific lysine residues in SUMO1 and SUMO2, resulting in normal development and heat stress tolerance while showing changes in sensitivity to other stress factors and hormones.
  • These findings suggest the importance of secondary modifications in plant defense and hormone signaling, and the new SUMO1(K0) variant can be used to further investigate SUMO conjugates and their functions.

Through crosstalk, FLAGELLIN SENSITIVE 2 (FLS2) and RESPIRATORY BURST OXIDASE HOMOLOG D (RBOHD) are involved in regulating the homeostasis of cellular reactive oxygen species (ROS) and are linked to the metabolic response of plants toward both biotic and abiotic stress. In the present study, we examined the metabolome of seedlings under drought and salt conditions to better understand the potential role of FLS2 and RBOHD-dependent signaling in the regulation of abiotic stress response. We identified common metabolites and genes that are regulated by FLS2 and RBOHD, and are involved in the response to drought and salt stress.

Fire is a common disturbance in many biomes, with both beneficial and detrimental effects on soil biology, which largely depend on fire intensity. However, little is known about the impact of fire on soil nematode communities in terrestrial ecosystem. In the present study, we investigated the effects of short-term prescribed fire on soil nematode communities and soil properties in an old-field grassland in Northern China.

Ethanol is a principal ingredient of alcoholic beverages with potential neurotoxicity and genotoxicity, and the ethanol-associated oxidative DNA damage in the central nervous system is well documented. Natural product may offer new options to protect the brain against ethanol-induced neurotoxicity. The male flower of (EUF) Oliver has been extensively utilized as the tea, the healthy hot drink on the market.

Extracellular DNA (eDNA) and intracellular DNA (iDNA) extensively exist in both terrestrial and aquatic environment systems and have been found to play a significant role in the nutrient cycling and genetic information transmission between the environment and microorganisms. As inert DNA sequences, eDNA is able to present stability in the environment from the ribosome enzyme lysis, therein acting as the historical genetic information archive of the microbiome. As a consequence, both eDNA and iDNA can shed light on the functional gene variety and the corresponding microbial activity.

Glioblastoma (GBM) is a central nervous system tumor with poor prognosis due to the rapid development of resistance to mono chemotherapy and poor brain targeted delivery. Chemoimmunotherapy (CIT) combines chemotherapy drugs with activators of innate immunity that hold great promise for GBM synergistic therapy. Herein, we chose temozolomide, TMZ, and the epigenetic bromodomain inhibitor, OTX015, and further co-encapsulated them within our well-established erythrocyte membrane camouflaged nanoparticle to yield ApoE peptide decorated biomimetic nanomedicine (ABNM@TMZ/OTX).

Article Synopsis
  • Previous research has shown that plants react to nutrient availability, but how they respond to different levels of nutrient enrichment, particularly nitrogen (N) and phosphorus (P), is not well understood.
  • A field experiment tested three levels of N (0, 10, and 40 g/m²/year) and P (0, 5, and 10 g/m²/year) to observe their effects on seed production in a dominant plant species in a temperate steppe.
  • Results indicated that N addition dramatically increased seed production, while P addition only enhanced seed output when nitrogen was also present, suggesting that nitrogen is the primary limiting nutrient for seed production in these ecosystems.

The distinguished properties of nanomaterials promote us to explore whether their intrinsic activities would be beneficial to disease treatment. Furthermore, understanding the molecular mechanism is thereby crucial for biomedical applications. Here, we investigate the therapeutic effects of single-walled carbon nanotubes (SWNTs) in a rat model of binge alcohol-induced neurodegeneration.

Article Synopsis
  • Climate change is affecting plants, insects, and their predators, which could change how they all interact with each other.
  • Scientists studied how higher levels of CO2, hotter temperatures, and less rainfall impact certain predator species like nematodes, spiders, ladybugs, and wasps.
  • They found that while some predators showed reduced survival with climate changes, most of them, especially spider and nematode species, actually did better in hunting their prey.

Seed germination plays an important role in mediating plant species composition of grassland communities under nitrogen (N) enrichment. Shifts of plant community structure with N-enhanced deposition in terrestrial ecosystems have occurred globally. Despite numerous studies about the effects of enhanced N deposition on mature plant communities, few studies have focused on seed germination.

Mass loss and nutrient release during litter decomposition drive biogeochemical cycling in terrestrial ecosystems. However, the relationship between the litter decomposition process and the decomposition stage, precipitation, and litter quality has rarely been addressed, precluding our understanding of how litter decomposition regulates nutrient cycling in various ecosystems and their responses to climate change. In this study, we measured mass loss as well as carbon and nutrient releases during the decomposition of 16 types of leaf litter under three precipitation treatments over 12 months in a common garden experiment (i.

Iron detection is one of the critical markers to diagnose multiple blood-related disorders that correspond to various biological dysfunctions. The currently available anemia detection approach can be used only for pre-treated blood samples that interfere with the actual iron level in blood. Real-time detection approaches with higher sensitivity and specificity are certainly needed to cope with the commercial level clinical analyses.

Unlabelled: Plant community diversity is conducive to maintain the regional ecosystems stability and ecosystem services. Seed germination is one of the main ways to regulate plant diversity, owing to seedling recruitment as a basis for plant community renewal. However, the exact mechanism of how plant litter affects seedling recruitment, and species richness is not yet fully understood.

Seed germination and seedling establishment play an important role in driving the responses of plant community structure and function to global change. Nitrogen (N) deposition is one of the driving factors of global change, which often leads to a loss in species richness in grassland ecosystems. However, how seed germination responds to N addition remains unclear.

Increasing fire risk and atmospheric nitrogen (N) deposition have the potential to alter plant community structure and composition, with consequent impacts on biodiversity and ecosystem functioning. This study was conducted to examine short-term responses of understory plant community to burning and N addition in a coniferous-broadleaved mixed forest of the subtropical-temperate transition zone in Central China. The experiment used a pair-nested design, with four treatments (control, burning, N addition, and burning plus N addition) and five replicates.

Changes in water and nitrogen (N) availability due to climate change and atmospheric N deposition could have significant effects on soil respiration, a major pathway of carbon (C) loss from terrestrial ecosystems. A manipulative experiment simulating increased precipitation and atmospheric N deposition has been conducted for 9 years (2005-2013) in a semiarid grassland in Mongolian Plateau, China. Increased precipitation and N addition interactively affect soil respiration through the 9 years.
