Mice with lymphatic dysfunction develop pathogenic lung tertiary lymphoid organs that model an autoimmune emphysema phenotype of COPD.

Am J Physiol Lung Cell Mol Physiol

January 2025

Division of Pulmonary and Critical Care, Department of Medicine, Weill Cornell Medicine, New York, New York, United States.

Article Synopsis
  • Researchers observed that mice lacking a protein called CLEC2, which affects lymphatic function, develop lung issues similar to emphysema in COPD due to the formation of tertiary lymphoid organs (TLOs).
  • Blocking the formation of these TLOs with an anti-CD20 antibody in these mice partially prevented the progression of emphysema and revealed that TLOs contributed to increased autoantibodies.
  • The study also found that chronic exposure to cigarette smoke enlarged TLOs and was linked to more severe lung injury, and similar lymphatic changes were noted in lung tissues from COPD patients with emphysema, suggesting a connection between lymphatic dysfunction and autoimmune responses in lung disease.

A Combination Therapy of Cyclophosphamide and Immunomodulating Agents in Cancer.

Curr Cancer Drug Targets

October 2024

Faculty of Health & Life Sciences, Management and Science University, Seksyen 13, 40100, Shah Alam, Selangor, Malaysia.

Cyclophosphamide is a precursor of alkylating nitrogen mustard and was initially claimed to have antineoplastic and immunosuppressive properties. However, the role of cyclo-phosphamide as an immune activator has also been reported, depending on the dosage used. The application of lower-dose cyclophosphamide has emerged as a potential approach to cancer treatment.

Computational tools can now facilitate screening precursors and selecting suitable biotransformation enzymes for producing new bioactive compounds. This study applied the data-mining approach to screen for candidate precursors of glycosyltransferases to produce new glucosides from 412 commercial natural compounds. Among five candidates, experimental results showed that only corylin could be glycosylated by the bacterial glycosyltransferase, BsUGT489.

Article Synopsis
  • * The researchers analyzed data from 685 students, finding that authoritative parenting positively correlated with access to books and motivation, which enhanced language abilities and reading skills, while authoritarian parenting had a negative effect due to lower family cohesion.
  • * The results suggest that while parenting styles do influence children's academic performance, the effects are complex and involve indirect pathways, implying that educators should consider a broader range of factors beyond just parenting styles in their assessments.

Surface availability of the dopamine (DA) transporter (DAT) critically influences DA transmission. Here, we present a protocol that describes the preparation of mouse ventral midbrain neurons, the expression of a new optical sensor, DAT-pHluorin, and the utilization of this sensor to analyze the surface availability of DAT in live neurons via fluorescent microscopy. This approach allows quantitative measures of basal surface DAT fraction under genetic backgrounds of interest and live trafficking of DAT in response to psychoactive substances.

The oncogenic transcription factor FOXM1 overexpressed in breast and other solid cancers, is a key driver of tumor growth and progression through complex interactions, making it an attractive molecular target for the development of targeted therapies. Despite the availability of small-molecule inhibitors, their limited specificity, potency, and efficacy hinder clinical translation. To identify effective FOXM1 inhibitors, we synthesized novel benzothiazole derivatives (KC10-KC13) and benzothiazole hybrids with thiazolidine-2,4-dione (KC21-KC36).

Endometriosis is a complex and heterogeneous condition affecting 10% of reproductive-age women, and yet, it often goes undiagnosed for several years. Limited observed heritability (7%) of large genetic association studies may be attributable to underlying heterogeneity of disease mechanisms. Therefore, we conducted this study to investigate genetic associations across sub-phenotypes of endometriosis.
