Topology is being widely adopted to understand and to categorize quantum matter in modern physics. A comprehensive understanding of the evolution of the immune landscape in humans across the entire lifespan at single-cell transcriptional and protein levels, during development, maturation and senescence is currently lacking. We recruited a total of 220 healthy volunteers from the Shanghai Pudong Cohort (NCT05206643), spanning 13 age groups from 0 to over 90 years, and profiled their peripheral immune cells through single-cell RNA-sequencing coupled with single T cell and B cell receptor sequencing, high-throughput mass cytometry, bulk RNA-sequencing and flow cytometry validation experiments. We revealed that T cells were the most strongly affected by age and experienced the most intensive rewiring in cell-cell interactions during specific age.

Objective: Direct electrical neurostimulation using continuous sinusoidal low frequency alternating currents (LFAC) is an emerging modality for neuromodulation. As opposed to the traditional rectangular pulse stimulation, there is limited background on the characteristics of peripheral nerves responses to sinusoidal LFAC stimulation; especially within the low frequency range (<50Hz). In this study, we demonstrate LFAC activation as a means to activate motor nerves by direct bipolar nerve stimulation via cuff electrodes, and characterize the factors of activation.

New Method: We develop and introduce the Dynamic Representation of Multimodal Activity and Markov States (DREAMS) framework, which embeds explainable artificial intelligence (XAI) techniques to model hidden state transitions during sleep using tensorized EEG, EMG, and EOG signals from 22 subjects across three age groups (18-29, 30-49, and 50-66 years). By combining Tucker decomposition with probabilistic Hidden Markov Modeling, we quantified age-specific, temazepam-induced hidden states and significant differences in transition probabilities.

Biochemical signals in native tissue microenvironments instruct cell behavior during many biological processes ranging from developmental morphogenesis and tissue regeneration to tumor metastasis and disease progression. The detection and characterization of these signals using spatial and highly resolved quantitative methods have revealed their existence as matricellular proteins in the matrisome, some of which are bound to the extracellular matrix while others are freely diffusing. Including these biochemical signals in engineered biomaterials can impart enhanced functionality and native-like complexity, ultimately benefiting efforts to understand, model, and treat various diseases.

Objectives: This study aims to elucidate the microbial signatures associated with autoimmune diseases, particularly systemic lupus erythematosus (SLE) and inflammatory bowel disease (IBD), compared with colorectal cancer (CRC), to identify unique biomarkers and shared microbial mechanisms that could inform specific treatment protocols.

Methods: We analysed metagenomic datasets from patient cohorts with six autoimmune conditions-SLE, IBD, multiple sclerosis, myasthenia gravis, Graves' disease and ankylosing spondylitis-contrasting these with CRC metagenomes to delineate disease-specific microbial profiles. The study focused on identifying predictive biomarkers from species profiles and functional genes, integrating protein-protein interaction analyses to explore effector-like proteins and their targets in key signalling pathways.
